Fluorometric estimation of dead cells in cell suspensions

Summary An objective vitality test is proposed. It is based on the fluorescence increment of ethidium bromide in the presence of dead cells, which is proportional to cellular DNA under conditions previously defined.Acknowledgments. Stimulation of hemin synthesis in Ehrlich ascites tumor cells by mouse liver RNA

Age-related changes of the pattern of non-histone proteins in active and condensed fractions of mouse liver chromatin and hepatocarcinoma

Summary Electrophoretic analysis of histones and non-histone acid-soluble proteins in active (nuclease sensitive) and inactive chromatin from liver of young and old CBA mice and in age-related hepatocarcinomas showed a higher ratio of NHP: histones in active chromatin in old cells. Some liver- and hepatoma-specific fractions of non-histone proteins have been identified as chromatin matrix proteins.

A new rapid method for collection and preparation of cell suspensions for electrom microscopy

Summary We describe a new rapid and simple method for collection and preparation of cell suspensions for electron microscopy; the cells are prefixed with glutaraldehyde in their culture medium, and are then compacted on a filter disc. Post-fixation in osmium, staining and dehydration are performed by transferring the filter disc and the cell pellet from one solution to the next. The pellet is easily separated from the filter disc just before treatment in propylene oxide. This method preserves the fine structure as well as the classical technique. Advantages are that numerous cells have the same orientation in the sections and that many samples can be taken in a very short time.Acknowledgments. The authors are indebted to Prof. H. The effect of X-irradiation on the citrate content in mouse liver

Zusammenfassung Nach einer Bestrahlung mit 690 R beruhen Veränderungen des Citratgehaltes in der Leber gefütterter Mäuse auf unregelmässiger Nahrungsaufnahme, während in 24 h hungernden Mäusen der Citratgehalt nach 690 R über 11 Tage fast unverändert bleibt und erst vom 12.–15. Tag nach der Bestrahlung ansteigt. Die Ergebnisse werden diskutiert im Hinblick auf Phosphofruktokinase — und Acetyl-CoA-Carboxylase-Aktivität.     

Dinemorphan N-demethylation by mouse liver microsomes

Dinemorphan, an antitussive drug, is N-demethylated in vitro by mouse liver microsomes with biphasic kinetics showing two apparent Km and Vmax. Moreover, dinemorphan N-demethylation is inhibited by CO, SKF-525A, metyrapone and it is specifically catalyzed by a phenobarbital-inducible form of cytochrome P-450.     

The influence of reserpine on incorporation of3H-thymidine into mouse liver DNA

Zusammenfassung Der Einbau von³H-Thymidin in die DNA der Leber von weiblichen weissen Mäusen wurde 60 h nach der Verabreichung von 2 mg/kg Reserpin und ausserdem 60 h nach Futterentzug an nicht reserpinisierten Tieren gemessen. Reserpin führt unabhängig von der unter dieser Substanz auftretenden Hypothermie und Anorexie zu einer Hemmung des³H-Thymidin-Einbaus in die DNS der Leber um etwa 60%.     

The nature of the solitary active cells of the central nervous system

Ceux-ci sont identifiés par une méthode de coloration argentine que l'on peut pratiquer directement sur les coupes enzymatiques. La recette de cette méthode est donnée.     

Intracellular distribution of folic acid in mouse liver

Zusammenfassung Eine bedeutende Menge der gesamten Folsäure der Mäuseleberzellen ist in den Mitochondrien vorhanden. Auch nach Inkubation des Leberhomogenats mit radioaktiver Folsäure wird der Grossteil der Radioaktivität im mitochondrialen Anteil gefunden.     

Antioxidant activity of mouse liver and kidney extracts

Zusammenfassung Die hemmende Wirkung wässeriger Leber- und Nierenextrakte von Mäusen auf die DOPA-Auto-Oxydation wurde geprüft. Leber- und Nierenwerte waren von gleicher Grössenordnung und etwa dreimal grösser als die früher für Mäuse- und Menschenhaut bestimmten Werte. Der Hemmfaktor besteht zum grossen Teil aus Eiweiss und konnte in einen -SH-Gruppen enthaltenden, hitzeempfindlichen Teil und einen -SH-Gruppen-freien, hitzestabilen Teil getrennt werden. Die mögliche Bedeutung solcher Hemmstoffe für die Zelle wird erörtert.
