植酸酶液体发酵条件及酶学性质的研究

植酸酶产生菌黑曲霉 90 3(Aspergillusniger)的最适产酶条件 :初始pH5 .0～ 6 .5 ,温度2 9± 1℃ ,培养时间 4d ,2 5 0ml三角瓶发酵液的装量为 5 0ml,一定量的Fe2 +和Mn2 +对产酶有促进作用 ;酶的最适作用pH为 5 .5～ 6 .0 ,最适作用温度为 37℃ ,该酶在 pH3.0～ 7.0 ,温度 30～ 5 0℃时稳定性较好。     

羔羊皱胃酶制备工艺和部分性质的研究

文章利用羔羊皱胃制备了凝乳酶,并对该酶的性质进行探讨．结果发现：超滤和真空干燥适用于羔羊皱胃酶的制备;羔羊皱胃酶的最适凝乳温度是55℃,最适酶pH是6．0,最适底物pH为5．0,最适底物浓度是100g·L^-1,最适钙离子浓度是8mmol·L^-1．     

褐色高温单孢菌PE-211产纤维素酶的酶学特性研究

对褐色高温单孢菌PE-211(Thermomomospora fuscd PE-211)形态学和生物学特征进行了研究,该菌适宜在pH8．0～10．0、55-60℃条件下生长．初步研究了该菌产生的纤维素酶(CMCase)的一些基本性质,结果表明．CMCase最适作用温度为75℃,最适作用pH为9．0,Ca^2 和Mn^2 对酶反应有促进作用,Pb^2 和Hg^2 时酶反应有抑制作用．这种酶制剂在洗涤剂工业中具有良好的应用前景．     

固定化脂肪酶的动力学研究

以壳聚糖为载体,用物理吸附固定化脂肪酶,对影响固定化过程的各种因素进行了考察,确定了最优条件,并比较了游离酶和固定化酶的最适温度、pH、酶的热稳定性以及表观Km。固定化酶水解的最适pH(pH7．0)稍低于游离酶(pH8．0),最适温度为50℃(游离酶为40℃)。固定化酶在50℃的半衰期(0．96h)是游离酶(0．32h)的3倍,在4℃的冰箱中贮存3个月活力变化很小。     

产脂肪酸菌种的筛选及部分酶学性质

从含油污泥中筛选出一株产脂肪酶活力较高的微球菌,此菌最适产酶条件：2％的蛋白胨为氮源,1％的橄榄油为碳源,pH8.0,28℃培养48h．酶学性质研究发现,在28℃和45℃脂肪酶有较高催化活力,其中28℃活力更高些,最佳反应pH为8．0,pH稳定范围在6．0～10．0之间,Ca2 和K 对酶有激活作用,Fe2 ,Cu2 和Mn2 等对酶有抑制作用．     

一种改进的漆酶酶活检测方法

以 2 ,2′ -连氮 -双 (3-乙基苯并噻唑 - 6 -磺酸铵 ) (简称ABTS)为漆酶A、B的底物 ,采用UV - 2 2 0 1型紫外 -可见分光光度计连续记录酶反应在 2 5℃ ,4 2 0nm波长下的时间历程曲线 ,并求取最初部分的斜率以计算漆酶酶活的方法 .该方法直观、方便、准确且可靠 .在 2 5℃的 0 .0 4mol/LBritton Robinson缓冲溶液中 ,漆酶A、B的最适 pH值分别为 4 .0和 4 .5 .在 2 5℃最适 pH值的Britton Robinson缓冲液中 ,由双倒数作图法求得漆酶A、B的米氏常数Km 分别是 6 .6 4× 1 0 - 5mol/L和 2 .2 1× 1 0 - 5mol/L ,漆酶A、B的最大米氏反应速度Vmax分别是 1 4 2 5μmol.M (L·min)和 32 5μmol.M (L·min) .漆酶A、B的酶活分别为 2 2 99μmol·min- 1·L- 1和 1 1 4 8μmol·min- 1·L- 1.     

红酵母菌产胡萝卜素的初步研究

报道一株分离于自然界的红酵母菌株 ,通过正交实验确定了菌株产生胡萝卜素的最佳培养基 ,并优化了菌株的培养条件 :温度 2 8℃和最佳pH 6.0及最适培养时间 3d ,在优化培养条件下色素产量为 0 .2 6mg/干细胞 (g) .     

糙皮侧耳Ax3产漆酶条件及部分酶学性质研究

对糙皮侧耳(Pleurotus ostreatus)Ax3菌的漆酶产生条件进行了研究，该菌产漆酶较高的条件是：培养温度25℃，培养基pH5．5，每升C／N含量0．252g／0．021g；加入0．05％～0．1％的吐温可提高漆酶酶活12倍。对漆酶的部分生化性质研究表明：最适反应温度25℃；最适反应pH2．8；琥珀酸缓冲液效果最好；金属离子：Mn^2 ，Cu^2 ，Zn^2 对漆酶有较强的激活作用，其中Mn^2 相对活性最高，Ag^ ，K^ ，Ca^2 ，Fe^2 对漆酶活性有抑制能力，其中Ag^ 的抑制作用明显。     

碱性脂肪酶产生菌的筛选与产酶条件及酶学性质研究

作者从泸州油脂厂附近的土壤中,筛选到一株产碱性脂肪酶活性较高的细菌,经初步鉴定为假单胞菌(Pseudomonassp.).经对该菌株进行产酶条件和酶学性质研究发现,该菌株的最适产酶发酵培养基为(%):酵母浸出汁1.5,黄豆粉1,NaCl0.2,K2HPO40.2,NaH2PO40.2,MgSO4·7H2O0.01,初始pH8.5.100mL三角瓶装液10mL,30℃,150r/min摇床培养48h,酶活最高可达19.1U/mL.酶的最适反应温度40℃,最适pH8.5,该酶具有较好的热稳定性.     

一种麦芽酸性磷酸酶的分离纯化及部分性质研究

经硫酸铵分级沉淀、CM-Sepharose CL 4B和DEAE-Sepharose CL 4B离子交换柱层析等步骤,从麦芽中分离提纯到一种酸性磷酸酶(ACPase,E．C．3．1．3．2),纯化倍数为33．06,酶液比活为88．27 U/mg．通过动力学方法测得其最适pH为5．4,酸碱稳定性较差,仅在pH 6．0左右的缓冲液中较稳定;最适温度为50 ℃,40 ℃以下有较好的稳定性;在最适条件下,该酶催化pNPP的米氏常数(Km)为4．696×10-4 mol/L．同时研究了一些金属离子和有机化合物对该酶     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。

橡胶颗粒沥青混合料低温抗裂性试验研究

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.
