外源乳酸脱氢酶基在在双歧杆菌中的表达

以大肠杆菌-双叉双歧杆菌的穿梭质粒PhJ为基础，插入从Lactococcus lactis总DNA中用PCR方法扩增得到的报告基因乳酸脱氢酶基因（ldh)，构建了双叉歧杆菌的表达质粒phj-ldh，并采用电转化法导入双叉双歧杆菌进行了表达研究，对双叉双歧杆菌总蛋白的SDS-PAGE分析结果表明，ldh报告基因的表达产物形成了明显的条带，Western-boltting结果确认了该条带为ldh基因的产物，LDH的酶活性定量测定结果表明，重组双叉双歧杆菌的蛋白粗抽提液中LDH的比活性为5.5U/mg，菌中LDH的表达量约为2mg/L，此工作为今后双叉双歧杆菌的基因工程奠定了基础。     

一个GAP启动子的分析及点突变体构建

甘油醛-3-磷酸脱氢酶基因(GAP)作为常见的持家基因在微生物中普遍表达,因而其启动子(PGAP)被认为是一个强的组成型启动子;首先,预测并克隆了双叉双歧杆菌的GAP启动子,并利用葡萄糖醛酸苷酶基因为报告基因,分析了PGAP在大肠杆菌和双歧杆菌中驱动表达的强度;不同碳源的发酵实验表明:PGAP在葡萄糖培养基中强度最高,而3种底物类似物对PGAP强度的影响各不相同;对PGAP预测的-10区进行了系列点突变,具有更接近sigma 70启动子共有序列的突变体p MGAP3表现出更高的驱动强度;最后,通过对代表性双歧杆菌PGAP序列的比对,表明在一些核心区域,如可能的TATA盒、转录起始位点、核糖体结合位点高度保守。     

绿色荧光蛋白基因在木葡糖酸醋杆菌中的表达

绿色荧光蛋白(green fluorescent protein,GFP)广泛应用于报告基因、基因的表达与调控以及微生物信号传导.以p MV24穿梭质粒为载体,实现了GFP在木葡糖酸醋杆菌(Gluconacetobacter xylinus)中的成功表达.采用PCR技术扩增出绿色荧光蛋白基因,连接至木葡糖酸醋杆菌表达载体p MV24中,构建成功的质粒命名为p MV24-gfp+.采用电转化技术将重组载体导入木葡糖酸醋杆菌中,转化子在荧光显微镜的蓝色激发光下发出绿色荧光,为木葡糖酸醋杆菌趋化性的观察及菌体中运动蛋白的分析提供了重要的理论依据.     

谷氨酸棒状杆菌新型诱导启动子的研究

以丰富和完善谷氨酸棒状杆菌的表达元件、筛选适合工业化应用的新型诱导启动子为研究目标,借助绿色荧光蛋白GFP为报告基因,构建启动子筛选工具质粒,在谷氨酸棒杆菌中表达,筛选出三种高效表达的诱导启动子.通过检测绿色荧光蛋白表达强度可知:丙酸启动子诱导表达强度最大,葡萄糖酸启动子次之,蔗糖启动子较弱;丙酸启动子最适诱导浓度为60 ug/m L、葡萄糖酸启动子2 mg/m L、蔗糖启动子2 mg/m L,表达强度比无诱导对照组分别增加4.39、3.86、2.74倍.     

绿色荧光蛋白基因在大黄鱼体内的表达

以绿色荧光蛋白基因为报告基因，以pIRESnco为载体质粒，制备含有报告基因的质粒，以肌肉注射的方式导入大黄鱼体内，每隔一周用荧光显微镜观察绿色荧光蛋白基因在鱼体不同部位的表达情况。结果表明，绿色荧光蛋白基因可以大黄鱼体内得到表达，表达时间高达28d以上。绿色荧光蛋白基因不仅可以在肌肉注射部都组织细胞得到表达，而且在其它部位的肌肉、肝脏、肾脏和心脏等组织中也有表达，但表达水平低于肌肉注射部位组织。     

弹性蛋白酶保护因子--Elafin结构基因腺病毒载体穿梭质粒的构建、鉴定及表达

构建能表达弹性蛋白酶保护因子———elafin基因腺病毒表达载体的穿梭质粒,为进一步包装能高效表达结构蛋白的腺病毒载体做准备.以含有elafin基因的真核质粒pEGFP-N1-Elafin为模板,PCR扩增elafin基因,PCR产物以SalⅠ、EcoRⅤ双酶切,定向插入到腺病毒穿梭质粒pAdTrack-CMV中CMV启动子下游SalⅠ与EcoRⅤ位点之间,获得重组腺病毒穿梭质粒pAdTrack-CMV-Elafin,通过SalⅠ及EcoRⅤ双酶切、PCR及插入片段序列测定对该质粒进行鉴定,将pAdTrack-CMV-Elafin转染293细胞,以RT-PCR及Western-Blot检测其在293细胞中的瞬时表达.结果表明:双酶切、PCR及测序鉴定证实,pAdTrack-CMV-Elafin穿梭质粒的插入片段为elafin基因,用pAdTrack-CMV-Elafin穿梭质粒转染293细胞后可见绿色荧光蛋白的表达,RT-PCR和Western-blotting表明其可在293细胞中瞬时表达.     

纳豆激酶基因重组表达载体的构建及其稳定性

利用PCR方法从分泌纳豆激酶的纳豆杆菌基因组DN中扩增纳豆激酶基因,将该基因克隆到表达载体PBV220上,筛选重组子,通过限制性内切酶和PCR技术分析,初步确定该重组子所携外源基因为纳豆激酶基因,用凝块溶解时间法（CLT）测出表达产物具有溶解血栓活性,证明该基因可在大肠杆菌中表达,对重组菌中重组质粒的稳定性进行研究,结果表明质粒的插入对宿主菌的生长没有太大影响,该质粒在宿主菌中具有良好的分离稳定性,但结构稳定性较差,SDS-PAGE分析结果表明基因产物为分泌型,蛋白表达量占菌体蛋白的12%左右。     

hIL-2/IFN-γ嵌合基因原核表达质粒的构建与表达

为构建hIL-2/IFN-γ嵌合基因原核表达质粒,筛选其高效表达工程菌,设计特异性引物,PCR扩增目的基因并克隆至pPROEX HTb质粒Nco Ⅰ/HindⅢ位点,经双酶切及序列测定筛选阳性克隆,IPTG诱导工程菌后以SDS-PAGE电泳鉴定目的蛋白的表达情况,并测定其抗原性和生物学活性,结果成功构建了工程菌Top1O F'[hIL-2/IFN-γ],经优化表达条件后,目的蛋白表达量约占菌体总蛋白的30%,分子质量约为34 ku.ELISA和Western-blot实验结果表明,目的蛋白能特异性结合抗IFN-γ抗体,呈阳性反应,生物学活性测定显示其IL-2和IFN-γ比活性分别为1.7×107U/mg与3.2×106U/mg.     

简化的人纤溶酶原饼环区5基因的克隆及其在大肠杆菌中的融合表达

从正常人肝脏组织中提取总RNA,合理设计引物,利用RT-PCR直接得到简化的hPK-5基因.将该基因克隆至原核表达质粒pGEX-1λT,将此重组载体转化大肠杆菌JM109,经PCR、酶切及测序鉴定阳性克隆.用IPTG诱导阳性克隆表达融和蛋白,SDS-PAGE观察表达产物.再通过Western-Blotting鉴定.简化的hPK-5基因的RT-PCR产物为264bp.通过酶切、测序等方法鉴定简化的hPK-5基因正确克隆至原核表达质粒pGEX-1λT中.重组质粒pGEX-1λT/predhPK-5在大肠杆菌中     

SAM合成酶基因的克隆及在酿酒酵母中的高效表达

采用PCR技术从S.cerevisiae TCCC 31012菌株的染色体DNA中扩增得到S-腺苷甲硫氨酸合成酶-2基因(sam2),将sam2克隆到酿酒酵母表达载体pACT2的强启动子PADHI控制下,以构建高效表达质粒,并在酿酒酵母亮氨酸缺陷型茵株YS58中表达.重组质粒经鉴定含有sam2基因.工程菌YS58-2表达产物经SDS-PAGE,结果显示重组菌S-腺苷甲硫氨酸合成酶的分子质量约为43ku,经凝胶电泳扫描,表达带约占菌体总蛋白的14%.YS58-2菌株培养72 h的SAM合成酶活力为16.5 U/mg茵体蛋白,较出发菌S.cerevisiae TCCC 31012提高了40.3倍,较受体菌YS58提高了32倍.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
