转甲状腺素蛋白基在毕赤氏酵母中的非融合表达

用限制酶将pSK-TTR质粒上TTR基因切下，分别插入分泌型载体pPIC9和pPIC9K，将重组的pPIC9K-TTR用BglⅡ酶切后，转化pichia pastoris GS115菌株，筛选获得G418高抗性转化子，转化子经摇瓶发酵，上清液用SDS-PAGE检测，有明显的rTTR表达，经DEAE-sepharose F.F.离子交换柱和Superdex75分子筛层析，得到蛋白线纯度大于95%的rTTR，经Western Boltting，分子质量和等电点测定等证明，毕赤氏酵母的表达rTTR与人血浆提取的TTR极为相似。     

人载脂蛋白基因apoAⅠ在毕赤氏酵母中的表达

将化学法合成的apoAⅠ基因插入分泌型载体 pPIC9K .将重组的 pPIC9K apoAⅠ用BglⅡ酶切后 ,转化PichiapastorisGS115菌株 ,筛选获得G4 18高抗性转化子 .转化子经摇瓶发酵和甲醇诱导 ,上清液用SDS PAGE检测 ,有明显的rApoAⅠ表达 .经Phenyl sepharose 6 (FastFlow)疏水层析柱和SephadexG 5 0 (coarse)分子筛层析 ,得到rApoAⅠ蛋白纯品 .经Westernblotting ,N末端氨基酸顺序测定证明 ,毕赤氏酵母表达的rApoAⅠ与人血浆提取的ApoAⅠ基本一致 .     

人载脂蛋白基因apoAI在毕赤氏酵母中的表达

将化学法合成的apoAI基因插入分泌型载体pPIC9K．将重组的pPIC9K—apoAI用BglⅡ酶切后，转化Pichia pastoris GS115菌株，筛选获得G418高抗性转化子．转化子经摇瓶发酵和甲醇诱导，上清液用SDS-PAGE检测，有明显的rApoAI表达．经Phenyl—sepharose 6(Fast Flow)疏水层析柱和Sephadex G-50(coarse)分子筛层析，得到rApoAI蛋白纯品．经Western blotting，N末端氮基酸顺序测定证明，毕赤氏酵母表达的rApoAI与人血浆提取的ApoAI基本一致．     

人骨形态发生蛋白6(hBMP6)在毕赤酵母中的分泌表达

以重组质粒pcDNA6A-BMP6为模板PCR扩增hBMP6成熟肽cDNA编码序列,将该序列克隆到酵母分泌表达表达载体pPIC9K中.重组质粒pPIC9K-hBMP6经SacI酶切线性化,电击转化毕赤酵母GS115,PCR法筛选阳性转化子,利用梯度浓度G418筛选到七株高拷贝整合的阳性转化子。用甲醇进行诱导表达,SDSPAGE及Western-blot检测结果表明hBMP6成熟肽以分子量约为34和18 kD两种不同糖基化修饰的单体形式存在,具有良好的免疫原性.     

人载脂蛋白基因apoAⅠ在毕赤氏酵母中的胞内表达

将PCR扩增得到的apoAⅠ基因片段插入胞内表达质粒pPIC3.5K,重组的pPIC3.5K apoAⅠ用BglⅡ酶切后,转化PichiapastorisGS115菌株,筛选获得G418高抗性转化子,再通过PCR鉴定证实apoAⅠ基因已整合到染色体上.其转化子经摇瓶发酵和甲醇诱导,收集细胞裂解后,用SDS PAGE与Westernblotting检测,证明胞内有明显的rApoAⅠ表达,其分子质量与人血浆提取的ApoAⅠ相同.     

表达人胰岛素前体的毕赤酵母菌株的构建

将人胰岛素前体 (HIP)基因插入到毕赤酵母Pichiapastoris的分泌表达质粒pPIC9K中,得到分泌表达质粒pPIC9K/HIP并用电转化法转化P.pastorisGS115。筛选出整合型His⁺Mut^s 菌株,进一步用G418筛选获得高拷贝转化子。经诱导培养后,SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)证明HIP在毕赤酵母中能有效分泌表达。对毕赤酵母中外源基因的稳定性进行了研究,结果表明外源基因在毕赤酵母中很稳定.     

重组质粒βhCG-pPIC9K的构建及嗜甲醇酵母(Pichia pastoris)的转化

目的 :为用基因重组技术表达hCG避孕疫苗抗原 ,构建在酵母细胞中表达的重组质粒βhCG -pPIC9K ,转化嗜甲醇酵母 .方法 :根据βhCG的cDNA序列设计两条引物 ,使上游带EcoRI酶切位点 ,下游带NotI酶切位点 ,以质粒 βhCG -PBSKS为模板 ,进行PCR扩增反应 ;将所得的DNA片段经EcoRI和NotI双酶切后用T4连接酶与pPIC9K质粒进行连接 ,然后导入大肠杆菌DH5α ,用PCR筛选阳性克隆并用双酶切鉴定重组子βhCG -pPIC9K .再用电打孔法转化酵母 (Pichiapastoris) ,用缺组氨酸MD平板筛选阳性株并在含G4 18的YPD培养基中筛选多拷贝插入菌株 .结果 :用所设计的引物扩增出两端分别带EcoRI和NotI酶切位点的βhCGDNA片段 ,插入pPIC9K并导入DH5α获得阳性克隆 ,经PCR反应和双酶切鉴定表明重组质粒是 βhCG -pPIC9K ,并转化嗜甲醇酵母获得耐受 4mg/mLG4 18的菌株 .结论 :构建了重组质粒 βhCG -pPIC9K并获得插入重组质粒 βhCG -pPIC9K的嗜甲醇酵母     

人血管抑制素基因克隆、表达和生物活性分析

应用PCR技术从人胎肝cDNA库中扩增了人血管抑制素基因。将克隆的基因重组进酵母质粒pPIC9K获得含该基因的重组质粒pPIC9KA3。用电激法将质粒pPIC9K转化毕节酵母GSll5，经PCR检测获得含人血管抑制素基因的酵母工程菌GSll5(pPIC8KA3)。再用G418筛选法，在含不同浓度的G418平板上筛选高拷贝整合的转化子。对高拷贝整合的转化子进行发酵培养和诱导表达。SDS—PAGE及Westem印迹分析显示：表达产物约占胞外蛋白的43％，相当于94mg/L，并具有免疫活性。并能抑制bFGF诱导的鸡胚尿囊膜新生血管的生成。还对用G418筛选高拷贝整合转化子的方法做了探索。     

人载脂蛋白基因αpoAⅠ在毕赤氏酵母中的胞内表达

将PCR扩增得到的αpoAⅠ基因片段插入胞内表达质粒pPIC3．5K，重组的pPIC3．5K-αpoAⅠ用BglⅡ酶切后，转化Pichia pastoris GS115菌株，筛选获得G418高抗性转化子，再通过PCR鉴定证实口αpoAⅠ基因已整合到染色体上．其转化子经摇瓶发酵和甲醇诱导，收集细胞裂解后，用SDS-PAGE与Western blotting检测，证明胞内有明显的rApoAⅠ表达，其分子质量与人血浆提取的ApoAⅠ相同．     

黑鲷抗菌肽hepcidin在毕赤酵母中的表达及其抗菌活性

鱼类抗菌肽hepcidin具有广谱抗菌活性.利用PCR技术扩增获得了黑鲷抗菌肽hepcidin (AS-hepc2)的前体肽和成熟肽 cDNA 序列,片段大小约250 bp.将其与毕赤酵母(Pichia pastoris) pPIC9K 质粒连接,构建了分泌表达载体pPIC9K/AS-hepc2 .电击法转化重组表达载体,经过G418筛选和选择性培养基以及PCR鉴定,得到的克隆子都为Mut+.以甲醇诱导pPIC9K/AS-hepc2,分泌表达上清中获得10 ku左右的表达产物,与预期的目的蛋白黑鲷hepcidin的前体肽和成熟肽的大小相符.表达试验证明,随时间的延长,表达产物量增多,至120 h 达到高峰.表达产物具有很强的热稳定性.用抑菌圈法测定重组蛋白Pro-AS-hepc2 的抗菌活性,发现能抑制革兰氏阳性菌和革兰氏阴性菌的生长.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
