针对无乳链球菌高度保守的表面免疫相关蛋白基因,采用环介导等温扩增技术(LAMP),设计4条特异性引物,优化反应体系,建立罗非鱼无乳链球菌快速诊断方法.研究结果表明,LAMP方法检测灵敏度达到30 CFU·m L-1,且对2株罗非鱼源无乳链球菌均能扩增出特异性片段,而对14株非无乳链球菌均未扩增出相应片段.建立的LAMP检测方法具有高度的特异性和灵敏性,反应在1 h内完成,为罗非鱼无乳链球菌病的快速诊断提供了一种重要的技术手段.  相似文献

食品及其原料中沙门氏菌的快速、现场检测对食品安全控制具有重要意义.根据沙门氏菌invA基因核苷酸序列设计一组引物,应用环介导等温扩增技术(LAMP),分别对7种不同血清型沙门氏菌和4种非沙门氏菌进行扩增,同时建立沙门氏菌人工污染的食品模型,比较了LAMP法与活菌计数检测的敏感性.结果表明,该方法仅对沙门氏菌产生特异性扩增,灵敏度高达336,mL-1,食品样品经细菌富集培养后,检测灵敏度高达8.25,g-1.所建立的检测沙门氏菌方法具有较高的特异性与灵敏性,操作简单、快速,可用于沙门氏菌污染食品的快速检测.  相似文献

森林病害主要是由病毒、细菌、真菌和寄生性生物引起的,其中真菌所致病害种类最多,约占森林病害的80%以上。环介导等温基因扩增(LAMP)技术是一种快速高效的DNA扩增方法,具有简单、快捷、特异性强、准确性高等特点,笔者论述了LAMP技术的原理及其在病毒、细菌、真菌和寄生性生物等方面的应用及研究。该技术广泛应用于食品、临床、环境、农业、工业和畜牧业等领域。环介导等温扩增技术为快速分子检测提供了一条新的途径,在林木病害检测领域将会有广阔的应用前景。  相似文献

为了鉴别猪瘟病毒(CSFV)野毒感染和疫苗接种,系统比较分析CSFV标准强毒株、疫苗株、不同基因亚型的野毒株的全基因组序列差异,设计一套分别针对猪瘟强毒与弱毒NS5B基因的环介导等温扩增(LAMP)引物,建立特异性的猪瘟强毒株与疫苗株的LAMP检测方法.LAMP扩增产物用罗丹明B指示剂或者琼脂糖凝胶电泳进行检测.该方法特异性好,比RT-PCR灵敏度高出1 000倍,且重复性稳定性良好,为快速准确地鉴别CSFV野毒感染和疫苗接种提供了有效的方法.  相似文献

以荚膜脂多糖(capsular polysaccharide,CPS)合成调控子resA为靶基因片段,设计了肺炎克雷伯菌特异性引物,建立了环介导恒温扩增技术(Loop-Mediated lsothermal Amplification,LAMP)检测方法.结果表明,3对LAMP引物对肺炎克雷伯菌不仅有菌种专一性,而且,具有相当高的灵敏度(2 CFU/100 g).因此,该方法有利于更快更准确的鉴定食品中肺炎克雷伯氏菌.  相似文献

为建立一种利用环介导等温扩增(LAMP)技术高效快速鉴定家鸡及早期胚胎性别的反应体系,本研究以W染色体上假基因序列EE 0.6序列为模板,设计LAMP引物,对反应温度、反应时间、Mg2+浓度、dNTP浓度等条件进行优化.结果表明,在25μL反应体系中,Mg2+终浓度为6 mmol/L,dNTP终浓度为1.5 mmol/L,63℃反应60 min时为最佳反应体系.在该反应体系下,LAMP成功地鉴定出家鸡性别的雌雄,准确率达到100%.此方法将成为一种快速有效地鉴定家鸡早期胚胎性别的新方法,且可以在生产实践中广泛推广应用.  相似文献

本研究采集同一地区三家规模化蛋鸡场的饲料、肛拭子和病死鸡样品共813份,分别采用本实验室改进的环介导等温扩增技术(LAMP)、普通PCR技术以及美国农业部沙门氏菌分离培养标准(USDA MLG 4.05)进行沙门氏菌检测,并对分离株进行血清型鉴定及ERICPCR分型.结果,三种方法均检出沙门氏菌阳性16份,检出率同为1.97%.其中病死鸡、饲料和肛拭子中检出率分别为11.29%、3.03%和1.02%.16株禽源沙门氏菌中有7种血清型,其中12株沙门氏菌ERIC-PCR分型相似性均小于90%.结果表明,本实验室改进的LAMP方法与PCR方法检出率相同,与USDA MLG 4.05的符合率为100%.16株沙门氏菌血清型多样,分子分型的相似度低.  相似文献

旨在建立一套LAMP体系对牦牛肉饼中掺杂的鸡肉和猪肉进行鉴定和检测.以牦牛、鸡和猪为研究对象,分别对牦牛的16S rRNA、鸡的12S rRNA和猪的12S rRNA-tRNA Val设计3组引物.通过模拟制作牦牛肉饼加工条件后,从肉饼中提取的DNA,进行LAMP扩增和电泳.结果表明,所设计的引物只能在对应的肉制品中扩增出目的条带,该法具有较高的特异性,可用于混合的生鲜牦牛肉或牦牛肉品中的动物源性成分的鉴定.将鸡肉和猪肉分别按一定的比例混合到牦牛肉中制成牦牛肉饼,采用建立的LAMP体系对其进行掺杂检测,鸡肉的检出限为0.25%,猪肉的检出限为0.125%.该方法在肉品的鉴别上具有简单、快速和特异性强的特点,在为熟肉制品种类来源的检测提供了新方法.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
