Attempts to infer DNA electron transfer from fluorescence quenching measurements on DNA strands doped with donor and acceptor molecules have spurred intense debate over the question of whether or not this important biomolecule is able to conduct electrical charges. More recently, first electrical transport measurements on micrometre-long DNA 'ropes', and also on large numbers of DNA molecules in films, have indicated that DNA behaves as a good linear conductor. Here we present measurements of electrical transport through individual 10.4-nm-long, double-stranded poly(G)-poly(C) DNA molecules connected to two metal nanoelectrodes, that indicate, by contrast, large-bandgap semiconducting behaviour. We obtain nonlinear current-voltage curves that exhibit a voltage gap at low applied bias. This is observed in air as well as in vacuum down to cryogenic temperatures. The voltage dependence of the differential conductance exhibits a peak structure, which is suggestive of the charge carrier transport being mediated by the molecular energy bands of DNA.  相似文献

Gene transfer from the chloroplast to the nucleus has occurred over evolutionary time. Functional gene establishment in the nucleus is rare, but DNA transfer without functionality is presumably more frequent. Here, we measured directly the transfer rate of chloroplast DNA (cpDNA) into the nucleus of tobacco plants (Nicotiana tabacum). To visualize this process, a nucleus-specific neomycin phosphotransferase gene (neoSTLS2) was integrated into the chloroplast genome, and the transfer of cpDNA to the nucleus was detected by screening for kanamycin-resistant seedlings in progeny. A screen for kanamycin-resistant seedlings was conducted with about 250,000 progeny produced by fertilization of wild-type females with pollen from plants containing cp-neoSTLS2. Sixteen plants of independent origin were identified and their progenies showed stable inheritance of neoSTLS2, characteristic of nuclear genes. Thus, we provide a quantitative estimate of one transposition event in about 16,000 pollen grains for the frequency of transfer of cpDNA to the nucleus. In addition to its evident role in organellar evolution, transposition of cpDNA to the nucleus in tobacco occurs at a rate that must have significant consequences for existing nuclear genes.  相似文献

The function of DNA during oxidative stress and its suitability as a potential building block for molecular devices depend on long-distance transfer of electrons and holes through the molecule, yet many conflicting measurements of the efficiency of this process have been reported. It is accepted that charges are transported over long distances through a multistep hopping reaction; this 'G-hopping' involves positive charges moving between guanines (Gs), the DNA bases with the lowest ionization potential. But the mechanism fails to explain the persistence of efficient charge transfer when the guanine sites are distant, where transfer rates do not, as expected, decrease rapidly with transfer distance. Here we show experimentally that the rate of charge transfer between two guanine bases decreases with increasing separation only if the guanines are separated by no more than three base pairs; if more bridging base pairs are present, the transfer rates exhibit only a weak distance dependence. We attribute this distinct change in the distance dependence of the rate of charge transfer through DNA to a shift from coherent superexchange charge transfer (tunnelling) at short distances to a process mediated by thermally induced hopping of charges between adenine bases (A-hopping) at long distances. Our results confirm theoretical predictions of this behaviour, emphasizing that seemingly contradictory observations of a strong as well as a weak influence of distance on DNA charge transfer are readily explained by a change in the transfer mechanism.  相似文献

Measurements of magnetic noise emanating from ferromagnets owing to domain motion were first carried out nearly 100 years ago, and have underpinned much science and technology. Antiferromagnets, which carry no net external magnetic dipole moment, yet have a periodic arrangement of the electron spins extending over macroscopic distances, should also display magnetic noise. However, this must be sampled at spatial wavelengths of the order of several interatomic spacings, rather than the macroscopic scales characteristic of ferromagnets. Here we present a direct measurement of the fluctuations in the nanometre-scale superstructure of spin- and charge-density waves associated with antiferromagnetism in elemental chromium. The technique used is X-ray photon correlation spectroscopy, where coherent X-ray diffraction produces a speckle pattern that serves as a 'fingerprint' of a particular magnetic domain configuration. The temporal evolution of the patterns corresponds to domain walls advancing and retreating over micrometre distances. This work demonstrates a useful measurement tool for antiferromagnetic domain wall engineering, but also reveals a fundamental finding about spin dynamics in the simplest antiferromagnet: although the domain wall motion is thermally activated at temperatures above 100 K, it is not so at lower temperatures, and indeed has a rate that saturates at a finite value-consistent with quantum fluctuations-on cooling below 40 K.  相似文献

The wavefunction is the complex distribution used to completely describe a quantum system, and is central to quantum theory. But despite its fundamental role, it is typically introduced as an abstract element of the theory with no explicit definition. Rather, physicists come to a working understanding of the wavefunction through its use to calculate measurement outcome probabilities by way of the Born rule. At present, the wavefunction is determined through tomographic methods, which estimate the wavefunction most consistent with a diverse collection of measurements. The indirectness of these methods compounds the problem of defining the wavefunction. Here we show that the wavefunction can be measured directly by the sequential measurement of two complementary variables of the system. The crux of our method is that the first measurement is performed in a gentle way through weak measurement, so as not to invalidate the second. The result is that the real and imaginary components of the wavefunction appear directly on our measurement apparatus. We give an experimental example by directly measuring the transverse spatial wavefunction of a single photon, a task not previously realized by any method. We show that the concept is universal, being applicable to other degrees of freedom of the photon, such as polarization or frequency, and to other quantum systems--for example, electron spins, SQUIDs (superconducting quantum interference devices) and trapped ions. Consequently, this method gives the wavefunction a straightforward and general definition in terms of a specific set of experimental operations. We expect it to expand the range of quantum systems that can be characterized and to initiate new avenues in fundamental quantum theory.  相似文献

When fluctuating fields are confined between two surfaces, long-range forces arise. A famous example is the quantum-electrodynamical Casimir force that results from zero-point vacuum fluctuations confined between two conducting metal plates. A thermodynamic analogue is the critical Casimir force: it acts between surfaces immersed in a binary liquid mixture close to its critical point and arises from the confinement of concentration fluctuations within the thin film of fluid separating the surfaces. So far, all experimental evidence for the existence of this effect has been indirect. Here we report the direct measurement of critical Casimir force between a single colloidal sphere and a flat silica surface immersed in a mixture of water and 2,6-lutidine near its critical point. We use total internal reflection microscopy to determine in situ the forces between the sphere and the surface, with femtonewton resolution. Depending on whether the adsorption preferences of the sphere and the surface for water and 2,6-lutidine are identical or opposite, we measure attractive and repulsive forces, respectively, that agree quantitatively with theoretical predictions and exhibit exquisite dependence on the temperature of the system. We expect that these features of critical Casimir forces may result in novel uses of colloids as model systems.  相似文献

The cochlear travelling wave is fundamental to the ability of the mammalian auditory system to resolve frequency. The seashell-shaped outer bone of the cochlea (the auditory inner ear) contains a spiral of cochlear fluid and the sensory tissue known as the cochlear partition. Sound travels down the ear canal to the eardrum, causing its flexible tympanic membrane to vibrate. This vibration is transmitted to the cochlea via the ossides. Motion of the stapes (the stirrup ossicle) sets the cochlear fluid in motion, which in turn sets the cochlear partition near the states in motion. The motion of the cochlear partition ripples down the cochlear spiral as a travelling wave, stimulating the cochlea's sensory hair cells. The wave peaks near the base (the stapes end) of the cochlea for high frequency tones and near the apex for low frequencies. The fundamental elements of the cochlear travelling wave are fluid pressure and motion and partition forces and motion. However, the wave's direct experimental study has to date relied almost solely on measurements of the partition motion. Here I report finely spaced measurements of intracochlear pressure close to the partition, which reveal the fluid component of the cochlear wave. The penetration depth of the wave is very limited, approximately 15 microm. Over a range of frequencies at least an octave wide, the depth is independent of frequency.  相似文献

The study of cell locomotion is fundamental to such diverse processes as embryonic development, wound healing and metastasis. Since microtubules play a role in establishing the leading lamellum and maintaining cell polarity, it is important to understand their dynamic behaviour. In vitro, subunits exchange with polymer by treadmilling and by dynamic instability. Disassembly events can be complete (catastrophic) or incomplete (tempered). In vivo, microtubules are in dynamic equilibrium with subunits with a half-time for turnover of 4-20 min. Microtubules grow by elongation of their ends and are replaced one by one with turnover being most rapid at the periphery. Although previous results are consistent with dynamic instability, we sought to directly test the mechanism of turnover. Direct observations of fluorescent microtubules in the fibroblast lamellum show that individual microtubules undergo rounds of assembly and disassembly from the same end. Reorganization of the microtubule network occurs by a tempered mode of dynamic instability.  相似文献

为了研究燃烧源产生细颗粒在过饱和条件下的长大特性,设计了基于冷气流与热水接触形成过饱和环境的生长管来促进细颗粒的长大.利用热风保温的方式防止测量窗口水汽的凝结,从而实现了直接测量细颗粒长大后的液滴粒径分布.利用实验系统研究了热水温度、初始颗粒浓度、气体流量对细颗粒长大特性的影响.实验结果表明:热水温度的上升对细颗粒的长大影响非常明显;而细颗粒的初始浓度增加则不利于细颗粒的长大;气体流量的增大在一定范围内对细颗粒的长大影响较小,而流量大于4 L/min时,则影响较大.在最优的实验条件下,平均粒径为0.197μm的细颗粒长大为5.411μm.  相似文献

Dynamical processes are commonly investigated using laser pump-probe experiments, with a pump pulse exciting the system of interest and a second probe pulse tracking its temporal evolution as a function of the delay between the pulses. Because the time resolution attainable in such experiments depends on the temporal definition of the laser pulses, pulse compression to 200 attoseconds (1 as = 10(-18) s) is a promising recent development. These ultrafast pulses have been fully characterized, and used to directly measure light waves and electronic relaxation in free atoms. But attosecond pulses can only be realized in the extreme ultraviolet and X-ray regime; in contrast, the optical laser pulses typically used for experiments on complex systems last several femtoseconds (1 fs = 10(-15) s). Here we monitor the dynamics of ultrafast electron transfer--a process important in photo- and electrochemistry and used in solid-state solar cells, molecular electronics and single-electron devices--on attosecond timescales using core-hole spectroscopy. We push the method, which uses the lifetime of a core electron hole as an internal reference clock for following dynamic processes, into the attosecond regime by focusing on short-lived holes with initial and final states in the same electronic shell. This allows us to show that electron transfer from an adsorbed sulphur atom to a ruthenium surface proceeds in about 320 as.  相似文献

The spin lifetime and diffusion length of electrons are transport parameters that define the scale of coherence in spintronic devices and circuits. As these parameters are many orders of magnitude larger in semiconductors than in metals, semiconductors could be the most suitable for spintronics. So far, spin transport has only been measured in direct-bandgap semiconductors or in combination with magnetic semiconductors, excluding a wide range of non-magnetic semiconductors with indirect bandgaps. Most notable in this group is silicon, Si, which (in addition to its market entrenchment in electronics) has long been predicted a superior semiconductor for spintronics with enhanced lifetime and transport length due to low spin-orbit scattering and lattice inversion symmetry. Despite this promise, a demonstration of coherent spin transport in Si has remained elusive, because most experiments focused on magnetoresistive devices; these methods fail because of a fundamental impedance mismatch between ferromagnetic metal and semiconductor, and measurements are obscured by other magnetoelectronic effects. Here we demonstrate conduction-band spin transport across 10 mum undoped Si in a device that operates by spin-dependent ballistic hot-electron filtering through ferromagnetic thin films for both spin injection and spin detection. As it is not based on magnetoresistance, the hot-electron spin injection and spin detection avoids impedance mismatch issues and prevents interference from parasitic effects. The clean collector current shows independent magnetic and electrical control of spin precession, and thus confirms spin coherent drift in the conduction band of silicon.  相似文献

The generation, manipulation and detection of spin-polarized electrons in nanostructures define the main challenges of spin-based electronics. Among the different approaches for spin generation and manipulation, spin-orbit coupling--which couples the spin of an electron to its momentum--is attracting considerable interest. In a spin-orbit-coupled system, a non-zero spin current is predicted in a direction perpendicular to the applied electric field, giving rise to a spin Hall effect. Consistent with this effect, electrically induced spin polarization was recently detected by optical techniques at the edges of a semiconductor channel and in two-dimensional electron gases in semiconductor heterostructures. Here we report electrical measurements of the spin Hall effect in a diffusive metallic conductor, using a ferromagnetic electrode in combination with a tunnel barrier to inject a spin-polarized current. In our devices, we observe an induced voltage that results exclusively from the conversion of the injected spin current into charge imbalance through the spin Hall effect. Such a voltage is proportional to the component of the injected spins that is perpendicular to the plane defined by the spin current direction and the voltage probes. These experiments reveal opportunities for efficient spin detection without the need for magnetic materials, which could lead to useful spintronics devices that integrate information processing and data storage.  相似文献

大位移井在深部地层的油气藏开采中得到了广泛应用,但同时面临着井眼清洁不充分、托压、卡钻等一系列问题,为了更好地指导超深大位移井的井眼清洁工作,需要进一步对超深大位移井的长裸眼段的岩屑动态运移特性进行研究。建立了考虑实际井眼轨迹、流体压降及悬浮层颗粒扩散的大位移井两层岩屑运移动态模型。该模型利用有限差分法进行求解,并利用已有文献的实验结果进行验证。对钻井和冲洗两种工况进行模拟研究,认识钻井液排量、钻井液密度、机械钻速、井眼半径等方面对岩屑运移的影响。研究结果表明:在钻井工况下,井斜角越小的井段,岩屑越难以沉积,且钻井液排量越高、钻井液密度越高、井眼尺寸越小、钻速越低,岩屑运移的效率越高;在冲洗工况下,井斜角越小,冲蚀效果越差,且钻井液排量越高、钻井液密度越高、井眼尺寸越小,岩屑运移的效率越高。研究结果对保障超深大位移井良好的井眼清洁具有指导意义。  相似文献

All-inorganic quantum dot light emitting diodes (QLEDs) have gained great attention as a result of their high stability under oxygen-rich, humid and high current working conditions. In this work, we have fabricated an all-inorganic QLED device (FTO/NiO/QDs/AZO/Ag) with sandwich-structure, wherein the inorganic metal oxides thin films of NiO and AZO were employed as hole and electron transport layers, respectively. The porous NiO layer with vertical lamellar nanosheets interconnected microstructure have been directly synthesized on the substrate of conductive FTO glass and increased the wettability of CdSe@ZnS QDs, which result in an enhancement of current transport performance of the QLED.  相似文献

以湘江流域为研究区域,选取县级及以上城市为评价单元,运用GIS分析技术从公路网密度、邻近度、通达性3方面分别对各评价单元的差异进行分析,并利用以上3个指标构建区域交通优势度综合评价模型。分析湘江流域交通优势度的空间分布格局和特征以及其形成成因.研究表明:湘江流域路网密度整体较低,城市间差异较大;邻近度和通达性以地级市为中心呈圈层分布;城市间交通优势度极化严重,交通优势度较高的地区主要在长株潭城市圈、衡阳市及周边地区:整个流域交通优势度成“H”字型分布格局.  相似文献

Absorption of ultraviolet light by DNA is known to lead to carcinogenic mutations, but the processes between photon absorption and the photochemical reactions are poorly understood. In their study of the excited-stated dynamics of model DNA helices using femtosecond transient absorption spectroscopy, Crespo-Hernández et al. observe that the picosecond component of the transient signals recorded for the adenine-thymine oligonucleotide (dA)18.(dT)18 is close to that for (dA)18, but quite different from that for (dAdT)9.(dAdT)9; from this observation, they conclude that excimer formation limits excitation energy to one strand at a time. Here we use time-resolved fluorescence spectroscopy to probe the excited-state dynamics, which reveals the complexity of these systems and indicates that the interpretation of Crespo-Hernández et al. is an oversimplification. We also comment on the pertinence of separating base stacking and base pairing in excited-state dynamics of double helices and question the authors' assignment of the long-lived signal component found for (dA)18.(dT)18 to adenine excimers.  相似文献
