Male Djungarian hamsters, reared under long (16L/8D) or short (10L/14D) days, were sacrificed at various ages during the day or night, or at night following a 30-min light pulse. The pineal melatonin rhythm matured similarly under long and short days by 20 days of age. The results are discussed in context of the hypothesis that melatonin mediates the photoperiod effects which forestall puberty in short-day reared hamsters.  相似文献

Summary The levels of melatonin in plasma were measured at hourly intervals for 24 h in 8 sheep, 4 under 8L:16D (short day) and 4 under 7L:10D:1L:6D (skeleton long day) after 38 days of exposure. Mean concentrations did not differ significantly between treatments (52 pg/ml under short days; 91 pg/ml for skeleton long days), but the levels were more stable during 24 h in the SD treatment. Under skeleton long days there were 3 peaks during the 10D scotophase, with low levels during the 6D scotophase and the 7L photophase.  相似文献

In male Djungarian hamsters (Phodopus sungorus) short photoperiods (L/D 8/16) with additional 1- or 5-min light-pulses 8 h after light-off were as effective as long photoperiods (L/D 16/8) in stimulating testicular recrudescence, increase in body weight and moult into summer pelage. The results are discussed with regard to the hypothesis that the pattern of melatonin release from the pineal gland is important in mediating photoperiodic effects in mammals.  相似文献

The pineal, serotoninergic and pigmented neurons are associated with light-dependent sleep/arousal, serving as a biological clock with a circadian rhythm. This rhythm is maintained by melatonin which serves to recognise the dark phase. The neural network that responds to seasonal variations in day/night length has not been identified. The present study demonstrates that melanocytes in human skin respond to changes in the duration of UV exposure, and can serve as a biological calendar. These responses are mediated by two indoleamines, serotonin and melatonin. Higher melatonin levels correspond to long nights and long days (short UV pulse), while high serotonin levels in the presence of melatonin reflect short nights and long days (long UV exposure). This response recapitulates the sleep/arousal patterns in animals exposed to large variations in day/night cycle that cause changes in coat colour from pure white in winter to complete repigmentation in summer.  相似文献

Summary Duration of the feeding stage and corresponding weight increase during the last larval instar of the codling moth,Laspeyresia pomonella, are controlled by JH. Larvae reared under short day conditions have a relatively high titer of JH during the last larval instar and enter diapause as mature larvae. They feed longer and become heavier than larvae reared under long day conditions, which have no JH during the last larval instar and pupate when mature. By application of the JH mimetic Altosid® during the first 2 or 3 days of the last larval instar, the duration of feeding activity of larvae reared under respectively long and short day conditions was prolongated and the larvae became significantly heavier. The feeding behaviour could only be influenced by the juvenoid as long as the feeding activity of the larvae had not yet ceased.  相似文献

Asexual planarians of the speciesDugesia dorotocephala exhibit a distinct circadian rhythm of fissoning (asexual reproduction) under the influence of normal photoperiod; fissioning occurs only at night. This rhythm is broken down by continuous illumination, continuous darkness or by decapitation. The fissioning rate increases when planarians are exposed to light for less than 1 hour/day or when they are decapitated. Fissioning of decapitated planarians is suppressed by continuous treatment with melatonin, whereas fissioning resumes when these are returned to normal culture water. Interestingly, fissioning occurs at night when decapitates are treated with melatonin in the daytime, while it is observed in the daytime with night-time melatonin treatment. Endogenous melatonin was detected by HPLC and RIA. The endogenous melatonin level is always higher in those heads collected in the scotophase than in those collected in the photophase. A type of neurosecretory cell, which may synthesize melatonin, is found in the assembly of photoreceptor cells.  相似文献

Summary Pineal N-acetyltransferase activity in the male Syrian hamster exhibited a daily rhythm; the maximal night-time value was 3.5-fold higher than the day-time value. When hamsters were exposed to light at night N-acetyltransferase declined within 30 min to 1/5 of its former activity. These results indicate that in the Syrian hamster the pineal melatonin rhythm may be regulated at least partly via changes in N-acetyltransferase activity. The authors are grateful to Mrs Marie Svobodová for her skillful technical assistance.  相似文献

Summary Daily injections of melatonin were found to retard testicular regression in hamsters exposed to LD 10:14, if the injections occurred in the morning (i.e., 0.5 h after lights on), but not if they occurred in the afternoon (i.e., 6 h after lights on). These results indicate that appropriately timed injections of melatonin can at least partially block the inhibitory effects of short days on gonadal activity in the photoperiodic hamster. We wish to thank Susan Losee and Susan Stice for excellent technical assistance and Gary Ellis for his comments. A melatonin rhythm was observed in the pineals of 18-day-old chick embryos incubated under a light-dark regime of 186 h. A low pineal melatonin content was found during the light phase of the day. Concentrations started to increase 2 h after dark onset and reached maximum levels after 4 h of darkness. The amplitude of the pineal melatonin rhythm increased considerably after 2 days and night-time concentrations in 20-day-old embryos were more than 5 times higher than in 18-day-old ones. Significant day/night differences in melatonin production were found both in pineals and eyes. Exposure of eggs to 1 h of light during the dark period decreased the high melatonin concentrations in the eyes but not in the pineals of the 20-day-old chick embryo. The results suggest that in this precocial bird at least part of the circadian system may already operate during embryonic life.  相似文献

Several novel type of lipopeptides were synthesized and evaluated for their ability to stimulate non-specific resistance againstLeishmania donovani infection. Peritoneal macrophages isolated from young male hamsters treated with muramyl dipeptide (MDP) and various synthetic lipopeptides (6 mg/kg i.p.) 7 days earlier, were cultured in vitro and challenged 24 h later withL. donovani promastigotes. One lipopeptide, Central Drug Research Institute (CDRI) compound 86/450, exhibited significantly higher immunostimulatory activity than MDP. Its prophylactic activity was further confirmed in hamsters by giving 2 split doses of 3 mg/kg of the compound spaced at 2 weeks, i.e. on day –7 and +7 of challenge withL. donovani amastigotes. The prophylactic effect lasted for 7 days following the last treatment with compound 86/450. The antileishmanial action of sodium stibogluconate (SAG) was also found to be enhanced by 16% in hamsters primed with compound 86/450.CDRI Communication No. 5034.  相似文献

Evidence is clear that each melatonin-producing cell in the chick pineal gland contains a circadian oscillator that continues to function in vitro, resulting in a prominent day/night rhythm of melatonin secretion. The aim of the present investigation was to examine whether the circadian organization of the gland has an electrophysiological correlate. To this end, single-cell recordings were made from isolated chick pineal glands kept in vitro under a light/dark cycle of 12:12 h, identical to that of the donors, or under continuous light or darkness. In all the glands investigated, a very small percentage of cells exhibited sodium-dependent spontaneous spike activity with a mean frequency below 10 Hz. The cells revealed rhythms with periods in the 15- to 60-min range and, additionally, exhibited ultradian and circadian rhythms in firing, with periods of 10.75+/-1.06 h and 26.25+/-1.26 h (mean +/- standard deviation), respectively. Most of the cells exhibited circadian rhythms with higher activity during daytime than at night, showing that the electrical activity and melatonin rhythm were out of phase. Under constant light or darkness, the circadian rhythm persisted. When the light/dark cycle of the donors was phase-advanced by 5 h, the cells revealed complete entrainment. We discuss whether the cells, albeit small in number, could function as a secondary ultradian/circadian oscillator contributing to the ultradian/circadian organization of the gland.  相似文献

Summary The locomotor activity of ovariectomized, adult golden hamsters was monitored under constant dim illumination. Thiourea treatment lengthened the period () of locomotor activity of thyroidectomized hamsters in comparison to pre-operative s and in comparison to the s of control hamsters. The results confirm hormonal modulation of the circadian system.Acknowledgments. This study was supported by grant HD-02982. L.J.B. was the recipient of a N.I.M.H. Traineeship (T32-MH-15860-01). Summary Male Syrian hamsters were fed a lithogenic diet containing 7% cellulose or 4.2% pectin. After 50 days, pectin was 76% and cellulose 64% less lithogenic than the control diet. Hamsters fed the control diet for 50 days were maintained on that diet for another 50 days or fed diets containing cellulose or pectin. These was a 52% increase in gallstone incidence in hamsters continued on the control diet and a 9% increase in those on cellulose. Pectin promoted regression of gallstones (by 52%).Acknowledgments. The melatonin rhythm: both a clock and a calendar   总被引:24,自引:0,他引:24
The paper briefly reviews the data which shows that the circadian production and secretion of melatonin by the pineal gland can impart both daily, i.e., clock, and seasonal, i.e., calendar, information to the organism. The paper summarizes the 3 patterns of nocturnal melatonin production that have been described. Clearly, regardless of the pattern of nocturnal melatonin production a particular species normally displays, the duration of nightime elevated melatonin is proportional to the duration of the night length. Since daylength under natural conditions changes daily the melatonin rhythm, which adjusts to the photoperiod sends time of year information to the organism. The melatonin receptors which subserve the clock message sent by the pineal gland in the form of a melatonin cycle may reside in the biological clock itself, namely, the suprachiasmatic nuclei (SCN). The melatonin receptors that mediate seasonal changes in reproductive physiology are presumably those that are located on the pars tuberalis cells of the anterior pituitary gland. Besides these receptors which likely mediate clock and calendar information, melatonin receptors have been described in other organs. Interestingly, the distribution of melatonin receptors is highly species-specific. Whereas the clock and calendar information that the melatonin cycle imparts to the organism relies on cell membrane receptors, a fact that is of some interest considering the high lipophilicity of melatonin, recent studies indicate that other functions of melatonin may require no receptor whatsoever.  相似文献

A freerunning rhythm of locomotor activity was observed between hibernation bouts in Turkish hamsters (mesocricetus brandti) kept in 10L:14D light-dark cycles at 10 +/- 1 degree C. The data further indicate an influence of natural hypothermia upon the circadian system and its ability to entrain to light-dark cycles.  相似文献

Active inhibition of mating behaviour in a male insect is reported here for the first time. InPyrrhocoris apterus L. (Heteroptera), the most important inhibitory pathway runs from the pars intercerebralis (PI) of the brain and does not pass through the corpora allata. The inhibitory activity of the PI is promoted by short day conditions and suppressed by long days. As the effect of photoperiod is delayed, transfer procedures enabled us to record daily rhythms in mating behaviour during short days. While the extirpation of the PI results in a discrete phase shift of the long day rhythm, there is a much less significant phase shift after this operation during short days. Thus the PI has been shown to mediate the effect of photoperiod on both the inhibition and the rhythm of mating behaviour.  相似文献
