Rapid effects of insulin on cyclic GMP location in an intact protozoan.

We studied rapid changes in location of cyclic GMP in Tetrahymena pyriformis. Insulin caused cGMP localization in cilia and near the plasma membrane (0.5-1 min). Later (1-5 min) cGMP localization was diffuse in cytoplasm with perinuclear accentuation. Inactive insulin analogs did not elicit these changes.     

Nucleolus DNA synthesis inVicia faba root-tip meristems

Summary Autoradiographs ofVicia faba lateral root-tip meristems were prepared following a 30 min pulse with³H-thymidine. 1/3 of all interphase nuclei in the meristem are labeled, most with a uniform distribution of silver grains. 2–5% of labeled nuclei show specific nucleolus DNA labeling.     

A newTrichoplusia ni antennal receptor neuron that responds to attomolar concentrations of a minor pheromone component

Summary A newly discovered third class of sensillum trichodea on the antenna ofTrichoplusia ni contains an olfactory receptor neuron that responds to (Z)-9-tetradecen-1-ol acetate at 1×10^–18 M. The threshold of this neuron is 100–1000 times lower than the thresholds of two previously described pheromone-sensitive neurons that detect (Z)-7-dodecen-1-ol acetate and (Z)-7-tetradecen-1-ol acetate. The sensitivity and selectivity of this receptor neuron is evidence that (Z)-9-tetradecen-1-ol acetate may be important for sexual behavior.     

A potent attractant of zoospores ofAphanomyces cochlioides isolated from its host,Spinacia oleracea

A highly potent attractant of zoospores ofAphanomyces cochlioides, a causal fungus of the root rot disease of spinach (Spinacia oleracea), was isolated from spinach roots, and its structure was determined by spectroscopic evidence and chemical synthesis as cochliophilin A (5-hydroxy-6,7-methylenedioxyflavone,1). A chromosorb particle prepared by soaking in solution of1 showed a potent attracting activity toward the zoospores using concentrations of1 above 10^–9 or 10^–10 M.     

In vivo effects of epinephrine in a freshwater fish

Summary In vivo effects of epinephrine were investigated in a freshwater teleost,Barbus conchonius Hamilton. Fish given 2 mg/kg epinephrine in a single i.m. dose showed significant hypocholesterolemia and elevated, liver and kidney cholesterol levels 1–8 h postinjection. Plasma amino nitrogen evinced a transient yet significant fall at 2 h followed by a significant increase after 24 h. A marked reduction occurred in the plasma FFA and organic PO₄ levels after 1–8 h. The results offer little evidence for a lipolytic effect of epinephrine in this species, and the changes in metabolite levels are attributable, in part, to the catecholamineinduced modification of insulin secretion.N.K. thanks the U.G.C. for the award of a research fellowship.     

Pectinoacetals A-C: novel sterol hemiacetals from the gorgonianCtenocella pectinata

Three polyhydroxylated sterol hemiacetals, pectinoacetals A-C (1–3) have been isolated as their acetyl derivatives (4–6) from the acetic anhydride treated organic extract of the Indo-Pacific gorgonianCtenocella pectinata. These natural products were found to undergo very rapid epimerization at the C-18 chiral center and thus exist only as an equilibrium mixture of two diastereomers. The structure assignments are based on spectral studies and chemical modifications of the natural products.     

Phosphoramidon enhances allatostatin-mediated inhibition of juvenile hormone biosynthesis in the corpora allta of the cockroach,Diploptera punctata

Use of the enkephalinase inhibitor phosphoramidon in the in vitro radiochemical assay for juvenile hormone biosynthesis enhanced allatostatin-mediated inhibition of hormone production by corpora allata of the cockroach,Diploptera punctata. Significant increases in inhibition in day 2 virgin female CA by AST 1 (at 10^–7 M) and AST 4 (10^–8–10^–7 M) were observed in the presence of phosphoramidon (10^–5M or greater). No significant increases in inhibition were seen in CA from day 6 mated females with AST 4 (10^–9–10^–7M) and phosphoramidon combined. Phosphoramidon alone had no effect on JH biosynthesis. Analysis of allatostatin content of the CA, as determined by ELISA, revealed that addition of phosphoramidon to the medium increased the endogenous allatostatin conten in CA of virgin and mated females. The similarity in primary structure between allatostatins and enkephalin-like peptides and their similar distribution makes it probable that phosphoramidon acts by preventing breakdown of allatostatins within the CA.     

Intracellular injection of cAMP and cGMP into snail neurones induces an increase in Na+-conductance

Summary Injection of cAMP and more rarely cGMP into the neurones of the snailHelix pomatia induces an increase in membrane conductance, membrane depolarization and excitation. The effect is theophylline-dependent and has a reversal potential near — 10 mV.     

Heat production by balling in the Japanese honeybee,Apis cerana japonica as a defensive behavior against the hornet,Vespa simillima xanthoptera (Hymenoptera: Vespidae)

Summary As an effective counterattack strategy against predacious hornets, especiallyVespa simillima xanthoptera, workers ofApis cerana japonica showed a distinct balling reaction, usually involving 180–300 bees. This produced heat for as long as 20 min, giving rise to temperatures inside the ball higher than 46°C, which is lethal to the hornet but not to the bees.     

The inhibitory effect of paraquat on histamine and isoproterenol induced changes of cyclic nucleotides in rat lung slices

Summary The incubation of rat lung slices with paraquat ion (10^–4 M) had no effect on cAMP and cGMP levels of the rat lung slices. The preincubation with the same concentration of paraquat inhibited the cAMP elevating effect of histamine (10^–5 M) and isoproterenol (10^–5 M) and reduced the cGMP level to approximately 50% of the level obtained without preincubation with paraquat.This work was supported by the Research Grant No. 5 R01 HL 19720-03 from NHLI Department of Health, Education and Welfare, Washington, D.C.     

Lipid accumulation by a cellulolytic strain ofAspergillus niger

Lipid accumulation by a cellulolytic mold,Aspergillus niger, was studied. The amount of lipid accumulated ranged from 13.6–16.6% on various carbon sources, namely glucose, xylose, avicel (microcrystalline cellulose) and bagasse (a natural lignocellulosic substrate). Neutral lipids, phospholipids and glycolipids of the mycelia varied from 41.0–46.2%, 34.9–38.4% and 18.7–22.6% of total lipids, respectively. Unsaturated fatty acids comprised around 80% of total fatty materials with linoleic and oleic acid predominating. Of the four nitrogen sources tested, NH₄Cl was the best source for lipid synthesis from cellulose (bagasse). Optimum temperature range for growth and lipid synthesis was 25–30°C.     

Biosynthesis of 1-alkenes in the defensive secretions ofTribolium confusum (Tenebrionidae); stereochemical implications

Summary The terminally unsaturated hydrocarbons of the defensive secretion ofTribolium confusum are biosynthesized from fatty acids by oxidative decarboxylation. The process involves an enantiospecific cleavage of the C–H bond of thepro-(S) C(3)–H atom and simultaneous decarboxylation of the acid into an 1-alkene and carbon dioxide via ananti-periplanar transition state geometry (anti-elimination). The stereochemistry of this biotranformation is identical in all respects with the same reaction in higher plants. The mechanism seems to be of general importance for the biosynthesis of many vinylic substructures of natural products from oxygen-containing precursors.     

Acyl fluorides as reactive mimics of aldehyde pheromones: hyperactivation and aphrodisiac inHeliothis virescens

Summary Substitution of fluorine for the aldehydic hydrogen provides behaviorally active, chemically reactive pheromone mimics. In male moths of the tobacco budwormHeliothis virescens, Z9–14:Acf and Z11–16:Acf cause hyperactivity and irreversible extension of the genitalia in over 80% of treated moths. Cell-to-cell transmission in the activation of in situ nematocytes in acontia ofCalliactis parasitica

Summary It is reported that Ca²⁺-induced discharge of in situ nematocytes of acontia ofCalliactis parasitica can occur by cell-to-cell transmission along the acontial filament at a speed that averages 9.8·10^–3 cm^–1. The discharge is preceded by protrusion of nematocytes that proceeds along the acontium at a slightly higher speed.     

Inhibition and sex specific induction of spawning by serotonergic ligands in the zebra musselDreissena polymorpha (Pallas)

Serotonin (5-hydroxytryptamine, 5-HT) stimulates spawning in the zebra mussel (Dressena polymorpha), a macrofouling European bivalve that has recently invaded North America. To develop methods of controlling zebra mussel spawning, two vertebrate serotonin antagonists, methiothepin and metergoline, known to bind with high affinity to snail 5-HT receptors, were tested for their ability to block 5-HT-induced spawning in zebra mussels. Methiothepin inhibited 5-HT-induced spawning at concentrations as low as 10^–6 M. Metergoline (10^–4 M) inhibited 5-HT-induced spawning; however, at lower concentrations (10^–8 to 10^–5 M), metergoline by itself significantly induced spawning in male, but not female zebra mussels. Metergoline (10^–5 M)-induced male spawning was inhibited by 10^–5 M methiothepin. Thus, methiothepin is the most effective inhibitor and metergoline the most powerful inducer of spawning yet tested in zebra mussels.     

Action,in vitro et in vivo,de l'angiotensine II sur le captage cardiaque de la noradrénaline

Summary The action of angiotensin II on cardiac uptake of norepinephrine was investigated in the rat in vivo and in vitro. In contrast to desipramine, neither infusion of subpressive (10 ng/kg/min) or pressive (50–150 ng/kg/min) amounts of angiotensin on intact and/or binephrectomized rats, nor incubation of cardiac slices with angiotensin II (10^–5; 10^–9 M) impair the accumulation of tritiated norepinephrine and the level of metabolites. It is thus concluded that there is no inhibiting action of angiotensin II on the cardiac uptake of norepinephrine.     

Effects of corpora cardiaca extract,furosemide and ion substitution on sodium and chloride flux in perfused Malpighian tubules ofLocusta

Treatment with the co-transport inhibitor, furosemide decreased³⁶Cl^– flux across perfused Malpighian tubules ofLocusta. However, exclusion of³⁶Cl^– from the bathing medium had not effect on²²Na⁺ flux whereas substitution of bathing medium Na⁺ by K⁺ increased³⁶Cl^– flux. Diuretic extract of corpora cardiaca increased²²Na⁺ (by 106%) and³⁶Cl^– (by 335%) fluxes differentially.     

Growth ofNostoc muscorum mutants in the presence of diuron (DCMU) and L-methionine-DL-sulfoximine

Summary Diuron (DCMU) is inhibitory to the photoautotrophic and photoheterotrophic growth of the N₂-fixing blue-green algaNostoc muscorum at concentrations of 1.0×10^–5 M and 2.0×10^–5 M, respectively. A mutant of this organism resistant to 5.0×10^–5 M DCMU under its photoheterotrophic growth conditions, with the ability to utilize DCMU as a carbon and nitrogen source for growth, and complete inability to grow photoautotrophically has been isolated. With the apparent defect in its photosynthetic ability, it is suggested that theDCMU ^r mutant lacks the step inhibited by 1.0×10^–5 M DCMU, and metabolizes DCMU by an existing enzyme system in the absence of such inhibition. That this enzyme may be glutamine synthetase (GS) is explained with the help of a L-methionine-DL-sulfoximine (MSO)-resistant mutant ofN. muscorum which is able to grow faster with 2.0×10^–5 DCMU and is known to contain an altered GS.Thanks are due to the Council of Scientific and Industrial Research, CSIR Complex, Govt. of India, New Delhi-110012, for appointing the author to the Scientists' Pool for undertaking researches on the physiological and genetic controls of nitrogen metabolism in blue-green algae, a part of which is presented in this literature.     

Phosphate transport inClaviceps sp. strain SD-58

Summary Phosphate uptake inClaviceps sp. strain SD-58 was found to be linear for 20 min, proportional to cell density in mg/ml, energy dependent, and taking place against a concentration gradient with a K_m value of 45.45×10^–5 M. Osmotic shock treatment to the cell caused a reduction in phosphate uptake associated with the release of binding protein. Partial restoration of uptake was observed on incubation of osmotically shocked cells with shock fluid. The results are discussed with reference to the effect of phosphate on alkaloid synthesis inClaviceps sp. strain SD-58.
