Carbamoyl phosphate synthetase (CPS) catalyzes one of the most remarkable reactions ever described in biological chemistry, in which carbamoyl phosphate is produced from one molecule of bicarbonate, two molecules of Mg2+ ATP, and one molecule of either glutamine or ammonia. The carbamoyl phosphate so produced is utilized in the synthesis of arginine and pyrimidine nucleotides. It is also employed in the urea cycle in most terrestrial vertebrates. Due to its large size, its important metabolic role, and the fact that it is highly regulated, CPS has been the focus of intensive investigation for nearly 40 years. Numerous enzymological, biochemical, and biophysical studies by a variety of investigators have led to a quite detailed understanding of CPS. Perhaps one of the most significant advances on this topic within the last 2 years has been the successful X-ray crystallographic analysis of CPS from Escherichia coli. Quite unexpectedly, this structural investigation revealed that the three active sites on the protein are widely separated from one another. Furthermore, these active sites are connected by a molecular tunnel with a total length of approximately 100 A, suggesting that CPS utilizes this channel to facilitate the translocation of reaction intermediates from one site to another. In this review, we highlight the recent biochemical and X-ray crystallographic results that have led to a more complete understanding of this finely tuned instrument of catalysis.  相似文献

Summary Fructose-1,6-P2 was immobilized by sodium borohydride reduction of the Schiff base formed with aminated agarose (AH-Sepharose 4B®). The coupling occurs with high yield (25 moles immobilized fructose-1,6-P2 per ml packed gel) at neutral pH and room temperature. Schiff base reduction thus provides a convenient and mild coupling procedure for sugar phosphates preserving their labile phospho ester bonds. As exemplified by a new isolation procedure for fructose-1,6-P2 aldolase from yeast, sugar phosphates insolubilized in this manner may be used for affinity chromatography of the corresponding enzymes, provided that contaminating unspecific phosphatases are removed in a preceding fractionation step.This work was supported by the Swiss National Science Foundation, grant No. 3.620-0.75. 在金盘电极表面制备L-半胱氨酸(L-Cys)自组装单分子膜,通过调节溶液的酸度使辣根过氧化物酶(HRP)带有正电荷,利用控制电位的方法在L-Cys自组装单分子膜上组装HRP,制备出辣根过氧化物酶/L-半胱氨酸/金电极(HRP/L-Cys/Au),进而研究金表面上固定化HRP的电化学表征方法以及对H2O2的催化还原作用。结果表明,该固定方法能很好地保持酶的催化活性,能有效催化H2O2分解,所制备的HRP/L-Cys/Au电极在2.2×10-5~2.2×10-2mol/L范围内,还原峰电流与H2O2的浓度呈良好线性关系,相关系数为0.996,对H2O2的检测下限为2.2×10-5mol/L,本文为生物分子的固定化提供了一种较好的方法,对生物传感器的研究具有重要意义。  相似文献

Syncoilin is a member of the intermediate filament protein family, highly expressed in skeletal and cardiac muscle. Syncoilin binds α-dystrobrevin, a component of the dystrophin associated protein complex (DAPC) located at the muscle cell membrane, and desmin, a muscle-specific intermediate filament protein, thus providing a link between the DAPC and the muscle intermediate filament network. This link may be important for muscle integrity and force transduction during contraction, a theory that is supported by the reduced force-generating capacity of muscles from syncoilin-null mice. Additionally, syncoilin is found at increased levels in the regenerating muscle fibres of patients with muscular dystrophies and mouse models of muscle disease. Therefore, syncoilin may be important for muscle regeneration in response to injury. The aims of this article are to review current knowledge about syncoilin and to discuss its possible functions in skeletal muscle. Summary Rat peritoneal mast cells incubated in vitro in the presence of L-[methyl-3H] methionine and exposed to histamine undergo a rapid but transient increase in phospholipid methylation. By using specific H1- and H2-receptor antagonists, and histamine analogues differing in their H2-receptor agonist potency, it has been demonstrated that this metabolic event is dependent on histamine binding to H2-receptors.We are grateful to Smith Kline and French Laboratories for the generous gift of histamine analogues.  相似文献

Certain rod-shaped bacteria have been reported to form elongated filamentous cells when exposed to marginal growth conditions, including refrigeration temperatures. To expand upon these observations, the filamentation of commensal Escherichia coli, E. coli O157:H7 and Salmonella spp was investigated, following exposure to certain, mildly stressing, levels of temperature, pH or water activity (aw), with levels of cellular protein being monitored during cell elongation, in some experiments. Our studies indicated that cellular filamentation could be demonstrated in all 15 strains of the above organisms tested, following exposure to marginal conditions achieved by incubation at high or low temperatures, high or low pH values and low aw. The level of environmental stress causing filamentation tended to be specific to the particular organisms. For example, Salmonella spp formed filamentous cells at 44 degrees C, whereas E. coli strains, including O157, grew by binary fission at that temperature, but formed filamentous cells at 46 degrees C. In addition, plate count techniques to enumerate bacteria during filamentation, failed to reflect the increase in cell biomass that was occurring, whereas measurements of protein concentration demonstrated the increase quite strikingly. These findings have important implications for our understanding of the ability of food-borne pathogens to cause disease, since the infectious dose of a microorganism implicated in an outbreak of such disease is typically determined by a viable count method, which could underestimate the number of potential infectious units present in a food that had been stored in such a way as to provide marginal growth conditions.  相似文献

Summary L-[3-2H,18O]glycerol was prepared and fed toEscherichia coli in order to determine the origin of the oxygen atom in the biosynthesized thiazole moiety of thiamine. Measurement by GC-MS of the isotope incorporation into the thiazole from this substrate confirmed that the 2 hydrogens and the oxygen on the C-3 carbon of glycerol are incorporated directly into the thiazole.  相似文献
