通过对Q Sepharose F.F.离子柱的制备及荞麦蛋白酶抑制剂(BTI)的初步纯化,探索用自制层析柱代替价格昂贵的预装柱来分离纯化生物活性物质的可能性.实验结果表明:自制的Q Sepharose F.F.强阴离子层析柱可以满足对目的产品进行分离纯化的实验要求,同时与购买的预装柱比较,自装柱具有较大的上样量,柱体积灵活可变以及低成本等特点.  相似文献

A large quantity of SARS-CoV virus was proliferated in Veto cells, inactivated with β-ptopiolactone, then purified by Sepharose 4FF column chromatography to prepare inactivated vaccine. The vaccine was identified by Western blot, mass spectrographic analysis, ELISA and electton microscopy. The vaccine with or without aluminum hydtoxide adjuvant was inoculated into female BALB/c mice at different dosages. The result showed that the antibodies to SARS-CoV were induced in the mice. The antibody levels induced by the vaccine with aluminum hydroxide were higher than those without aluminum hydroxide.  相似文献

目的:对铜绿假单胞菌(Pseudomonas aeruginosa,PA)的阻遏蛋白LexA进行基因克隆、重组表达及蛋白纯化.方法:采用PCR法从PAO1株基因组中扩增出1 086 bp的LexA基因,将此基因片段插入到表达载体pET32a( )上,并在大肠杆菌BL21(DE3)中表达.包涵体洗涤后用8 mol/L尿素溶解,以镍离子亲合柱层析作为第1步纯化,Superdex75凝胶过滤层析作为第2步精细纯化,反相高相液相色谱法(HPLC)测定蛋白的浓度.结果:LexA以包涵体形式表达,表达量为45%,经镍离子亲合柱层析纯化后LexA蛋白纯度达到85%以上,回收率大于80%,镍离子亲合柱层析和凝胶过滤层析两步纯化目的蛋白,经HPLC测定,最终目的蛋白的纯度为98.97%.结论:在pET32a( )表达系统中实现了PA的LexA蛋白的高效表达,两步纯化法获得高纯度的目的蛋白,为进一步鉴定LexA靶位点奠定了基础.  相似文献

为大规模制备rhBMP-2m,将高表达rhBMP-2m的工程菌株进行高密度发酵、温度诱导表达,收集菌体,经裂菌和超声洗涤得到并纯化的包涵体。洗涤后的包涵体中,rhBMP-2mdi蛋白量的75.1%。为获得高纯度的rhBMP-2m,采用尿素作变性剂将包涵体溶解,经SP-SepharoseFF柱和Q-SepharoseFF柱两步纯化。rhBMP-2m包涵体经pH6.5尿素溶解后,经SP-Sepharose FF柱纯化,蛋白纯度为91%;若rhBMP-2m在碱性尿素中作用时间超过48h,再经Q-SepharoseFF柱纯化后,纯化产物中在还原性SDS-PAGE中会出现少量与rhBMP-2m二聚体大小接近的蛋白带,显示出碱性尿素对rhBMP-2m的共价修饰作用,会严重地影响rhBMP-2m的纯化效果和活性;若24h内完成分离纯化则影响较小,纯化后rhBMP-2m单体的纯度达98%以上。本研究表明尿素适合rhBMP-2m包涵体的大规模纯化。文章对使用尿素的利弊进行了讨论,提出大规模制备蛋白质时最好避免在碱性环境使用高浓度尿素。  相似文献

Human Cytomegalovirus (HCMV) DNA polymerase gene was overexpressed in insect cells using the baculovirus transfer system. A6. 2 kb HCMV Rsr II-EcoRI DNA fragment with intact HCMV pol gene coding sequence was engineered into NheI site of vector pBlueBac under the control of polyhedrin promoter of Autographa californica nuclear polyhedrosis virus (AcNPV). Recombinant AcNPV carried HCMV pol gene was generated by cotransfection of Spodoptera frugiperta cell (SF21) with AcNPV DNA and baculovirus transfer vector with HCMV pol gene. Infection of SF21 cell with recombinant virus lead to the expression of 140 kD peptide of HCMV specific DNA polymerase at the level approximately 2 mg per 108 cells. The polypeptide was purified from the infected SF21 cells by a series of column chromatography to homogeneity. The purified enzyme had a molecular weight of 140 kD and reacted with antiserum specific for HCMV DNA polymerase. It exhibited both 3′–5′ and 5′–3′ exonuclease activities. 利用硅胶柱层析、高效液相色谱从丽江产华丽牛肝菌子实体的甲醇提取物中分离得到7个化合物,通过波谱学方法分别鉴定为:麦角甾-ergosta-7,22-二烯-3β,5α,6β-triol(1),麦角甾-4,6,8(14),22-四烯-3-酮(2).麦角甾-5,7,22-三烯醇-3β-醇(3),麦角甾-7,22-二烯-3β-醇(4),D-阿洛醇(5),大黄素(6),大黄素甲醚(7).所有化合物均为首次从华丽牛肝菌中分离得到.  相似文献

Cystic fibrosis is associated with a defect in epithelial chloride ion transport which is caused by mutations in a membrane protein called CFTR (cystic fibrosis transmembrane conductance regulator). Heterologous expression of CFTR produces cyclicAMP-sensitive Cl(-)-channel activity. Deletion of phenylalanine at amino-acid position 508 in CFTR (delta F508 CFTR) is the most common mutation in cystic fibrosis. It has been proposed that this mutation prevents glycoprotein maturation and its transport to its normal cellular location. We have expressed both CFTR and delta F508 CFTR in Vero cells using recombinant vaccinia virus. Although far less delta F508 CFTR reached the plasma membrane than normal CFTR, sufficient delta F508 CFTR was expressed at the plasma membrane to permit functional analysis. delta F508 CFTR expression induced a reduced activity of the cAMP-activated Cl- channel, with conductance, anion selectivity and open-time kinetics similar to those of CFTR, but with much greater closed times, resulting in a large decrease of open probability. The delta F508 mutation thus seems to have two major consequences, an abnormal translocation of the CFTR protein which limits membrane insertion, and an abnormal function in mediating Cl- transport.  相似文献

用直接转化法,将构建有肝片吸虫保护性抗原基因FH3的植物表达载体pBI121-FH3,导入感受态的根瘤农杆菌EHA105;以重组的根瘤农杆菌为介导,用叶盘法转化苜蓿外植体,筛选得到抗Kan转化外植体;抗性外植体通过诱导丛生芽和诱导生根,再生出完整植株,提出植株总DNA进行Southern blot,结果表明,FH3基因已经整合入苜蓿基因组中。  相似文献

为使用外源表达载体表达并纯化有活性的水稻ATP酶蛋白Os AAA1,构建了p ET-32a-Os AAA1原核表达载体并进行体外IPTG诱导表达和Ni+柱亲和层析纯化.利用SDS-PAGE和Western Blot检测了目的蛋白.结果表明:将成功构建的p ET-32a-Os AAA1原核表达载体,转化到大肠杆菌Origami菌株后,在70~100 KD之间检测到可溶性目的蛋白带,并优化了诱导表达的较适温度、IPTG诱导浓度和诱导表达时间.故成功构建了p ET-32a-Os AAA1原核表达载体并获得了可溶性Os AAA1目的蛋白,为其后续研究奠定了基础.  相似文献

WSSV可经注射、投喂或共居感染克氏螯虾和日本沼虾,病毒感染的组织匀浆物注射后发病很快,但蔗糖介质纯化的WSSV注射后发病较慢,Ficoll-400介质纯化的WSSV注射后发病时间介于前二者之间。中国对虾细胞内WSSV一般每个囊膜只含一个核衣壳,但此毒种经克氏螯虾反复增殖后,多粒包埋的病毒粒子明显增多。从病虾的每克鳃可提出约0.66mg的WSSV。含WSSV的血淋巴或组织匀浆液感染8种昆虫细胞后,传代2次仍未观察到病毒在其中复制。  相似文献

The worldwide importance of human hepatitis B virus infection and the toll it takes in chronic liver disease, cirrhosis and hepatocarcinoma, make it imperative that a vaccine be developed for worldwide application. Human hepatitis B vaccines are presently prepared using hepatitis B surface antigen (HBsAg) that is purified from the plasma of human carriers of hepatitis B virus infection. The preparation of hepatitis B vaccine from a human source is restricted by the available supply of infected human plasma and by the need to apply stringent processes that purify the antigen and render it free of infectious hepatitis B virus and other possible living agents that might be present in the plasma. Joint efforts between our laboratories and those of Drs W. Rutter and B. Hall led to the preparation of vectors carrying the DNA sequence for HBsAg and antigen expression in the yeast Saccharomyces cerevisiae. Here we describe the development of hepatitis B vaccine of yeast cell origin. HBsAg of subtype adw was produced in recombinant yeast cell culture, and the purified antigen in alum formulation stimulated production of antibody in mice, grivet monkeys and chimpanzees. Vaccinated chimpanzees were totally protected when challenged intravenously with either homologous or heterologous subtype adr and ayw virus of human serum source. This is the first example of a vaccine produced from recombinant cells which is effective against a human viral infection.  相似文献

N-糖酰胺酶F(PNGase F)是由革兰氏阴性菌——脑膜炎脓杆菌分泌的一种分子量为34.8 kDa的去糖基化酶.本研究通过PCR技术扩增出945 bp的N-糖酰胺酶F基因,经酶切、连接,构建了大肠杆菌表达载体pET28a-PNGase F.将pET28a-PNGase F转入大肠杆菌BL21中,16℃诱导表达及Ni柱纯化后,SDS-PAGE鉴定,获得超过95%纯度的可溶性表达的N-糖酰胺酶F.对Na+依赖的中性氨基酸转运蛋白1(SNAT1)和2(SNAT2)进行脱糖基化作用检测,结果证明:纯化的N-糖酰胺酶F对SNAT1和SNAT2具有高效的脱糖基化作用.  相似文献

新城疫病毒(NDV)ND-xx08毒株经10 d龄SPF鸡胚增殖后,提取其基因组RNA并反转录成cDNA,用NDV F基因特异性引物,经PCR扩增后获得与F基因预期大小一致的DNA片段。将NDV F基因片段克隆到pMD18-T载体上,并进行EcoR I和Hind III双酶切鉴定和测序鉴定。结果显示,ND-xx08毒株F基因片段的长度为1 662 bp,共编码554个氨基酸,F蛋白的裂解位点为112R-R-Q-K-R-F117,是典型强毒株氨基酸序列结构。将NDV ND-xx08株F基因的47 bp到420 bp序列与新城疫病毒基因型I至基因型Ⅸ毒株的相同序列绘制病毒基因进化树,显示ND-xx08分离株属于基因Ⅶe型。将NDV ND-xx08株F全基因与国内外发表的23株NDV F基因核苷酸序列和氨基酸序列的同源性比较分析,结果表明,其核苷酸序列的同源性在82.7%~97.8%之间,氨基酸同源性在87.5%~97.7%之间。  相似文献

诺卡氏菌株(Nocaridasp.)82除去菌体的发酵液经硫酸镀沉淀、DEAE-纤维素离子交换层析、SephadexG-200凝胶过滤,得到高纯度的β-淀粉酶。用SDS-PAGE测定酶蛋白分子量为53000,不具亚基;酶反应最适pH和温度分别为7.0和60℃,有较高的热稳定性;Fe2十和Zn2+对酶活力有促进作用,而Hg2+对酶活力有抑制作用。  相似文献
