三硫二苄基通过抑制JAK/STAT3信号通路抑制A549细胞的增殖和转移

目的 探讨三硫二苄基(DTS)对非小细胞肺癌(NSCLC)细胞系A549细胞增殖、转移和侵袭的抑制作用及其机制。方法 克隆形成和CCK-8检测DTS对A549细胞增殖的影响;Annexin V-FITC/PI双染色法与免疫印迹检测DTS对A549细胞凋亡及凋亡相关蛋白Bax, Bcl-2表达的影响;划痕实验和Transwell侵袭实验检测DTS对A549细胞迁移和侵袭的影响;免疫印迹检测DTS对A549细胞上皮间质转化(EMT)和JAK/STAT3信号通路相关蛋白表达的影响;利用IL-6可以激活STAT3信号通路,利用划痕实验,Transwell侵袭实验和Western blot免疫印迹分析验证DTS对A549细胞增殖和转移的作用依赖JAK/STAT3信号通路。结果 我们的结果显示DTS能抑制A549细胞增殖、迁移、侵袭并诱导其凋亡;DTS诱导促凋亡基因Bax的表达,降低抗凋亡基因Bcl-2的表达;DTS减弱EMT转化,且与JAK和STAT3磷酸化相关;IL-6通过激活STAT3信号通路可以挽救A549细胞的迁移能力。结论 DTS具有干预JAK/STAT3信号通路,从而抑制A549细胞...     

一种新的Bax异构体Baxθ促进细胞凋亡(英文)

Bcl-2蛋白家族由一类含有特征性的Bcl-2同源结构BH、且主要调控细胞死亡的蛋白组成.Bax是一个含有多个特征性BH结构域、促进凋亡的蛋白.克隆了一个全新的Bax异构体Baxθ,其cDNA由外显子1,4,5,6组成.MTC表达谱分析显示Baxθ在人体各组织中广泛且不同强度的表达.将BaxθORF插入pEGFP-N1载体,转染HEK293细胞,观察到一个15 ku的融合蛋白,提示Baxθ编码的蛋白是与Baxβ共用同一个C端,且使用一个全新的翻译起始位点.尽管Baxθ缺乏BH3结构域,但它在HEK293细胞中过表达时能促进细胞凋亡.截断体实验分析发现Baxθ第43~69位氨基酸残基可能与其促进细胞凋亡的活性有关.BH3结构域可能不是Bcl-2蛋白家族诱导细胞凋亡的唯一功能区域.     

绿豆胰蛋白酶抑制剂BBI诱导肺腺癌A549细胞凋亡的研究

研究了绿豆胰蛋白酶抑制剂BBI对人肺腺癌A549细胞增殖抑制作用和对肺腺癌A549细胞凋亡的影响.采用MTT法检测绿豆BBI对人肺癌A549细胞的增殖抑制作用;流式细胞仪检测和光镜观察绿豆BBI诱导A549细胞凋亡;Western-Blot检测凋亡蛋白Caspase3表达情况.结果显示:绿豆BBI对人肺腺癌A549细胞的增殖有明显抑制作用;绿豆BBI可以促使人肺腺癌A549细胞出现明显的染色质浓缩和凋亡小体,诱导人肺腺癌A549细胞的凋亡.说明绿豆BBI能抑制人肺腺癌A549细胞增殖、诱导人肺腺癌?A549细胞凋亡,实验为绿豆BBI抗肿瘤效应提供了依据.     

“聊红椿”提取物对非小细胞肺癌的抑制作用

目的探讨"聊红椿"提取物对非小细胞肺癌的抑制效果,研究其作用机制。方法通过有机溶剂萃取法,提取"聊红椿"中有效成分;将其作用于人肺癌细胞系A549,MTT法检测A549细胞增殖情况;流式细胞仪考察A549细胞凋亡情况;进一步通过荧光定量PCR和Western blot考察细胞凋亡因子Caspase-3,Bax,Bcl-2,cleaved PARP的mRNA和蛋白水平表达情况;构建肺癌裸小鼠皮下移植瘤模型,将低、中、高剂量"聊红椿"提取物分别腹腔注射干预治疗,作用一定时间后,测量小鼠肿瘤生长情况。结果 HPLC分析发现,与正常臭椿相比,"聊红椿"提取物中有高含量有效成分臭椿酮;MTT实验结果发现,"聊红椿"提取物能够明显抑制A549细胞增殖;流式细胞仪检测结果发现,"聊红椿"提取物能够诱导A549细胞发生凋亡;荧光定量PCR和Western blot实验结果发现,"聊红椿"提取物能够使得细胞凋亡因子Caspase-3,Bax和cleaved PARP含量显著下降,抗凋亡因子Bcl-2含量显著增加;"聊红椿"提取物作用于肺癌裸小鼠皮下移植瘤模型后,肿瘤质量和直径明显下降,且与处理浓度呈反比。结论"聊红椿"提取物能够通过影响相关细胞凋亡因子的表达,显著抑制非小细胞肺癌细胞增殖,促进细胞凋亡。     

姜黄素与阿霉素联合应用抗肿瘤的协同作用

研究姜黄素与阿霉素联合应用对人纤维肉瘤HT1080、人乳腺癌MCF-7、人肺癌A549细胞的增殖抑制作用及诱导肿瘤细胞凋亡的协同作用,探讨其抗肿瘤协同作用的机制,为临床寻找有效的化疗增敏剂提供实验依据和理论基础.MTT法测定细胞增殖抑制率,结果表明5μmol/L姜黄素与阿霉素联合用药对HT1080、MCF-7、A549细胞均表现出协同抗肿瘤作用;Hoechst 33258荧光染色观察A549细胞凋亡的形态学变化;Westernblot法检测A549细胞bcl-2蛋白的表达,结果表明姜黄素与阿霉素联合应用可显著诱导A549细胞发生凋亡,其协同抗肿瘤的机制可能与下调抑凋亡蛋白bcl-2表达有关.

细胞凋亡是细胞按照自身的程序,自己结束其生命的过程。细胞凋亡受细胞内外许多因素的调节,Bcl-2蛋白家族是目前研究得较多的一种。在骨骼肌细胞和心肌细胞内,Bcl-2家族成员之间形成二聚体,调节细胞是否进入凋亡程序。运动量和运动强度对骨骼肌和心肌的细胞凋亡有重要影响。     

几种新的植物甾醇类衍生物对人类癌细胞系增殖的影响

探讨几种新的植物甾醇类衍生物抑制人肝癌细胞系HepG2和人乳腺癌细胞系MCF-7生长及诱导其凋亡的生物学效应.采用形态学观察,溴化二甲噻唑二苯四氮唑(MTT)还原法和流式细胞仪(FCM)检测3种新的植物甾醇衍生物在不同作用时间、处理浓度时,对HepG2和 MCF-7细胞生长的影响.实验结果表明: 3种新的植物甾醇衍生物对2种细胞系的生长具有明显抑制作用,且呈现一定的对浓度和时间的依赖效应,并具有诱导细胞凋亡的功能.     

Bcl-2蛋白质家族调控细胞凋亡机制的研究进展

细胞凋亡是一种受基因调控的响应凋亡刺激的细胞程序性死亡方式.通过线粒体途径引发的凋亡主要由Bcl-2(B-cell lymphoma 2)蛋白质家族成员之间复杂的相互作用进行调控,然而科学界对其具体的相互作用模式一直存在争议.首先综述了近年来Bcl-2蛋白质家族成员之间相互作用的生物学机制,然后总结了细胞凋亡具有双稳性和该家族成员相互作用模式的数学模型,最后通过对目前流行的3种作用模式(即直接激活模式、间接激活模式、统一模式)进行数值模拟和分岔分析,认为统一模式是更合理的作用模式.以期能更好地理解病理细胞中可能存在的作用机制,从而为因凋亡异常引起的癌症、阿尔兹海默症等疾病的治疗和控制提供思路.     

绿豆胰蛋白酶抑制剂BBI诱导肺腺癌A549细胞凋亡

研究了绿豆胰蛋白酶抑制剂BBI对人肺腺癌A549细胞增殖抑制作用和对肺腺癌A549细胞凋亡的影响.结果显示:绿豆BBI对人肺腺癌A549细胞的增殖有明显抑制作用,绿豆BBI可以促使人肺腺癌A549细胞出现明显的染色质浓缩和凋亡小体,诱导人肺腺癌A549细胞的凋亡.说明绿豆BBI能抑制人肺腺癌A549细胞增殖、诱导人肺腺癌A549细胞凋亡,为绿豆BBI抗肿瘤效应提供了依据.     

水飞蓟宾通过上调P15~(INK4B)和P21~(WAF1/CIP1)活化JNK诱导人胰腺癌细胞G1期阻滞及细胞凋亡

目的:探讨水飞蓟宾对胰腺癌As PC-1细胞的增殖抑制作用及其作用机制.方法:MTT法和克隆形成抑制实验观察水飞蓟宾对人胰腺癌As PC-1细胞的增殖抑制作用,碘化丙锭(PI)单染色检测细胞周期改变,Annexin V-FITC/PI双染流式细胞术检测细胞凋亡水平,Western blotting检测细胞周期及细胞凋亡相关蛋白的表达.结果:不同浓度的水飞蓟宾对胰腺癌As PC-1细胞的生长均有抑制作用,且呈剂量-效应和时间-效应关系(P0.05),水飞蓟宾作用于As PC-1细胞48、72 h的IC50浓度分别为224.20、87.25μmol/L;克隆形成抑制实验显示,随着水飞蓟宾浓度增加,As PC-1细胞克隆形成逐渐减少.细胞周期检测结果显示,随着水飞蓟宾浓度的增加,胰腺癌As PC-1细胞出现明显G1期阻滞;水飞蓟宾处理组细胞的周期蛋白Cyclin D1、Cyclin E2、Cyclin A、Cyclin B1表达下降,细胞周期蛋白激酶CDK4、CDK6表达不变,细胞周期素依赖性蛋白激酶抑制蛋白P15INK4B、P21WAF1/CIP1表达升高,与流式检测的结果相一致.不同浓度水飞蓟宾作用48 h后,出现明显的凋亡细胞群;同时发现Caspase-9、Caspase-3活化降解,Caspase3下游效应蛋白PARP出现切割条带.JNK蛋白表达增加并磷酸化活化,Bcl-2蛋白家族中抗凋亡蛋白Bcl-2、Bcl-x L、Mcl-1表达明显降低,促凋亡蛋白Bax表达基本不变,BH3-only蛋白Bclxs、Bid、Bim表达增加.结论:水飞蓟宾明显抑制胰腺癌细胞增殖,通过诱导P15INK4B、P21WAF1/CIP1表达阻滞细胞周期在G1期,并通过诱导JNK活化激活线粒体细胞凋亡途径,进而诱导胰腺癌As PC-1细胞凋亡.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
