抗人PD-1高亲和力抗血清的制备及其在PD-1免疫印迹分析中的应用

利用纯化的可溶性人PD-1胞外域(sPD-1)蛋白为免疫原免疫小鼠,获得抗血清.免疫印迹显示该抗血清与原核表达的sPD-1有特异性反应,但与某些菌体蛋白存在交叉反应.经菌体蛋白吸附法处理后,交叉反应明显减弱.以商业化抗人PD-1抗体和小鼠sPD-1抗血清分析经亲和层析富集的活化的Jurkat T细胞表达的PD-1,只有自制的抗血清鉴定到特异反应条带,其表观分子质量为49 ku.     

人小肠三叶因子的克隆及酶联免疫吸附检测

人小肠三叶因子的cDNA,克隆到原核表达载体pGEX-4T-1中,经IPTG诱导后,hITF以GST-hITF融合蛋白的形式表达,融合蛋白占细菌总蛋白的15%左右。经过Glutathione-Sepharose 4B亲和柱和Sephacry1 S 100分子筛层析纯化了hITF。SDS-PAGE和氨基酸分析证明得到了hITF纯品。以天然hITF为抗原免疫青紫蓝兔制备了抗血清,用纯化的IgG建立了hITF的酶联免疫吸附检测(ELISA)方法,给出了标准工作曲线。应用于纯化过程中样品峰的确立。     

AtPCS1基因的原核表达及多抗血清的制备

利用RT-PCR扩增拟南芥螯合肽合成酶(AtPCS1)基因全序列,构建原核表达载体pET28a-AtPCS1并测序.将该载体转化大肠杆菌BL21,用0.75mmol/LIPTG诱导融合蛋白AtPCS1-His的表达,纯化该融合蛋白.用纯化的融合蛋白免疫小鼠,ELISA法测定抗血清的效价可达1:2000.抗血清与融合蛋白及拟南芥总蛋白的western-blot结果表明:制备的抗血清具有较高的活性和特异性.     

人胎肝金属硫蛋白的分离纯化及酶联免疫吸附检测

人胎肝经匀浆、离心、乙醇沉淀后,通过Sephadex G-75、DEAE-Sepharose Fast Flow柱层析分离,获得金属硫蛋白的两种亚型(MT-1和MT-2)。经氨基酸组成分析证明是均一的。MT-1为抗原免疫青紫兰兔制备了抗血清,以纯化的IgG建立了人MT的酶联免疫吸附检测(ELISA)方法,并给出了标准工作曲线。     

谷胱甘肽转硫酶多克隆抗体的制备及应用

将含有谷胱甘肽转硫酶 ( GST)基因的 p GEX质粒转入大肠杆菌 TG1后 ,在 0 .5 mmol/L异丙基硫代 - β- D-半乳糖苷 ( IPTG)诱导培养下表达了 GST,用亲和纯化得到的 GST免疫新西兰大白兔 ,获得了滴度 ( ELISA)在 2× 1 0 5以上的抗血清 ,纯化后的抗体已成功地应用于两种新的融合蛋白 GST- FLP和 GST- Pre S1的免疫印迹检测和亲和层析法纯化     

SARS-CoV棘突蛋白片段在E.coli中的表达及免疫活性分析

SARS-冠状病毒(SARS-CoV)棘突蛋白(Spike,S)是构成病毒包膜突起的主要成分,属于型膜糖蛋白,全长1255aa,在病毒入侵宿主细胞中起重要作用.本文利用生物信息学技术,界定冠状病毒SARS-CoVS蛋白的抗原决定区域,通过PCR扩增相应片段,克隆至原核表达载体pET-H,得到四个重组质粒pET-HS1、pET-HS3、pET-HS4、pET-HS5.经序列测定证实后,转化大肠杆菌BL21(DE3),IPTG诱导获得表达,产物经紫外薄层扫描显示目的蛋白占菌体细胞总蛋白量的20%~30%.经Westernblot检测,表达蛋白与恢复期SARS病人血清呈高反应原性.纯化相应蛋白免疫小鼠,所获抗血清效价达1∶3×103.与SARS诊断试剂反应呈阳性,这一工作将为进一步研究S蛋白的定位、功能及SARS诊断提供帮助.     

人源程序性死亡配体PD-L2蛋白的原核表达与抗血清制备

利用原核表达系统获得包涵体表达形式的重组蛋白h PD-L220~123(Ig V结构域)和h PD-L220~208(Ig V结构域和Ig C结构域);将重组蛋白利用Ni-NTA柱亲和层析纯化与复性后,作为免疫原免疫Balb/c雌性小鼠制备鼠抗人的多抗血清,经ELISA法检测其效价均达到1∶106;Western-blot实验结果表明,制备的多抗血清均能特异性识别h PD-L2蛋白.以制备的多抗血清进行IHC实验评估与鉴定,结果显示:以h PD-L220~123重组蛋白为免疫原制备的多抗血清阳性较弱且定位模糊,以h PD-L220~208重组蛋白为免疫原制备的多抗血清具有很强的膜定位且背景清晰.     

牛奶中过敏原组分的分离纯化及主要过敏原的鉴定

目的:分离纯化并鉴定牛奶中引起过敏反应的主要致敏原组分,并分析其与组胺释放的关系.方法:等电点沉淀、Sephadex G-50凝胶层析和DEAE-Sepharose离子交换层析等技术分离纯化牛奶过敏原组分.脱脂乳浸液免疫C57/BL6小鼠制备抗血清.Western-Blotting分析脱脂乳致敏原组分.间接ELISA检测抗血清中slgE和sIgG;同时荧光法检测各致敏组分体外诱发免疫小鼠腹腔肥大细胞组胺释放率.结果:分离纯化出酪蛋白、β-乳球蛋白、α-乳白蛋白等致敏组分(纯度>90%).免疫印迹表明酪蛋白是主要致敏原组分.抗血清sIgG水平差异显著,sIgE水平没有大的变化;组胺释放率与sIgG水平是一致的,而与sIgE水平没有明显相关性.酪蛋白抗血清sIgG效价最高为1:25 600,组胺释放率高达(73.28±9.31)%.结论:牛奶中主要过敏原组分鉴定为酪蛋白,对牛奶过敏症的体外诊断治疗以及低/无过敏原配方奶的研制有一定的意义.     

克氏原螯虾主要变应原原肌球蛋白的一个片段区基因的克隆表达、纯化及免疫原性鉴定

克隆克氏原螯虾主要过敏原原肌球蛋白的一个片段区基因,表达并纯化该蛋白,检测其免疫原性.提取克氏原螯虾总RNA,设计特异性引物,RT-PCR克隆得到目的基因;将目的基因连入pMD19-T载体,提质粒酶切鉴定并测序;将测序正确的片段连入原核表达载体pET-32a(+)上,并转入BL21(DE3)宿主表达菌中;IPTG诱导表达;通过Ni2+亲和层析(FPLC)纯化目的蛋白;利用Western-blotting和ELISA检测该重组蛋白的免疫原性.克隆获得目的基因,其片段长为291 bp,编码97个氨基酸.重组蛋白纯化后经SDS-PAGE鉴定,目的蛋白大小与理论值相符.Western-blotting和ELISA结果表明该蛋白与克氏原螯虾过敏病人混合血清中IgE结合,具有免疫原性.     

绿色荧光蛋白GFP多克隆抗体的制备及在多种细胞中的特异性鉴定

绿色荧光蛋白是当前在生物及生物医学研究当中应用最为广泛的一种荧光标记蛋白.为开发一种高灵敏度、高特异性以及简单经济的GFP多克隆抗体,并使之适用于在跨物种细胞里表达的GFP蛋白进行检测,本文利用gfp基因cDNA全序列分别构建了带有6×His和GST标签的原核表达载体pET28A-gfp和p GEX-2T-gfp,并将其转入大肠杆菌(Escherichia coli)BL21(DE3)细胞进行诱导表达,而后进行聚丙烯酰胺凝胶电泳(SDS-PAGE)表达鉴定,分别获得了相对分子质量为2.6×10~4和5.2×10~4的重组6×His/GST-GFP融合蛋白.将亲和纯化的6×His-GFP融合蛋白免疫新西兰大白兔,采集第5次免疫后血清用ELISA测定效价为1﹕32,000.抗血清依次经Protein A亲和纯化和GST-GFP抗原抗体纯化后,用免疫印迹实验(Western blot)方法检测抗体特异性,结果表明制备的多克隆抗体能很好地识别在莱茵衣藻(Chlamydomonas reinhardtii)、HEK293细胞和大肠杆菌中表达的GFP,对继续开展GFP相关研究具有重要意义.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
