黑果枸杞的组织培养和快速繁殖   总被引:2,自引:0,他引:2  
本研究以黑果枸杞(Lycium ruthenicum)为对象,对其愈伤组织诱导、不定芽分化、诱导生根的培养条件进行筛选,并探究了黑果枸杞实验室试管苗的大田移栽.结果表明,诱导愈伤组织最适培养基为:MS+0.5mg·L-1 6-BA+0.1mg·L-1 2,4-D+10g·L-1蔗糖,出愈率达88%,愈伤组织生长状态良好;不定芽分化增殖的最适培养基为:MS+0.2mg·L-1 6-BA+10g·L-1蔗糖,不定芽增殖倍数达5.05倍;诱导生根最适培养基为:MS+0.1mg·L-1 IBA+5g·L-1蔗糖,生根率为95%.逐级炼苗后的试管苗大田移栽成活率达90%.  相似文献

以1/2MS为基本培养基,添加不同浓度的NAA、IBA和6-BA,配置16种培养基进行试验,筛选最适宜于金钗石斛兰生根的培养基.结果显示:1/2MS+0.2 mg/L 6-BA+1.0 mg/L NAA+0.5 mg/L IBA及1/2MS+0.5 mg/L 6-BA+1.0 mg/L NAA+0.2 mg/L IBA培养基最适宜于金钗石斛兰生根,生根率达100%,而且根系生长健壮.将金钗石斛兰生根组培苗移栽于6种不同的基质中,结果表明,以棕榈外壳作为移栽基质最佳,移栽成活率达100%,且幼苗生长良好.  相似文献

以大别山冬青(Ilex dabieshanensis)组培苗为试验材料,探索不同培养基及基质对组培苗生根及移栽成活率的影响。研究结果表明:(1)最适宜大别山冬青生根的培养基为1/2 MS培养基;(2)最适宜大别山冬青生根培养的生长调节剂配比为1/2MS+萘乙酸(NAA)0.3 mg·L-1+吲哚丁酸(IBA)0.2 mg·L-1,生根率高达95%;(3)最佳移栽基质是蛭石、珍珠岩、泥炭土体积比为2:1:2的基质,控制环境条件,大别山冬青生长状况良好,成活率可达85%。  相似文献

以不同植物激素组合和不同发育阶段的茎段为实验因子,开展富贵籽茎段诱导腋芽生长及其增殖和试管苗培育的实验研究。结果:(1)MS+NAA 0.1 mg·L-1+BA 0.50 mg·L-1的培养基配方较适合于从富贵籽无菌苗和当年萌发的嫩枝茎段诱导腋芽生长与增殖;(2)MS+NAA 0.1 mg·L-1+BA 1.50 mg·L-1的组合配方能较好的诱导富贵籽1年生茎段腋芽生长与增殖。(3)1/2MS+NAA 0.50 mg·L-1+IBA 0.30 mg·L-11的组合配方用于不定芽体诱导生根和培育试管苗的效果最好。  相似文献

以俄罗斯引进的蓝靛果忍冬优良无性系Berel(L.caerulea var.kamtschatika X altaica)、Rus1(L.caerulea var.kamtschatika)和Rus2(L.caerulea var.regeliana X altaica)为研究对象,以其健壮枝条上的茎尖及茎段为外植体,进行组织培养研究,建立适合俄罗斯蓝靛果忍冬外植体生长、扩繁、生根的基本培养程序.结果表明:俄罗斯蓝靛果忍冬的3个优良无性系以MS+1.0 mg/L 6-BA+0.2 mg/L IBA为初代培养基效果均较好,但继代增殖培养基和生根培养基差异较大.其中,适合Berel增殖的培养基为MS+2.0 mg/L 6-BA+0.2 mg/L IBA,增殖系数达到5.6,生根培养基为WPM+1.5 mg/L IBA;适合Rus1和Rus2继代增殖的培养基为WPM+0.5 mg/L 6-BA+0.2 mg/L IBA,增殖系数达到6.8和7.6,生根培养基为1/2MS+0.2 mg/L IBA;适合俄罗斯蓝靛果忍冬试管苗移栽的基质为草炭土+河沙(体积比1∶1)的混合基质.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
