大熊猫核糖体蛋白S12亚基基因(rpS12)的cDNA克隆及序列分析

根据已报道的部分哺乳动物核糖体蛋白S12亚基基因(rpS12)的相关信息设计引物,运用RT-PCR技术,从大熊猫的肌肉组织总RNA中成功克隆了核糖体蛋白S12亚基基因的表达序列,对其进行了克隆、测序及分析.结果表明:大熊猫核糖体蛋白S12亚基基因的表达序列全长为422bp,开放阅读框(ORF)为399bp,编码132个氨基酸的蛋白质,该蛋白的相对分子质量为1.45×104,PI为6.81,含有1个酪蛋白激酶Ⅱ磷酸化位点,3个N-豆蔻酰化位点和一个核糖体蛋白S12 signature位点.该基因的表达序列及其编码的氨基酸序列与已报道的部分哺乳动物有很高的相似性.     

大熊猫核糖体蛋白S15A亚基基因（rps15A）的克隆及序列分析

根据已报道的部分物种的核糖体蛋白S15A亚基基因(rps15A)的相关信息设计引物, 运用RTPCR和TouchdownPCR技术, 分别克隆了大熊猫核糖体蛋白S15A亚基的cDNA和结构基因序列, 并进行测序及分析. 结果表明: 大熊猫核糖体蛋白S15A亚基基因的表达序列全长为460 bp,开放阅读框(ORF)为393 bp, 编码130个氨基酸, 结构基因序列全长为6091 bp, 含有4个外显子和3个内含子. RPS15A蛋白的相对分子质量为14.84 kD, pI为10.62. 拓扑预测显示含有5个类型的功能位点, 即: 1个依赖cAMP和cGMP的蛋白激酶磷酸化位点, 2个蛋白激酶C磷酸化位点, 1个乙酰化位点, 1个N 糖基化位点及1个核糖体蛋白S8 signature位点. 进一步对RPS15A蛋白的结构分析及其与部分脊椎动物和果蝇RPS15A蛋白的同源性分析, 发现该基因的表达序列及其编码的氨基酸序列具有很高的相似性, 这表明真核生物核糖体蛋白亚基S15A在进化中具有较高的保守性.     

大熊猫核糖体蛋白L34基因cDNA克隆与蛋白特性

为进一步研究RPL34的结构基因,也为更深入开展大熊猫分子生物学研究积累科学资料,本研究采用RT-PCR方法,首次成功克隆了大熊猫的核糖体蛋白L34基因的cDNA序列,对克隆序列进行了测序及初步分析,并利用RPL34蛋白构建系统树.结果表明:大熊猫RPL34基因的表达序列全长为379 bp,ORF为354 bp,编码117个氨基酸的蛋白质,该蛋白分子量为13.292 9 kD,等电点(pI)为11.48,含有5种类型共11个功能位点.进一步分析发现,该基因与已报道的人、牛、褐家鼠、小家鼠、非洲爪蟾和斑马鱼等物种的编码序列及其编码的氨基酸序列同源性很高,蛋白质分子量、pI非常接近,功能位点基本相同,说明核糖体蛋白L34基因及其编码蛋白在进化过程中非常保守;进化树分类结果与传统分类一致,表明L34蛋白能很好的反映物种间的进化关系.     

元江普通野生稻金属硫蛋白基因的获得和序列分析

对SMARTTM技术构建的元江普通野生稻叶片cDNA文库进行随机测序,获得了元江普通野生稻的金属硫蛋白基因cDNA序列.该序列全长525 bp,开放阅读框长186 bp,编码62个氨基酸,10个半胱氨酸集中分布在肽链的N端和C端,该蛋白的分子量为6.42 kD,理论等电点为5.21.氨基酸序列(Blastp)同源性分析表明该基因属于金属硫蛋白家族.     

大熊猫核糖体蛋白S26亚基基因(rps26)的cDNA克隆及序列分析

为了解大熊猫核糖体蛋白亚基rps26基因的结构特点及其与已报道的人和其他哺乳动物核糖体蛋白亚基基因rps26的异同,以大熊猫的肌肉组织为材料,根据已报道的部分哺乳动物核糖体蛋白S26亚基基因(rps26)的相关信息设计引物,运用RT-PCR技术,成功地克隆了核糖体蛋白亚基基因rps26的表达序列,并对其进行了测序及初步分析.结果表明:大熊猫rps26亚基基因的表达序列长为413 bp,开放阅读框(ORF)为348 bp,编码115个氨基酸的蛋白质,该蛋白的相对分子质量为13.025 2×103,等电点为11.61.拓扑预测显示该蛋白含有3个功能位点:1个N-糖基化位点,1个酪蛋白激酶Ⅱ蛄姿峄?位点和1个核糖体蛋白S26e signature位点.进一步分析发现,大熊猫rps26基因与已报道的人、西藏黄牛、野猪、褐家鼠和小家鼠5个哺乳动物物种的表达序列其编码的氨基酸序列具有很高的相似性:编码序列同源性分别为90.23%、91.67%、92.82%、87.36%和86.78%;氨基酸序列同源性均为99.86%.     

棉铃虫泛素基因的克隆及序列分析

泛素介导的泛素-蛋白酶体通路(Ubiquitin-proteasome pathway,UPP)是真核细胞内依赖ATP的非溶酶体蛋白质降解途径,该途径对细胞内蛋白的选择性降解起着重要作用.设计一对简并引物,从棉铃虫Helicoverpa armigera卵巢细胞Ha831中克隆了泛素基因的编码区,GenBank登录号AY456195.序列分析表明,该编码区的长度为228bp,编码76个氨基酸、其编码蛋白的相对分子质量为8 560,等电点为6.56.同源性比较发现,棉铃虫泛素基因与其他真核生物泛素基因在氨基酸水平上具有96%以上的相似性,而与棉铃虫核多角体病毒泛素的相似性为76%,所有已知的泛素关键功能位点在该泛素蛋白中均保守存在.     

小菜蛾泛素基因的克隆与分析

运用RT-PCR技术,从小菜蛾幼虫总RNA反转录产物中扩增出泛素基因的cDNA序列.利用RNA干扰(RNAi)技术将一段体外合成的与小菜蛾泛素延伸蛋白基因UBA52同源的dsRNA注入四龄幼虫体内,以抑制小菜蛾UBA52基因的表达.半定量RT-PCR结果显示,72 h后沉默组小菜蛾幼虫体内UBA52基因的mRNA表达量显著下调.本实验成功沉默小菜蛾泛素基因,此RNAi干扰体系的建立为利用此技术分析小菜蛾及其他昆虫基因的功能提供了参考.     

角毛壳菌泛素结合酶基因的克隆和特性分析

利用BlastX将角毛壳菌(Chaetomium cupreum)几丁质诱导的菌丝体cDNA文库中的ESTs序列与NCBI的Nr(Non-redundant)数据库进行相似性搜索,获得泛素结合酶(ubiquitin conjugating enzyme,E2)基因的ESTs.以此结果设计引物进行RT-PCR,克隆该基因的cDNA序列(CcE2).该序列长600bp,开放阅读框444bp,编码147个氨基酸组成的多肽,蛋白分子量为16.6ku,是亲水性蛋白.BlastP分析表明该基因进化相当保守,与其他真菌的E2具有高度的相似性;三维结构分析显示CcE2仅包含一个功能结构域,在结构域的中央是决定CcE2活性的半胱氨酸残基活性位点.     

青岛文昌鱼核糖体蛋白AmphiL 37a基因的克隆和同源性分析

对青岛文昌鱼神经胚cDNA进行测序,获得了3个AmphiL 37a的EST,经拼接得到编码文昌鱼核糖体蛋白的AmphiL37a基因全长cDNA序列并演绎出AmphiL37a的氨基酸序列.通过对AmphiL37a蛋白的结构分析及其与多种脊椎动物和无脊椎动物中同种蛋白的同源性进行比较,发现AmphiL37a具有典型的四个半胱氨酸的锌指结构,与人、鼠、鸡的L37a,尤其与果蝇、隐板石鳖等高等无脊椎动物核糖体L37a具有较高的同源性,说明文昌鱼在进化上更接近于高等无脊椎动物;同时说明核糖体蛋白L37a基因在进化上具有较高保守性.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. What＇s more, the linguistic phenomena in a speaking community can be understood more thoroughly.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
