Purification of RNA from animal cells using diethyl-pyrocarbonate

Summary Extraction of RNA from animal cells by a method using diethyl-pyrocarbonate yielded 50–60% of the total RNA. RNA purified by a hot phenol-SDS method from adenovirus 2 infected cells showed about 9% homology with adenovirus DNA, and RNA purified by diethyl-pyrocarbonate-SDS showed over 7% hybridization. Profiles of RNA prepared by both methods were identical when studied by polyacrylamide gel electrophoresis.Acknowledgment. Increase in membrane conductance by adrenaline in parotid acinar cells

Summary It is shown that excitation of the - or -adrenoceptors in mouse parotid acinar cells causes a marked reduction of surface cell membrane resistance. The -adrenoceptor induced membrane effect is an increase in K conductance. The -adrenoceptor induced membrane effect does not seem to be mediated by cyclic AMP.

RNA and DNA metabolism in liver cells of normal and cancer-bearing mice

Résumé Le métabolisme des acides nucléiques des cellules du foie de souris de la souche C₃H normales et atteintes d'une tumeur maligne, a été étudié à divers intervalles de temps avec de l'adénine marquée.Dans le cas des souris normales, la proportion de RNA incorporée dans le noyau par rapport au cytoplasme se modifie fortement. Dans le cas des souris atteintes d'une tumeur maligne, ce rapport reste stable pendant la durée de l'expérience.     

Lesions in folic acid metabolism induced by primidone

Résumé Les auteurs ont étudié le mécanisme de la toxicité de la primidone (Mysoline) chez certains microorganismes. Ces recherches montrent que le métabolisme de l'acide folique est inhibé par le primidone et cela en trois étapes: (a) la réduction de l'acide folique en acide dihydrofolique, (b) le métabolisme des ptéridines nonconjugées et (c) la phosphorylation de la thymidine.

Effect of immune and normal RNA on the immunological response induced by rat red blood cells in mice

Resumen Se inmunizaron ratones BALB con glóbulos rojos de ratas de una línea endocriada. A estos ratones se les extrajo el ARN del bazo y se inyectó a otro grupo de animales de la misma cepa. Además, se inyectó el ARN proveniente de bazo de ratón normal a un segundo grupo de animales. Cinco días después de la inyección del ARN normal e inmune, ambos grupos, y un tercero, testigo, recibieron una inyección de glóbulos rojos de rata.

Effect of lactic acid on the aggregation of human platelets induced by ADP adrenaline and collagen

Zusammenfassung Nachweis, dass die Milchsäure die durch ADP und Adrenalin induzierte Aggregation der menschlichen Thrombozyten hemmt und keinen Einfluss auf die Kollagen-induzierte Plättchenaggregation in vitro hat. Die Hemmwirkung ist unspezifisch, kommt sie infolge der pH-Verschiebung in saurer Richtung zustande.

The metabolism of methadone by cultured mammalian cells

Summary Rat hepatoma tissue culture cells and mouse leukemic cells were found to metabolize [1-³H] methadone to at least 2 unidentified radioactive compounds. These results suggest that cultured cells may be useful models for studying methadone metabolism by specific cell types.This study was supported by United States Public Health Service Research Grant No. MH 15622. Adrenaline (A) is synthesized in the adrenal medullae by the enzyme phenylethanolamine-N-methyltransferase (PNMT). After surgical removal of the adrenal medullae tissue A levels ranged from 22% of control in the heart to 125% of control in the liver. Use of a novel assay to measure tissue A formation revealed that many tissues can synthesize A using PNMT and another enzyme that N-methylates both noradrenaline and dopamine. These enzymes are non-neuronal, inducible and synthesize a major fraction of tissue and urine A.
