Summary Following 2/3 hepatectomy + CCl₄, the labelling- (³H-thymidine) and mitosis-index of liver epithelia increases 8 h later than after an unique 2/3 hepatectomy, and the peak of compensatory regeneration is reached 16 h later. Then the curve declines and 48 h post operationem reparative regeneration due to CCl₄ necrosises starts.

Zur Methodik der Artefaktausschaltung bei Anwendung von Diazoniumsalzen in der Histochemie (Phosphatasen,Esterasen usw.)

Summary A post-incubation with 1% sulfanilic acid solution in 10% CH₃COOH (approx. 2–10 min) or 5–10% potash alum or 0.5 mol AlCl₃ in 0, 1n HCl-after the histochemical reaction with diazonium salt-prevents N₂ formation. This gas originates from decomposition of tissue adsorbed diazonium salt which occurs after mounting in glycerin jelly. The inhibition of diazo-group disintegration depends, in the case of sulfanilic acid, on the formation of a diazoamino compound or double salt production with aluminium ions in the case of AlCl₃.

Zum Chemismus der Kadmiumreaktion im Blutserum

Summary The authors have recently introduced a new reaction, which allows to estimate the lability of the Serum Proteins. It is measured by the turbidity caused by a solution of CdSO₄. It is shown by electrophoresis before and after the CdSO₄-reaction has taken place, that the turbidity is not originating from any single protein fraction, but from all of them, though to a different degree.     

Quantitative Untersuchungen über die Proteinsyntheseaktivität von Epidermiszellen während der Extremitätenregeneration der Urodelen

Summary In the regenerating urodele limb the epidermal cells increase their volume significantly. The nucleocytoplasmic volume ratio and the protein synthesis rate obviously change in a similar manner. But in this scheme the incorporation pattern of the cells in the wound epithelium and the apical cap differs from that of all other epidermal cells by an approximately constant rate.

Die Konstitution der Salamander-Alkaloide

Summary Four alkaloids have been isolated from the skin gland of the Feuer- and Alpensalamander (Salamandra maculosa andatra Laur.). The structure, stereochemistry and absolute configuration of the principal alkaloid, samandarin, have been elucidated by chemical, optical, and X-ray-crystallographic methods. The basic structure of samandarin is derived from 5-androstan by the insertion between C-2 and C-3 of ring A of an NH group which, along with an oxygen bridge between C-1 and C-3, constitutes an oxazolidine ring; at C₁₆ there is a-hydroxyl group. By virtue of its origin and constitution samandarin occupies a unique position among alkaloids.

Isolation of human epidermal stem cells by adherence and the reconstruction of skin equivalents

The isolation of human epidermal stem cells is critical for their clinical applications. In the present study, we isolated three populations of epidermal keratinocytes according to their ability to adhere to collagen type IV: i.e., rapidly adhering (RA), slowly adhering (SA), and non-adhering (NA) cells. The aim of this study was to characterize RA cells and to investigate the possibility of using these cells for epidermis reconstruction. To identify RA cells, flow cytometric analysis was performed using anti-₆ integrin and anti-CD71 antibodies. RA cells express high levels of ₆ integrin and low levels of CD71, which are considered as markers of an epidermal stem cell nature. Furthermore, electron microscopy showed that RA cells are small and have a high nuclear to cytoplasmic ratio, whereas SA and NA cells have well-developed cellular organelles and abundant tonofilaments. Western blot analysis showed that RA cells are slow cycling and express p63, a putative epidermal stem cell marker, whereas SA and NA cells express c-Myc, which is known to regulate stem cell fate. To compare epidermal regenerative abilities, skin equivalents (SEs) were made using RA, SA, and NA cells. The epidermis constructed from RA cells was well formed compared to those formed from SA or NA cells. In addition, only SEs with RA cells expressed ₆ integrin and ₁ integrin at the basal layer. These results indicate that RA cells represent epidermal stem cells and are predominately comprised of stem cells. Therefore, the isolation of RA cells using a simple technique offers a potential route to their clinical application, because they are easily isolated and provide a high yield of epidermal stem cells.Received 2 July 2004; received after revision 20 August 2004; accepted 10 September 2004

Summary The placentae of 17 to 19 days pregnant mice respond to subcutaneous serotonin (15 mg/kg) with vascular changes which lead to the death of the fetuses in 3–4 h. Pretreatment of the females withd-lysergic acid derivatives preserves the life of the young. Thus, LSD showed an ED₅₀ of only 30 g/kg, while UML (Deseril®) gave an ED₅₀ of 3.7 g/kg. PML takes an intermediate position. At a 50% efficiency level, UML (Deseril®) antagonises the effect of a 4000 times higher dose of serotonin.     

Prednisolon-Na-succinat und die Phagozytose der Leukozyten

Summary Intravenous administration of 3 mg/kg body weight prednisolon-Na-succinate (P) decreased, after 24 h, by 30% the bacterial phagocytosis of leukocytes of rabbits. After 48 h, the effect became a slight increase. At a dosis of 0.3 mg/kg, P exerts a stimulating effect which ceases within some hours. Inin vitro experiments, it showed increased or decreased action depending upon the concentration. When perfused through the liver, the efficacy of P rises strikingly.     

l-Methionin als Regulator der Spannung von Collagen

Summary Collagen fibres produce tension in hypermolar salt solutions such as 5M NaClO₄. This tension can be increased by formaldehyde 0.035M (or more). These reactions are reversible in 0.15M NaCl and can be repeated many times.l-Methionine in 0.01M solution inhibits the reversal. It is supposed that this effect is caused by inhibition of the aldol-aldimin reaction which restitutes the crosslink in the collagen fibre.     

Der Einfluss der experimentellen Leberschädigung auf Gesamtfett und ungesättigte Fettsäuren in Serum und Leber

Summary In experiments on rats with CCl₄-induced liver damage the total lipids and the di- to tetra-enoic fatty acids were estimated. The acute experiment of 72 h duration showed a strong augmentation of total lipids and a transitory diminution of arachidonic acid in serum and liver which two months after the intoxication still persisted together with a marked rise in linolenic acid. The parallelism of changes of saturation of the fatty acids in serum and liver being statistically significant indicates that the process of desaturation is most probably localized in the liver itself.     

Die Mitosehäufigkeit in der normalen Epidermis der Maus nach einmaliger DMSO-Einwirkung

Summary 12 h after local application of DMSO on the epidermis of the mouse, increased mitotic activity has been observed, the maximum occurring 24 h after the onset of the experiment. Subsequently a distinct reduction of the frequency of mitosis can be seen with a minimum occurring about 53 h after the beginning of experiment. The initial mean value is reached 12 h after.     

Zur chemischen Konstitution der herzaktiven Krötengifte

Summary Degradation of bufalin-acetate with KMnO₄ in acetone gave methyl 3--acetoxy-14-hydroxy-14-iso-ætiocholanate and 3--acetoxy-14-hydroxy-14-iso-20-keto-pregnane-21-acid-(2114)-lactone. This proves that bufalin and digitoxigenin differ only with regard to the nature of the lactone ring, the molecular structures being otherwise identical.

Versuche zum enzymatischen Abbau der «gebundenen» gibberelline vonPharbitis purpurea

Summary Seeds and fruit walls ofPharbitis purpurea contain in addition to the free gibberellins A₈, A₃, A₅, more polar gibberellin-like substances, so-called bound gibberellins tentatively namedPharbitis and. After purification by thin layer chromatography, these 2 substances were treated with both Ficin and Emulsin in buffered solutions of pH 6.2. After treatment with Emulsin, the free gibberellins A₈ and A₅, after treatment with Ficin the gibberellins A₃ and A₅, were detectable in thin layer chromatograms.     

Nachweis der Bisulfit-Addukte (α-Oxysulfonsäuren) von Carbonylverbindungen in den mit SO2 behandelten Erbsenkeimlingen

Summary The intoxication of 15-day-old green pea seedlings with 1% gaseous SO₂ causes an important concentration fall of some free ketoacids (pyruvic, oxalacetic, -ketoglutaric) and of glyceraldehyde in the roots and shoots. This fall is a consequence of the reaction of these carbonyl compounds with SO₂ in the plant tissue under the formation of bisulphite adducts (-hydroxysulfonic acids), which have been chemically proved and quantitatively estimated.     

Interaction between PGE2 and EGF receptor through MAPKs in mouse embryonic stem cell proliferation

Identifying the small molecules that permit precise regulation of embryonic stem (ES) cell proliferation should further support our understanding of the underlying molecular mechanisms of self renewal. In the present study, we showed that PGE₂ increased [³H]-thymidine incorporation in a time and dose dependent manner. In addition, PGE₂ increased the expression of cell cycle regulatory proteins, the percentage of cells in S phase and the total number of cells. PGE₂ obviously increased E-type prostaglandin (EP) receptor 1 mRNA expression level compare to 2, 3, 4 subtypes. EP1 antagonist also blocked PGE₂-induced cell cycle regulatory protein expression and thymidine incorporation. PGE₂ caused phosphorylation of protine kinase C, Src, epidermal growth factor (EGF) receptor, phosphatidylinositol 3-kinase (PI3K)/Akt phosphorylation, and p44/42 mitogen-activated protein kinase (MAPK), which were blocked by each inhibitors. In conclusion, PGE₂-stimulated proliferation is mediated by MAPK via EP1 receptor-dependent PKC and EGF receptor-dependent PI3K/Akt signaling pathways in mouse ES cells. Summary After administration of various C₂-compounds as well as leucine-2-¹⁴C toCatharanthus roseus shoots and glycine-2-¹⁴C toStrychnos nux vomica plants no specific incorporation into the non-tryptophan C_9–10 moiety of indole alkaloids was observed. The results indicate that glycine-2-¹⁴C is transformed into serine and is incorporated via tryptophan into strychnine.     

Kinetik der hepatischen Farbstoffaufnahme von Indocyaningrün. Einfluss von Bilirubin und Natriumglykocholat

Summary Kinetics of hepatic uptake of indocyanine green, a dye which is used for evaluation of liver function, were studied in the rat. The results indicate that the relationship between ICG-dose and initial hepatic dye uptake obeys Michaelis-Menten kinetics suggesting an interaction of the dye with a carrier or fixed site in the liver cell. Thus it was possible to calculate maximum ICG-uptake (v _max ) and the Michaelis constant (K _m ) of this transport system from several submaximal values.v _max was 7.65 (6-06-9.65)²² mg per 100 g liver/min and K _m 0.56 (0.31–0.81)²². Under the influence of substances which inhibit the elimination of dyes by the liver the parametersv _max and K _m showed changes which allowed characterization of the type of inhibition. While sodium glycocholate had no influence on maximum hepatic ICG-uptake and the Michaelis constant bilirubin caused a significant increase of K _m to 1.29 (0.68–1.90)²² without significantly changingv _max . These data suggest that bilirubin interferes with hepatic uptake of indocyanine green by competitive inhibition and that uptake of bile acids is dependent on a different mechanism.

Summary The measurement of meson masses in the cosmic radiation with a slowly expanding wilson chambre indicates mesons of different masses. There are at least two different masses, one between 100m ₀ and 200m ₀, and the other exceeding 200m ₀.
