High mycoplasmal infection ratio in gastric cancer tissues suggests a possible association between my-coplasma infection and tumorigenesis.Because TNF-α plays an important role in carcinogenesis caused by microbes in-fection and P37 is a major immunogen of mycoplasma hy-orhinis(M.hyor.),investigating whether P37 could induce expression and secretion of TNF-α will be very significant fo elucidate the possible molecular mechanism of gastric car-cinogenesis involved with M.hyor.At the present study,we cloned full gene of p37 by PCR and mutated the 7 codes of TGA into TGG firstly,then expressed the P37 protein suc-cessfully with pGEX-4T-1 vector in E.coli,which was veri-fied with Western bolt.By RT-PCR and sensitive L929 cell toxic assay,we found that P37 protein could induce expres-sion and secretion of TNF-α from human peripheral blood mononuclear cells,and the inducing activity of P37 could be dramatically blocked by McAb PD4.These results suggest that the induction of TNF-α secretion by P37 probably plays an importan role in diseases caused by M.hyor.infection and needs to be further investigated.  相似文献

以Cd2 浓度1.25、2.503、.75 mg.kg-1腹腔注射染鲫后,用3H-胸腺嘧啶脱氧核苷掺入法研究镉对鲫外周血单个核细胞(PBMC)增殖的影响.检测鲫PBMC的增殖.结果显示:1.25 mg.kg-1组与对照组在不同时间放射性活度(dpm)的差异均无显著性;0d和3d时各染镉组与对照组dpm的差异均无显著性;5d时2.50 mg.kg-1及3.75 mg.kg-1组与对照组及1.25 mg.kg-1组dpm的差异有显著性且呈剂量效应,7d和10d时dpm与5d时基本相同;14d时3.75 mg.kg-1组与其余组dpm的差异有显著性,但已出现缩小的趋势.因此低浓度Cd2 对鲫PBMC增殖的影响不大,高浓度Cd2 则能诱导鲫PBMC增殖抑制.  相似文献

Cellular immune response is a major barrier to xenotransplantation, and cell adhesion is the first step in intercellular recognition. Flow-cytometric adhesion assay has been used to investigate the differential adhesions of monocyte (Mo), natural killer cell (NK) and T lymphocyte (T) present within human peripheral blood mononuclear cells (PBMC) to porcine aortic endothelial cells (PAEC), and to demonstrate the effect of human interferon-γ (hIFN-γ) or/and tumor necrosis factor-α (hTNF-α) pretreatment of PAEC on their adhesiveness for different PBMC subsets. The preferential sequence for PBMC subset binding to resting PAEC is Mo, NK and T cells, among which T cells show the slightest adherence; hTNF-α can act across the species, and augment Mo, NK and T cell adhesion ratios by 40%, 110% and 3 times, respectively. These results confirm at the cell level that host Mo and NK cells are major participants in the cellular xenograft rejection, thereby, providing a prerequisite for further studying the human Mo/NK-PAEC interactive mechanisms.  相似文献
