鼠源单抗3G1单链抗体基因的构建及序列分析

利用Rr—PCR方法，从分泌抗汉滩病毒mAb的杂交瘤细胞系3Gl中扩增出抗体VH和VL基因，通过PCR方法获得1inker—VL基因，进而利用加端PCR技术，在扩增出的单抗体VH和1inker—VL基因两端加上限制性酶切位点，分别克隆入pUCl8中并测序．3Gl VH基因长360bp，编码120个氨基酸；VL基因长324bp，编码108个氨基酸．在pUCl8载体中将VH和1inker—VL基因连接成ScFv基因．     

CSDC2蛋白免疫优势肽段分析及其多克隆抗体的制备

目的 为制备含有C2的冷休克结构域(cold shock domain containing C2, CSDC2)的特异性抗体，本试验预测了CSDC2蛋白的免疫优势肽段并制备多克隆抗体，可为进一步探究其功能奠定基础。方法 本研究以绒山羊CSDC2蛋白全长氨基酸序列为基础，应用在线分析工具Expasy分析亲疏水性；SOPMA和Predictprotein共同分析二级结构；DNAstar分析抗原指数、表面可及性、柔韧性；TMHHM 2.0预测跨膜结构；SWISS-MODEL预测三级结构；SignalP 5.0预测信号肽；NetPhos 3.1预测磷酸化位点；SVMTriP在线分析软件预测抗原表位，综上分析获得CSDC2免疫优势肽段，制备多克隆抗体；ELISA检测多克隆抗体效价，Western blot验证抗体特异性。结果 CSDC2为不稳定亲水蛋白，无跨膜结构和信号肽区域，具有22个磷酸化位点，二级结构主要由无规则卷曲、α-螺旋和延伸链构成。最终选取绒山羊CSDC2蛋白N-端15-33氨基酸序列(HSPKSPVWPTFPFHREGSR)为特异免疫肽段，诱导兔产生CSDC2蛋白特异性IgG抗...     

生物信息学及其在病毒研究中的应用

介绍生物信息学的原理与方法及其在病毒研究中的应用。保守序列承担着极其重要的功能，通过多重序列对齐搜寻保守序列，是生物信息学方法的基础；敏感位点是一种反映蛋白质或核酸功能的特定模式，因此，通过数量关系的优化推导敏感位点，可用于分离病毒蛋白质与核酸相互作用位点；核苷酸和氨基酸序列只有形成了三级或四级结构才能表现功能，通过同源建模预测蛋白质的高级结构，有助于疫苗的研制、抗病毒药物的筛选以及药物的分子设计。预测RNA的三级结构大多从RNA折叠入手．病毒蛋白质三级结构预测比较成功的是日本脑炎病毒包膜糖蛋白的三级结构。     

苏云金芽孢杆菌毒素Cry7Aa1三维结构模型

运用生物信息学软件和网络数据库资源,采取同源建模的方法以毒素蛋白Cry8Ea1为模板预测了苏云金芽孢杆菌毒素Cry7Aa1的三级结构并分析了相关的生物学功能.结果表明,Cry7Aa1为3个结构域组成的近似球状蛋白,Cry7Aa1在氨基酸序列上与Cry8Ea1有40%的同源性,在三维结构_ECry7Aa1与Cry8Ea1有高度的相似性,它们结构域Ⅰ几乎完全相同,在结构域Ⅱ之间和结构域Ⅲ之间存在较小差异.Cry7Aa1与Cry8Ea1的表面电势分布不同.2种毒素具有相似的毒杀机理.模型经Ramachandran图和Verify 3D检验具有合理性.     

抗体在植物中的表达及应用前景

抗体是动物机体在外源物质(抗原)的刺激下由免疫系统及淋巴细胞产生,并能与抗原特异结合的免疫球蛋白。抗体作为机体自然免疫系统的一部分,具有特异性结合抗原、中和病毒以及与血清中一种被称为“补体”的蛋白质协同完成免疫应答反应等生物学功能,可用于治疗和诊断。抗原抗体的关系类似钥匙与锁的关系,一把锁只有相应的钥匙可以打开。正是由于抗体的高度特异性、亲和性及其性能的多样性,使抗体成为一种具有特殊用途的蛋白。成熟的抗体由抗体的重链和轻链双链组成,而结合抗原的单链抗体及催化抗体的设计进一步     

不同物种间乙酰辅酶A羧化酶的差异性和相似性

采用生物信息学方法对GenBank, SwissProt中的拟南芥、大豆、甘蓝型油菜、小麦等物种的乙酰辅酶A羧化酶核苷酸和氨基酸序列进行了比对分析,进而对其组成成分、理化性质、疏水性/亲水性、跨膜结构域、蛋白质二级结构、功能结构域、三级结构数进行了预测和分析.结果表明： ACC基因具有完整的开放阅读框,长约50683 bp,编码氨基酸相对分子质量为93.41 kD,理论等电点为7.27,是亲水性不稳定蛋白,二级结构均以α-螺旋和不规则卷曲为主要构件.通过进化树分析可分别将8种生物分为2组： A支包括小麦等7个物种,B支仅含甘蓝型油菜.     

模式识别方法预测蛋白质二级结构的研究

提出一种预测蛋白质二级结构的模式识别方法。该法首先对大量已知结构的蛋白质实验数据进行分析，找出鉴别蛋白质不同结构成分的有效信息，即设计分类器，然后实现对未知蛋白质二级结构的预测。用此方法对６４０个实验样本进行了研究，得到较高的预测精度，表明方法是有效的。还对实验结果进行了分析；讨论了有限样本对分类器性能的影响。     

拟穴青蟹(Scylla paramamosain)NOS蛋白结构分析及同源建模

为了进一步认识拟穴青蟹一氧化氮合成酶(S.paramamosain nitric oxide synthetase,SpNOS)蛋白的结构和功能,深入研究该蛋白的生物学特性,本研究采用生物信息学方法,对SpNOS蛋白质的理化性质、二级结构及三级结构等进行生物信息学分析,并在获得三级结构的基础上进行同源建模.分析结果表明,拟穴青蟹NOS蛋白与眼斑龙虾NOS蛋白组成较为接近.二级结构以α螺旋和随机卷曲为主.NOS全长1214个氨基酸,其空间结构分别以2G60、3HR4、1QGY的A链为模板,分三段进行建模,得到合理的空间结构.实验数据为深入研究一氧化氮合成酶的蛋白结构和免疫功能的关系奠定了基础.     

重组人源性抗HBsAg单链抗体的纯化与性质鉴定

对大肠杆菌表达的人源性抗乙型肝炎表面抗原（HBsAg)单链抗体（scFv)进行了纯化研究，并对单链抗体活性及其结构进行了鉴定。大肠杆菌表达的单链抗体包涵体，用8mol/L尿素裂解，经过Ni离子螯合亲和层析和凝胶过滤两步纯化后，纯度（ω）达到97％以上，再通过透析复性除去尿素。经间接ELISA和竞争性ELISA证明，复性后的scFv具有与HBsAg结合的活性，而且对鼠源抗HBsAg单克隆抗体的抑制率可达40.3%，基质辅助激光解析飞行时间质谱（matrix assisted laser desorption ionization,MALDI-TOF-MS)的结果证明，重组的scFv分子量与理论值相符，肽质量图谱的结果证明重组单链抗体的一级结构与理论序列是完全相符的，并确定了scFv蛋白中二硫键的位置。     

新型Vip3-like蛋白的从头预测建模及其结构域分析

Vip3-like蛋白对小菜蛾等鳞翅目昆虫有杀虫活性,是苏云金芽孢杆菌营养期杀虫蛋白Vip3的同源蛋白.为建立Vip3-like三维结构的理论模型并对其结构域进行生物信息学分析,选择从头预测的方法对Vip3-like蛋白三维结构的理论模型进行预测分析.分析结果表明,Vip3-like蛋白由1个Vip3A-N超家族结构域和3个糖类结合结构域组成.与模式蛋白Vip3Aa相比,Vip3-like是一种分子量更大的蛋白质,且2种蛋白的表面静电势分布也存在差异.其Vip3A-N结构域仅由α螺旋和无规则卷曲组成,没有发现β折叠.3个糖类结合结构域主要由无规则卷曲和β折叠组成,仅存在1个α螺旋结构.结构域分析预示无规则卷曲和β折叠可能在Vip3-like和受体结合的过程中扮演着重要角色.通过比较分析,发现Vip3Aa和Vip3-like蛋白在三级结构上存在较大差异,仅仅在局部结构存在一定的相似性,Vip3-like可能并不属于Vip3Aa类蛋白家族而属于一种全新的Bt营养期杀虫蛋白质.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. What＇s more, the linguistic phenomena in a speaking community can be understood more thoroughly.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
