Triacylglycerol hydrolase: role in intracellular lipid metabolism

Recent scientific advances have revealed the identity of several enzymes involved in the synthesis, storage and catabolism of intracellular neutral lipid storage droplets. An enzyme that hydrolyzes stored triacylglycerol (TG), triacylglycerol hydrolase (TGH), was purified from porcine, human and murine liver microsomes. In rodents, TGH is highly expressed in liver as well as heart, kidney, small intestine and adipose tissues, while in humans TGH is mainly expressed in the liver, adipose and small intestine. TGH localizes to the endoplasmic reticulum and lipid droplets. The TGH genes are located within a cluster of carboxylesterase genes on human and mouse chromosomes 16 and 8, respectively. TGH hydrolyzes stored TG, and in the liver, the lipolytic products are made available for VLDL-TG synthesis. Inhibition of TGH activity also inhibits TG and apolipoprotein B secretion by primary hepatocytes. A role for TGH in basal TG lipolysis in adipocytes has been proposed. TGH expression and activity is both developmentally and hormonally regulated. A model for the function of TGH is presented and discussed with respect to tissue specific functions.     

A comparative study of tryptophan metabolism in skins and livers of black and albino rats

Summary The skin of black rats contains less tryptophan than the skin of albino rats, possibly because the activity of tryptophan pyrrolase is higher. Effect of glucagon on lipid metabolism

Zusammenfassung Die Wirkung von Glukagon auf den Fettstoffwechsel gesunder Hunde wird untersucht. Intravenöse Verabreichung von 1 mg kristallinem Glukagon bewirkt weder im Serum-Cholesterin- noch im -Gesamt-lipidgehalt wesentliche Abweichungen und auch die Veränderung der Lipoproteinfraktionen ist nicht signifikant. Die Blutzuckerwerte sind bei sämtlichen Tieren stark erhöht.     

Factor in human urine inhibiting lipid metabolism

Zusammenfassung Ein im Urin hungernder oder kachektischer Personen vorhandener Faktor, führt bei Mäusen nach s.c. Injektion im Blut zur Senkung des Ketonkörperspiegels. Der Leberfettgehalt wird herabgesetzt und eine schwache, jedoch regelmässig auftretende Erhöhung des Blutzuckerspiegels herbeigeführt. Bei der Ratte hemmt der Faktorin vitro die Freisetzung von Fettsäuren aus dem epididymalen Fettgewebe. Er beeinflusst möglicherweise den im Harn hungernder Menschen vorhandenen lipid mobilizing factor antagonistisch.     

The comparative metabolism of ethylene dimethanesulphonate and ethylene dibromide

Zusammenfassung Äthylenglykol-dimethansulfonat (EDS) wird von der Ratte zu N-Acetyl-S-(-hydroxyäthyl)-cystein (V), N-Acetyl-S-(-hydroxyäthyl)-cystein-S-oxyd (VI), und Methansulfonsäure abgebaut. Äthylendibromid (EDB) gibt ausser V und VI, S-(-Hydroxyäthyl)-cystein (III) und wird weiter zu Kohlendioxyd und anorganischem Sulphat via S-(-Hydroxyäthyl)-cystein-S-oxyd (IV) abgebaut. Keine der erwähnten Stoffwechselprodukte sind gegen Tumoren oder gegen Spermatogenese wirksam.

Alterations in lipid metabolism following trauma in rabbits

Zusammenfassung Nach Knochenverletzung beim Kaninchen kommt es im Blut zu einer drastischen Erhöhung der gesamten Lipide, freien Fettsäuren und des Cholesterols, während die Veränderungen des Serum-Phospholipids nur geringfügig ist. Die Erhöhung bleibt bis zum 9. Tag nach erfolgtem Knochenbruch.

Effects of porcine β-lipotropic hormone on lipid metabolism

Zusammenfassung Es wird gezeigt, dass ein aus Schweinehypophysen gewonnenes lipotropes Hormon (-LPH), in vitro und in vivo bei verschiedenen Tierarten untersucht, eine Erhöhung der Lipolyse herbeiführt. Die Wirkung war deutlich bei Kaninchen, Meerschweinchen und Schweinen, während bei Hunden, Ratten und Mäusen keine lipolytische Wirkung nachzuweisen war.     

Stimulation of methemoglobin reduction by selenium: a comparative study with erythrocytes of various animals

The extent of stimulation of methemoglobin (metHb) reduction by selenite depends upon the level of reduced glutathione (GSH) in the erythrocytes. The reason for the species difference in the effect of selenite was discussed with respect to species differences in the GSH levels in erythrocytes.     

Phospholipase D: a lipid centric review

Phospholipase D (PLD) hydrolyzes the phosphodiester bond of the glycerolipid phosphatidylcholine, resulting in the production of phosphatidic acid and free choline. Phosphatidic acid is widely considered to be the intracellular lipid mediator of many of the biological functions attributed to PLD. However, phosphatidic acid is a tightly regulated lipid in cells and can be converted to other potentially bioactive lipids, including diacylglycerol and lysophosphatidic acid. PLD activities have been described in multiple organisms, including plants, mammals, bacteria and yeast. In mammalian systems, PLD activity regulates the actin cytoskeleton, vesicle trafficking for secretion and endocytosis, and receptor signaling. PLD is in turn regulated by phosphatidylinositol-4,5-bisphosphate, protein kinase C and ADP Ribosylation Factor and Rho family GTPases. This review focuses on the lipid precursors and products of mammalian PLD metabolism, especially phosphatidic acid and the roles this lipid performs in the mediation of the functions of PLD.
