Summary 2 isozymes of acid phosphatase have been identified by polyacrylamide disc gel electrophoresis in the ovary and mature, unfertilized eggs ofB. rerio. Histochemically, the enzyme appears to be localized in preyolk bodies of previtellogenic oocytes and in yolk platelets of vitellogenic and postvitellogenic oocytes. The contents of the cortical granules at all stages of oocyte differentiation were acid phosphatase negative.  相似文献

Obesity, diabetes, and related metabolic disorders are major health issues worldwide. As the epidemic of metabolic disorders continues, the associated medical co-morbidities, including the detrimental impact on reproduction, increase as well. Emerging evidence suggests that the effects of maternal nutrition on reproductive outcomes are likely to be mediated, at least in part, by oocyte metabolism. Well-balanced and timed energy metabolism is critical for optimal development of oocytes. To date, much of our understanding of oocyte metabolism comes from the effects of extrinsic nutrients on oocyte maturation. In contrast, intrinsic regulation of oocyte development by metabolic enzymes, intracellular mediators, and transport systems is less characterized. Specifically, decreased acid transport proteins levels, increased glucose/lipid content and elevated reactive oxygen species in oocytes have been implicated in meiotic defects, organelle dysfunction and epigenetic alteration. Therefore, metabolic disturbances in oocytes may contribute to the diminished reproductive potential experienced by women with metabolic disorders. In-depth research is needed to further explore the underlying mechanisms. This review also discusses several approaches for metabolic analysis. Metabolomic profiling of oocytes, the surrounding granulosa cells, and follicular fluid will uncover the metabolic networks regulating oocyte development, potentially leading to the identification of oocyte quality markers and prevention of reproductive disease and poor outcomes in offspring.  相似文献

Summary Degeneration of ovarian oocytes occurred to a remarkable extent in rats with polycystic ovaries induced by dehydroepiandrosterone acetate (DHA-Ac) administration. The ratio of degeneration oocytes, compared with the total oocytes examined, finally exceeded 70%.We thank Mrs M. Aikawa and Miss R. Araki for laboratory assistance, and Dr K. Simultaneous addition of an aliquot of body fluid obtained from the surf clam, Spisula solidissima, enhanced oocyte germinal vesicle breakdown induced with serotonin but not with KCl. When the body fluid and serotonin were added sequentially to the oocytes, potentiation did not occur. Body fluids of both males and females were effective at a 200-fold dilution. The factor is stable when treated with heat, acid, base, trypsin and pronase. It is hydrophobic and not dialyzable through tubing with a molecular weight cutoff of 1000 daltons. Female fertility and offspring health are critically dependent on the maintenance of an adequate supply of high-quality oocytes. Like somatic cells, oocytes are subject to a variety of different types of DNA damage arising from endogenous cellular processes and exposure to exogenous genotoxic stressors. While the repair of intentionally induced DNA double strand breaks in gametes during meiotic recombination is well characterised, less is known about the ability of oocytes to repair pathological DNA damage and the relative contribution of DNA repair to oocyte quality is not well defined. This review will discuss emerging data suggesting that oocytes are in fact capable of efficient DNA repair and that DNA repair may be an important mechanism for ensuring female fertility, as well as the transmission of high-quality genetic material to subsequent generations.  相似文献

Summary The distribution of alkaline phosphatases has been studied in male and female gonads of some isopod crustaceans by means of Gomori's reaction.Monophosphatases are found on all Feulgen-positive nuclear structures, in the nucleolus and in the cytoplasm of young oocytes. They disappear from the cytoplasm in auxocytosis, during yolk formation, although they persist in the germinal vesicle. The authors think that the cytoplasmic localization is coincident with ribonucleic acid distribution.From the experiments related in the following note, nucleolar phosphatases appear to be in some way different from the nuclear (sensu stricto) and cytoplasmic phosphatases.Diphosphatases do not follow so closely the nucleic acid localization. They are absent from cytoplasm; in the nucleolus they are not distributed uniformly but show characteristic structure; in the nucleus they are not localized in the chromosomes but homogeneously diffused.  相似文献

Summary Examination of numerous fragmented oocytes in the ovary revealed an asynchronous relationship between nuclear and cell divisions. The asynchrony was observed in non-fragmented oocytes as well and was considered to be one of the common processes leading towards oocyte elimination in the ovary. The present study additionally demonstrates the morphology of follicle cell degeneration observed on the surface of fragmented oocytes.The authors are very grateful to Mr T. Horii, Dr K. Part of the ribosomes crystallize in the ovarian follicles during hibernation. The association between ribosomes crystals and endoplasmic reticulum membranes depends on the cell types of the follicle. The cytoplasm of oocytes with a nucleolus shows ribosome crystals. In larger oocytes, without nucleolus, there is no ribosome crystallization. The ribosomes which crystallize would be newly synthesized.  相似文献

Progesterone reinitiates in vitro meiosis in Xenopus laevis oocytes. This action of the hormone can be abolished by the exotoxin of vibrio cholerae. The concentration of toxin which inhibits 50% of the progesterone (10 muM) action in about 2.5 pM. Binding experiments using 125I labelled toxin demonstrated the existence of high affinity binding sites (KD approximately 0.2 nM) located probably on the surface of the oocytes.  相似文献

Summary Activator-deficient phosphodiesterase from bovine heart was stimulated by heat-treated dialyzed extracts from starfish oocytes. The same dose-response curve was obtained with extracts from oocytes which had or had not been released from prophase block by 1-methyladenine. Such extracts were shown to contain calmodulin by affinity chromatography on troponin I covalently bound to sepharose and SDS polyacrylamide gel electrophoresis. The results suggest that no change in calmodulin content occurs in starfish oocyte following meiosis reinitiation.  相似文献

Summary The incorporation of tritiated uridine into the follicular epithelium of degenerating oocytes of the zebrafish,Brachydanio rerio, was studied by radio-autography. After incubation times from 1–12 h, the labelling of the follicular epithelial cells of degenerating oocytes was several times higher than in normal developing follicles. This indicates an increase of RNA-synthesis, probably necessary for the formation of lytic enzymes.  相似文献

Since rare cases of sex chromosome anomalies such as XXX and XXY were observed in the offspring of our XO breeder mice, we performed a cytogenetic analysis of metaphase II oocytes of XO mice to determine whether any changes in chromosomal configurations occur. We found a significantly increased incidence of unpartnered single chromatids in metaphase II oocytes of XO mice. Such single chromatids may contribute to embryonic aneuploidy. In addition, the tendency of the X-chromosome to segregate non-randomly to the oocyte rather than to the polar body was confirmed.  相似文献

The comparative study of fertilization, with the same sperm sample, of in vitro matured oocytes and freshly ovulated ones, shows a new aspect of mammalian oocyte maturation. While 80% of freshly ovulated oocytes are fertilized, in vitro matured eggs are not fertilizable. They present the ability to be penetrated by spermatozoa 4 to 6 hrs. only after HCG injection. This is therefore not on tubal influence but depends on an oocyte specific factor which appears during the end of intrafollicular maturation.  相似文献

Technical elaboration of in vitro incubation of Parascaris equorum gonads with 3H-Uridine has permitted, for the first time, the study of RNA synthesis during oogenesis along the whole gonadic tube. In germ cells, oocytes in diakinesis (oviduct) and in division of maturation (uterus) show no label. On the contrary oogonia and growing oocytes in ovary are labelled. RNA synthesis is always detected in all parietal cells but is more active in oviduct and uterus where the gonadic wall is particularly developed.  相似文献

Summary Findings in the human ovary confirmed the fact already pointed out in animal ovaries that under the influence of oestrogens the germinal epithelium is capable of forming new oocytes. Whether the germinal epithelium of the mature woman can form new oocytes under normal conditions cannot be determined as yet, but the present findings allow the conclusion that neo-oogenesis during the reproductive period of women seems possible. Further investigations, now in progress, are required to determine this possibility.  相似文献

Summary The reproductive activity of virgin females of anAcanthoscelides obtectus strain originating from the high Colombian plateau was variable. Some females did not synthesize vitellogenin during imaginal life while others produced mature oocytes. In the case of virgin females without reproductive activity, the supply of pollen in the adult diet did not stimulate synthesis and incorporation of vitellogenin. On the other hand, the supply of pollen always induced a higher vitellogenin titre in the haemolymph of females which produced oocytes, and stimulated ovarian production.  相似文献

Summary During the development ofBombyx mori (monovoltin race) ecdysteroid levels were determined in oocytes, eggs, and haemolymph of larvae, and in the haemolymph of pupae. In haemolymph, the only RIA reactive materials are ecdysone and ecdysterone. In oocytes and eggs, other ecdysteroids are also detected. During larval instars, the ecdysteroid levels are low whereas they are very high during morphogenetic periods. During embryonic diapause, the ecdysone titer decreases during the cessation of morphogenesis.  相似文献

Oocytes incubated for 1 hr. in saline containing 1 mg/ml of antipain, leupeptin or chymostatin do not reinstate meiosis when progesterone or ionophore A 23187 are added. On the contrary such oocytes remain able to undergo meiosis when injected with cytoplasm of maturing oocytes (MPF), with Rabbit phosphorylase kinase or with Beef protein kinase. These results demonstrate that the loss of responsiveness to progesterone or ionophore is not due to toxic effects but rather to some interference with a key event which occurs prior to MPF action.  相似文献
