Biotin-dependent carboxylases include acetyl-CoA carboxylase (ACC), propionyl-CoA carboxylase (PCC), 3-methylcrotonyl-CoA carboxylase (MCC), geranyl-CoA carboxylase, pyruvate carboxylase (PC), and urea carboxylase (UC). They contain biotin carboxylase (BC), carboxyltransferase (CT), and biotin-carboxyl carrier protein components. These enzymes are widely distributed in nature and have important functions in fatty acid metabolism, amino acid metabolism, carbohydrate metabolism, polyketide biosynthesis, urea utilization, and other cellular processes. ACCs are also attractive targets for drug discovery against type 2 diabetes, obesity, cancer, microbial infections, and other diseases, and the plastid ACC of grasses is the target of action of three classes of commercial herbicides. Deficiencies in the activities of PCC, MCC, or PC are linked to serious diseases in humans. Our understanding of these enzymes has been greatly enhanced over the past few years by the crystal structures of the holoenzymes of PCC, MCC, PC, and UC. The structures reveal unanticipated features in the architectures of the holoenzymes, including the presence of previously unrecognized domains, and provide a molecular basis for understanding their catalytic mechanism as well as the large collection of disease-causing mutations in PCC, MCC, and PC. This review will summarize the recent advances in our knowledge on the structure and function of these important metabolic enzymes. 相似文献

Compared with rats fed a normal diet, the activity of the protein inhibitor of acetyl CoA carboxylase in rat liver doubled after 48 h of fasting. Conversely, acetyl CoA carboxylase activity was diminished by nearly one half. In animals fasted and then subsequently refed a fat free diet, acetyl CoA carboxylase activity was elevated by nearly 9-fold, with a concomitant decrease in the activity of the protein inhibitor by about 9-fold as compared to fasted rats. Hence it appears that the regulatory protein inhibitor for acetyl CoA carboxylase is of physiological significance for fatty acid biosynthesis. 相似文献

Summary Cold-stored blood cells of rats, which were depleted of potassium and had gained an equivalent amount of sodium instead, were incubated with 500 mg% glucose at 37°C in serum. Under these conditions an extrusion of sodium from and a re-entry of potassium into the cells could be observed. Desoxycorticosterone, when added in a concentration of 1 mg per ml, was able to slow down the rate of this exchange to three fifths of the rate of untreated cells. With blood of adrenalectomised rats, the same effect of DOC was observed, although the rate without DOC was somewhat lower than in the blood of normal animals. There was no statistically significant difference between the DOC-effect in blood of normal and adrenalectomised rats. 相似文献

The influence of sodium intake on the gene expression and circulating levels of atrial natriuretic factor (ANF) was investigated in unanesthetized rats with one-kidney, one-clip renal hypertension. After clipping, the rats were maintained for 3 weeks either on a salt-deficient (n = 11) or a regular-sodium diet (n = 10). Animals which had received the regular-sodium diet exhibited significantly higher ANF mRNA levels in their right and left atria than salt-restricted animals, whereas there was no significant difference in plasma ANF levels. 相似文献

Summary The cardiac catecholamine content of Sabra rats and their 2 genetically derived substrains, hypertension prone and resistant rats, was studied by high pressure liquid chromatography and electrochemical detection. Both in the control period and after sodium and DOCA administration the cardiac noradrenaline level is higher in hypertension resistant rats than in Sabra rats, and also higher than in hypertension prone rats. This finding suggests that a reduction of the cardiac sympathetic nervous tone is involved in the genetic resistance to sodium. 相似文献

When D-alpha-tocopherol is administered i.v. to vitamin E deficient rats, significant amounts of this vitamin are bound to a nucleoprotein complex in hepatic nuclei, and this complex can be solubilized by high concentrations of sodium chloride (0.6 M). The bound vitamin in this complex, extractable by ethanol, was found to be identical with authentic alpha-tocopherol by thin layer chromatography. 相似文献

Summary After water loading, rats bearing ocular grafts of the pars intermedia of the pituitary showed that the content of sodium increased in urine, whereas potassium excretion and diuresis did not vary significatively. Summary The sodium concentration in single frog muscle fibres vacuolated by glycerol treatment was significantly higher than in devacuolated fibres. Acute renal failure (ARF) was associated with increased urinary thromboxane (TXA2) excretion and lessened excretion of sodium (UNaV) and fractional excretion of sodium (FENa%). The inhibitor of thromboxane A2-synthetase OKY-046 enhanced sodium excretion and fractional excretion of sodium in normal and saline loaded animals whereas it partially prevented the reduction in sodium excretion and creatinine clearance and significantly increased fractional excretion of sodium in glycerol treated rats suggesting a partial protection against the development of acute renal failure. 相似文献

Résumé Dans le foie de rats porteurs de tumeurs (épithéliome de Guérin et sarcome de l'estomac), les activités de la cystathionase, de la décarboxylase de l'acide cysteine sulfinique et de la sérine déhydrase sont considérablement diminuées: cette diminution est de l'ordre de 50%, 15 jours après implantation de ces tumeurs. Si à ce moment le sarcome (qui ne provoque pas de métastases) est chirurgicalement extirpé, on observe une restauration des activités enzymatiques qui, après 15 jours, est totale pour la cystathionase et très significative pour la décarboxylase.
