Depression of spontaneous and ionophore-induced neurotransmitter release by Salmonella.

Exposure of frog neuromuscular junctions to heat-killed, lyophilized Salmonella typhimurium (SR 11) produces an early increase in spontaneous transmitter release followed by depression of release and blockade of the obligatory release usually induced by ionophore X537A.     

Ionophoric activity of the antibiotic X.14547 A in the mitochondrial membrane

Summary Release of Ca⁺⁺, Mg⁺⁺ and K⁺ by the carboxylic ionophore X-14547 A was studied in the mitochondrial membrane. A comparison was made with A.23187 (Calcimycin) and X.537 A (Lasalocid A) under the same experimental conditions. It was shown that in this test system X.14547 A is primarily a K⁺ carrier comparable with X.537 A.     

The influence of insulin,cAMP and the calcium ionophore X 537 A on the growth of the cartilage anlagen of limb buds in vitro

Summary Limb buds of 11-day-old mouse embryos were cultured for 6 days with insulin, dibutyryl cAMP and X 537 A. The cartilage anlage was reduced by insulin and enlarged by dibutyryl cAMP and X 537 A. The effects are due to changes in the amount of intercellular substance.

Effect of X-537A- on the phosphorylated protein in sarcoplasmic reticulum vesicles.

The effect of the antibiotic X537A on the phosphorylated ATPase (E approximately P) was investigated. The results show that X-537A depresses the level of E approximately P which is dependent on the Ca2+ gradient, while the Ca2+-independent E approximately P is not affected.     

Role of calcium in the reinitiation of oocyte meiosis in Sabellaria alveolata (L.) (Annelida, Polychaeta)]

Meiotic maturation is triggered by ionophores A 23187 and X 537 A, even in absence of extracellular calcium. CaMg free medium alone initiates 100% maturations while Ca free SW triggers varying percentages of maturation and Mg free SW has no effect.     

Effect of X-537A on the phosphorylated protein in sarcoplasmic reticulum vesicles

Summary The effect of the antibiotic X-537A on the phosphorylated ATPase (EP) was investigated. The results show that X-537A depresses the level of EP which is dependent on the Ca²⁺ gradient, while the Ca²⁺-independent EP is not affected. Acknowledgments. Summary In this note we claim that the two words and , which were used in Greece to indicate the geometrical point, had both been introduced in the scientific language in the first half of the IV^th century B.C., and that they became popular independently of each other in two different cultural circles, those of philosophy and of mathematics, respectively.We propose therefore, in contrast to a conjecture by Heiberg, a new explanation for the predominance of during the golden age of Greek mathematics and for the resurgence of during the period of decadence.

Role of calcium in the histamine release induced by D-galactosamine from rat mast cells

Rat peritoneal mast cells were isolated and purified by differential centrifugation in Ficoll. Cells pooled from three to four rats were suspended at approximately 10(6) cells/ml in a buffered salt solution and incubated for 1 h at 37 degrees C in 300 microliter volumes in the absence or presence (9 X 10(-4) M) of calcium chloride. Addition of D-galactosamine hydrochloride (DGM; 2.8 X 10(-4)M) caused (in addition to basal release) a mean +/- SEM percent histamine release of 15.7 +/- 5.2 in the presence of Ca++ and 19 +/- 4.9 in the absence of Ca++ (p greater than 0.05). It is suggested that D-galactosamine does not require extracellular Ca++ for the release of histamine from the rat mast cell.     

Inhibition of prostaglandin-induced cyclic AMP accumulation in the rat anterior pituitary by alrestatin.

Alrestatin at 25-1 X 10(-4) M inhibited the accumulation of cyclic AMP induced by prostaglandin E2, but not theophylline, in the rat anterior pituitary in vitro. Somatostatin, at lower concentrations, inhibited both; maximal inhibition of the prostaglandin effect was greater with alrestatin. As cyclic AMP is considered to be a mediator in induced-hormonal release, it appears from the present findings that alrestatin may be of potential use in altering hormonal release.     

Mutagenicity of melanin from human red hair

Summary TheSalmonella typhimurium histidine reversion test of Ames et al. was used to demonstrate that pheomelanin, the red-brown polymeric pigment produced in human skin and hair, becomes mutagenic after exposure to long wave-length UV-light; a finding consistent with the UV-induced somatic mutation hypothesis for the origin of freckles and the high susceptibility of redheads and blonds to sunlight-induced skin cancers.Acknowledgments.S. typhimurium strains were kindly provided by Dr Bruce N. A mutant of the antibiotic resistance factor R124 with altered copy number

Summary A mutation conferring increased antibiotic resistance onSalmonella typhimurium strain 11G carrying R124 was plasmid determined; strains harbouring the mutant plasmid contained more DNA as ccc plasmid than those harbouring R124. The increased copy number was manifested at all growth rates at all growth rates tested.This work was supported by a grant (to R. J. Effect of the polychlorinated biphenyl preparation,Aroclor 1242, on the quantity of neurosecretory material in the medulla terminalis X-organ of the fiddler crab,Uca pugilator

Summary Exposure of the fiddler crab,Uca pugilator, to the PCB preparation, Aroclor 1242, produces an increase in the quantity of neurosecretory material in the medulla terminalis X-organ. This Aroclor apparently inhibits release, but not synthesis, of one or more neurohormones.This investigation was supported by grant PCM75-03029 A01 from the National Science Foundation.     

Increased susceptibility ofTrypanosoma lewisi infected,or decomplemented rats toSalmonella typhimurium

Summary Rats infected withTrypanosoma lewisi or decomplemented by injection of cobra venom factor or complement activating factor of trypanosomes were found to be more susceptible to infection withSalmonella typhimurium. Antimutagenic unusual amino acids from plants

Summary Five unusual amino acids were identified as antimutagens against spontaneous mutation ofSalmonella typhimurium TA100: L-azetidine-2-carboxylic acid (1) from Liliaceae plants, -(methylenecyclopropyl)glycine (2) fromLitchi chinensis seeds, and 2-amino-4-methylhex-5-ynoic acid (3), hypoglycin A (4), and (2S, 4R)-2-amino-4-hydroxyhept-6-ynoic acid (5) fromEuphoria longana seeds. The absolute stereochemistry of5 was determined by its chiral synthesis from L-allylglycine, proving that5 is the C-4 epimer of the amino acid previously isolated from dried longan seeds.This work was partly supported by a Grant-in-Aid form the Ministry of Education, Science, and Culture, Japan, No. 57740283 (to HK).     

Paneth cell α-defensins in enteric innate immunity

Paneth cells at the base of small intestinal crypts of Lieberkühn secrete high levels of α-defensins in response to cholinergic and microbial stimuli. Paneth cell α-defensins are broad spectrum microbicides that function in the extracellular environment of the intestinal lumen, and they are responsible for the majority of secreted bactericidal peptide activity. Paneth cell α-defensins confer immunity to oral infection by Salmonella enterica serovar Typhimurium, and they are major determinants of the composition of the small intestinal microbiome. In addition to host defense molecules such as α-defensins, lysozyme, and Pla2g2a, Paneth cells also produce and release proinflammatory mediators as components of secretory granules. Disruption of Paneth cell homeostasis, with subsequent induction of endoplasmic reticulum stress, autophagy, or apoptosis, contributes to inflammation in diverse genetic and experimental mouse models.     

Experimental alteration of biomagnetic interactions among bean seeds

Summary Interactional patterns among bean-seed (Phaseolus vulgaris) clusters distributed on a table differed statistically significantly between when in clockwise rotating fields (table or magnetic) and when in counterclockwise fields or as nonrotating controls. It is postulated that clockwise rotation shortens the range of interactions.The author expresses his gratitude to Carol S. Chow and Kate M. Differences in chromosome A arrangement betweenDrosophila madeirensis andDrosophila subobscura

Summary The proximal half of the A (=X) chromosome ofD. madeirensis has a gene arrangement very similar to the A1 or A6 inversions found inD. subobscura. Polytene chromosome analysis of hybrids betweenD. madeirensis and strains ofD. subobscura homozygous for such inversions shows, however thatD. madeirensis has a gene arrangement different from any known forD. subobscura. These results provide evidence for a greater differentiation of the X chromosome in these species than has previously been described; it seems that the X chromosome is the only one that has undergone structural variation during the speciation process.     

Hemodynamic studies in a parabiotic model of portal hypertension

Splanchnic and systemic hemodynamic studies were performed in a rat model of parabiosis and portal hypertension. A portal hypertensive and a normal rat were surgically united side to side. A hyperdynamic circulation, characterized by increased cardiac index (413 +/- 26 vs 318 +/- 23 ml X min-1 X kg-1; p less than 0.05) and portal venous inflow (9.61 +/- 1.29 vs 6.33 +/- 0.36 ml X min-1 X 100 g b.wt-1; p less than 0.05), was found in all the portal hypertensive rats but not in the normal parabiotic partners. These results do not support the existence of a transferable humoral factor mediating the hyperdynamic circulatory state of chronic portal hypertension.
