The structure of amoorastatone and the cytotoxic limonoid 12-hydroxyamoorastatin.

2 new limonoid-type terpenes have been isolated from an aqueous extract of seeds produced by the Eastern Himalayan (India) plant Aphanamixis grandifolia B1. By interpreting principally mass spectral and nuclear magnetic resonance data, the structures of 12-hydroxyamoorastatin (2b) and amoorastatone (3) were elucidated. Unequivocal evidence for the 12-hydroxyamoorastatin structural assignment was obtained by chemical conversion to sendanin (4). Amoorastatin derivative 2b was found to significantly inhibit growth of the murine P388 lymphocytic leukemia cell lines but amoorastatone in the same system was inactive. In a comparative biological study, sendanin (4) and anthothecol (7) were also found significantly to inhibit growth of the P388 cell line, while rohitukin (8) and limonin (9) were found to be inactive.     

The structure and functions of the presenilins

The presenilins (PSs) were new proteins discovered in 1995 to be involved, among other functions, in the molecular mechanisms leading to Alzheimers disease. These proteins have been the subject of many investigations since then to elucidate their molecular structures and functions. Until now, the conclusions about PS structure have been discordant, but the 8-TM structure has been accepted by the Alzheimers community, with the evidence for the 7-TM structure largely ignored. Here the evidence is reviewed for the 6-TM, 7-TM, 8-TM and other proposed models of PS topography and possibilities offered for the differences in interpretation of the various sets of data. The conclusion is that at this stage, the 7-TM model for cell surface PS is most likely the correct one.Received 22 December 2004; accepted 26 January 2005

Zusammenfassung Die Struktur desepi-Limonol-jodoacetates (C₂₆H₃₁O₈) COCH₂I, wurde durch dreidimensionale Röntgenanalyse bestimmt. Es gelang, die Lage sämtlicher Atome in zwei kristallographisch unabhängigen, jedoch chemisch identischen Molekeln zu ermitteln. Dieses Resultat stellt eine vollständige Bestimmung der Konstitution und der Stereochemie des Limonins dar.

Zusammenfassung Die Massenspektren und¹H- und¹³C-NMR-Spektren der Gelsemium-Alkaloide Gelsemin, Gelsedin und Gelsevirin wurden aufgenommen und vollständig analysiert. Gelsevirin wurde durch Reduktion in Gelsemin übergeführt und besitzt die Struktur des N_a-Methoxygelsemins.
