以黄嘴白鹭(Egretta eulophotes)卵壳为材料,电镜下观察其超微结构,利用能谱分析其元素组成,并与其它鹭类进行比较,结果表明:黄嘴白鹭卵壳的基本结构与其它鹭科鸟类相似,包括壳膜、锥体层、柱状层和表层;但是,黄嘴白鹭卵壳的超微结构和组成元素与其它鹭科鸟类有所不同;黄嘴白鹭卵壳厚度为159.6μm±0.282 8(n=3),薄于池鹭(Ardeolabacchus),白鹭(Egretta garzetta)、牛背鹭(Bubulcus ibis)和夜鹭(Nyctycoraz nyctycoraz);黄嘴白鹭卵壳的主要元素由钙(17.49%±1.621)(n=3)、碳(21.95%±1.712)(n=3)、氧(60.81%±1.601)(n=3)组成;壳膜的主要组成元素包括钙(4.765%±0.261 6)(n=3)、氧(50.50%±0.487 9)(n=3)、碳(38.77%±0.459 6)(n=3)、硫(0.950 0%±0.056 6)(n=3)、镁(0.215%±0.212)(n=3).  相似文献

为测定PELA微粒中胰岛素含量,建立反相高效液相色谱方法. 以Zorbax300SB-C18高效液相色谱柱分离;检测波长为220 nm;流动相组成为:V(H2O(含φ(TFA)=0.15%))∶V(CH3CN)=66∶34;保留时间为4.3 min. 结果在ρ(胰岛素)为10~200 μg/mL范围内,峰面积(A)与胰岛素质量浓度(ρ)成线性变化,其线性方程为:ρ=7.82×10-2A +9.88×10-2,相关系数r=0.999 95. 日内差和日间差分别为0.6%和0.7%. 当ρ(胰岛素)为60, 80和100 μg/mL时的回收率分别为(98.1±0.5)%, (99.6±1.5)%和(99.4±1.0)%(n=6). 测得胰岛素在PELA微粒中的包封率为(56.3±3.3)%(n=6).  相似文献

应用全细胞膜片钳记录技术在大鼠新鲜分离DRG神经元上,观察预加多巴胺D_2受体选择性激动剂R(—)—NPA对GABA—激活电流的调制作用.绝大部分受检细胞86.2%(50/58)对外加GABA敏感.10~(-6)~10~(-3)mol/LGABA 引起一剂量依赖性有明显去敏感作用的内向电流.对GABA敏感的50个受检细胞中13个细胞引起一较大的无明显去敏感的内向电流,其他的无反应.预加R(一)—NPA30秒后再加GABA,则对GABA—激活电流幅值的影响如下:抑制的为76%(38/5O),增强的为2%(1/50),无明显作用的为22%(11/5O).在其浓度为10~(-4)mol/L、10~(-5)mol/L、10~(-6)mol/L、10~(-7)moV/L时抑制(X±SE)分别为19.1%12.8(n=6)、42.49%±3.2(n=6)、83.6%±1.2(n=6)、8.69%±1.8(n=6).细胞内加蛋白激酶抑制剂H7后,R(—)—NPA对GABA的抑制作用完全消除(n=16).  相似文献

阿尔泰狗哇花总黄酮和总皂苷定量分析方法研究   总被引:2,自引:0,他引:2  
采用分光光度法,以槲皮素为对照品经AlCl3-NaAc显色测定总黄酮含量,以齐墩果酸为对照品经香草醛-高氯酸显色测定总皂苷含量.分析结果表明,原药材中总黄酮和总皂苷的含量分别为17.79和40.86 mg/g,线性范围分别为30.6~112.2μg(r=0.999 7)和20~60μg(r=0.999 7),平均回收率分别为97.7%(RSD=2.4%,n=5)和98.7%(RSD=0.98%,n=5).该法简便、准确、灵敏度高、重现性好,也可用于阿尔泰狗哇花总黄酮和总皂苷产品的含量测定.  相似文献

通过检测乳腺UDH、ADH、DCIS和浸润性导管癌(infiltrative ductal carcinoma,IDC)中Survivin、Bcl-2和Ki-67的表达,从凋亡、增殖等方面初步探讨乳腺癌的某些发病机制及其可能的临床意义.选取四川大学华西医院病理科1996年~2004年手术切除乳腺标本94例,经三位有经验的病理医师复习原切片,按照2003年版《WHO乳腺与女性生殖系统肿瘤与遗传学分类》标准,筛选出UDH 22例,ADH8例,DCIS36例和IDC28例.运用免疫组化Envision法检测Survivin、Bcl-2和Ki-67的表达情况.Survivin在UDH、ADH、DCIS和IDC中表达的阳性细胞百分率分别为3.27%±8.02%(0%~34%)、2.25%±2.92%(0%~8%)、9.71%±14.16%(1%~34%)和19.45%±17.81%(1%~60%).四组病变的表达差异有显著性(F=6.983,P<0.05).在IDC中表达的阳性细胞百分率分别比UDH、ADH和DCIS高(P<0.05).②Bcl-2在UDH、ADH、DCIS和IDC中表达的阳性强度分别为2.32±0.72(1~3)、2.38±0.52(2~3)、1.39±1.08(0~3)和1.04±1.11(0~3).四组病变的表达差异有显著性(P<0.05).UDH、ADH的阳性强度分别明显高于DCIS、IDC(P<0.05).③Ki-67在UDH、ADH、DCIS和IDC中的增殖指数分别为2.97±3.66%(0.5%~12%)、2.75±3.09%(0.5%~10%)、9.81±10.25%(2%~40%)和20.21±16.50%(5%~85%).四组病变的表达差异有显著性(F=11.560,P<0.05).IDC分别比UDH、ADH和DCIS的Ki-67增殖指数高(P<0.05).④Survivin与Ki-67增殖指数显著正相关性(r =0.697,P<0.05),而与Bcl-2表达呈负相关性(r =-0.425,P<0.05).得到以下结论 ①乳腺癌的发生过程中可能表现为增殖增加,凋亡抑制增加.②Survivin、Bcl-2可能都参与乳腺癌的发生、发展.  相似文献

对几种脊椎动物 (家鸽、蟾蜍、鲫鱼 )的红细胞渗透脆性、红细胞沉降率及红细胞比容几项生理特性进行了初步比较研究 .结果 :①家鸽红细胞最大渗透脆性为 0 .82 %± 0 .13 %NaCl溶液 (0 .5 0 %~ 1.0 0 % ) ,最小渗透脆性为 0 .44 %± 0 .0 4%NaCl溶液(0 .3 5 %~ 0 .5 0 % ) ;蟾蜍分别为 0 .2 1%± 0 .0 3 %NaCl溶液 (0 .18%~ 0 .3 0 % )和 0 .14 %± 0 .0 3 %NaCl溶液 (0 .10 %~ 0 .2 0 % ) ;鲫鱼分别为 0 .2 8%± 0 .0 4%NaCl溶液 (0 .2 2 %~ 0 .3 0 % )和 0 .2 0 %± 0 .0 3 %NaCl溶液 (0 15 %~ 0 .2 5 % ) .②家鸽红细胞沉降率为 4.2 4±2 .2 3mm/ 1h末 (1~ 10mm/ 1h末 ) ;蟾蜍为 11.0 3± 4.2 6mm/ 1h末 (4~ 2 7mm/ 1h末 ) ;鲫鱼为 3 .80± 0 .62mm/ 1h末(3~ 5mm/ 1h末 ) .③家鸽红细胞比容为 48.73 %± 7.11% (3 4.8%~ 5 9.2 % ) ;蟾蜍为 2 2 .77%± 8.5 2 % (7.3 %~ 41.8% ) ;鲫鱼为3 9.40 %± 7.5 3 % (2 2 .3 %~ 48.7% ) .本文对上述几种脊椎动物的红细胞生理特性的结果进行了统计学显著性检验 .  相似文献

异常黑胆质成熟剂总糖和总皂苷含量测定   总被引:1,自引:1,他引:0  
以葡萄糖为对照品.采用苯酚一硫酸法测定了异常黑胆质成熟剂中总糖含量;以甘草酸铵为对照品.采用香草醛-高氯酸法测定了异常黑胆质成熟剂中总皂苷含量.结果显示,葡萄糖浓度在0.002 3~0.0146 mg/mL范围内线性关系良好.该方法加样回收率为97.5%(n=9).精密度为1.86%(n=5),供试品总糖含量为57.99%±1.36%(n=3).甘草酸铵浓度在0.011 5~0.057 7 mg/mL范围内线性关系良好.该方法加样回收率为90.4%(n=5).精密度为1.14%(n=5).供试品总皂苷含量为4.69%±0.08%(n=3).此两种方法精密度高、准确性较强、重现性好,分别可以用作异常黑胆质成熟剂中总糖和总皂苷含量的测定.  相似文献

建立了一种高效毛细管电泳方法,用于缬沙坦和苯磺酸氨氯地平的同时分析:以30mmol/L磷酸钠缓冲液(pH=7.47)作为电解质溶液,检测波长237nm,分离电压15kV,高度差进样10s.同时进行了方法学验证:缬沙坦在0.050 4~0.504 0mg/mL内线性良好(r=0.999 8,n=6),迁移时间和峰面积的相对标准偏差(RSD)分别为0.3%和1.3%(n=6),回收率为98.98%~99.79%;苯磺酸氨氯地平在0.050 4~0.504 0mg/mL内线性良好(r=0.999 6,n=6),迁移时间和峰面积的RSD为0.6%和1.4%(n=6),回收率为98.22%~99.46%.通过上述高效毛细管电泳法,缬沙坦和苯磺酸氨氯地平在15.0min内分离良好,分离度(R=29.8).此方法快速,灵敏,实用.  相似文献

用放射配体结合实验方法确定钩藤提取物是否能与阿片受体发生相互作用,具体以3H-diprenorphine为标记配体,反应体系及反应条件同饱和结合实验,选用不同浓度的吗啡(10-10~10-5nmol/L)或钩藤提取物(0.125~4mg/mL)为竞争药物,考察钩藤提取物与转染阿片受体的结合水平.结果表明,钩藤提取物(0.125~4 mg/mL)浓度依赖地竞争3H-diprenorphine(1nmol/L)与μ、δ、κ3种阿片受体的结合,其IC50值分别为(1.27±0.86)mg/mL(n=3)(、0.59±0.21)mg/mL(n=4)和(1.20±0.24)mg/mL(n=3),其Ki值分别为(0.41±0.27)mg/mL(n=3)、(0.26±0.09)mg/mL(n=4)和(0.38±0.08)mg/mL(n=3),说明钩藤提取物非选择性地与μ、δ、κ三种阿片受体结合.  相似文献

以火麻仁粕和未脱壳火麻籽粕为原料,研究了脱壳处理对火麻蛋白回收率、色泽、溶解度、持水性、持油性、乳化性及乳化稳定性、起泡性及起泡稳定性和消化性的影响。结果表明:从脱壳处理后的火麻仁粕中提取的火麻仁蛋白在蛋白质回收率(83.90%±0.69%)、蛋白质质量分数(88.56%±0.65%)和色泽上都显著(P<0.05)优于从未脱壳火麻籽粕中提取的火麻蛋白的蛋白质回收率(45.88%±0.51%)、蛋白质质量分数(81.97%±0.81%)和色泽。脱壳处理不改变火麻蛋白的溶解度曲线和氨基酸组成,但火麻仁蛋白的功能特性都优于未脱壳火麻蛋白。经过体外模拟消化后,火麻仁蛋白消化率达到92.66%±0.23%,显著(P<0.05)高于未脱壳火麻蛋白的消化率(78.93%±1.12%)。研究表明,脱壳处理可作为从火麻籽粕中提取高品质火麻蛋白的一种必要手段。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
