粉防己碱逆转人早幼粒白血病HL60而药细胞对三尖杉…

无细胞毒性浓度的粉防己碱，明显地增强三尖杉酯碱对人早幼粒白血病ＨＬ６０耐药细胞的生长抑制作用，降低集落形成率，但对敏感ＨＬ６０细胞没有增强作用，ＤＰＨ荧光标记法测定表明，粉防己碱对细胞膜流动性没有影响。     

线粒体参与了三尖杉酯碱诱导的HL-60细胞凋亡

用流式光度术研究了三尖杉酯碱诱导人早幼粒白血病ＨＬ－６０细胞及其转染了ｂｃｌ－２基因的细胞株ＨＬ－６０／Ｂｃｌ－２线粒体膜电势的变化,数据表明线粒体损伤介导了ＨＴ诱导的ＨＬ－６０细胞的凋亡过程。     

抗三尖杉酯碱的HL60细胞蛋白质磷酸化

研究了抗三尖杉酯碱的ＨＬ６０细胞蛋白质磷酸化的变化。经差速离心得到纯膜蛋白，抗性细胞有一高度磷酸化的１１０ｋｕ的蛋白质存在，免疫沉淀ｃ－ＫＡＦ－１蛋白激酶，抗性细胞内ｃ－ＲＡＦ－１蛋白激酶磷酸化程度明显提高，其活性被蛋白激酶Ｃ抑制剂ＣａｌｐｈｏｓｔｉｎＣ明显地抑制。结果表明：ＨＬ６０细胞对三尖杉酯碱的抗药性与蛋白质高度磷酸化有关；抗性细胞内ｃ－ＲＡＦ－１蛋白激酶磷酸化程度明显提高，可能与多药抗药性和抗细胞调亡有关。     

星状孢子素和金雀异黄素对人早幼粒白血病细胞系的诱导分化作用

以人早幼粒白血病细胞ＨＬ－６０细胞为模型，研究了星状孢子素和金雀异黄素对该细胞的诱导分化作用，结果表明，ＨＬ－６０细胞经２种药物分别处理后，细胞增殖能力均呈下降趋势，细胞形态学变化明显，大部分细胞趋于成熟，硝基兰四氮唑（ＮＲＴ）还原能力增强，ＮＢＴ阳性细胞百分率均达８０％以上，说明此２种药物对ＨＬ－６０细胞有明显的诱导分化作用。ＨＬ－６０细胞酸性磷酸酶活力显著升高，则揭示该细胞沿粒细胞方向趋于分化。

用对蛋白激酶具有强烈抑制、作用广泛的抑制剂ｓｔａｕｒｏｓｐｏｒｉｎｅ（Ｓｔａ），研究敏感和抗三尖杉酯碱的人白血病ＨＬ６０细胞中凋亡和多药抗药性的关系。Ｓｔａ均能诱导２种细胞发生典型的凋亡，但抗性细胞发生凋亡需更长的时间、凋亡的细胞数减少。Ｓｔａ增加柔红霉素在抗性细胞内的积聚，说明其能逆转多药抗药性，在抗性细胞凋亡过程中，ｍｄｒｌ基因表达没有变化，ｃ－ｍｙｃ稍有增加，结果显示：Ｓｔａ能诱导敏感和抗三     

高三尖杉酯碱对白血病细胞分化和肿瘤转移的影响

为研究高三尖杉酯碱（Ｈｏｍ）对白血病细胞分化和肿瘤转移的影响．采用四氮唑蓝法检测ＨＬ－６０细胞的分化及测定游离磷含量观测肿瘤细胞Ｎａ＋－Ｋ＋－ＡＴＰ酶和Ｍｇ＋＋－ＡＴＰ酶活性．肿瘤转移模型采用小鼠皮下接种Ｌｅｗｉｓ肺癌观测肺转移灶数和小鼠足跖接种Ｐ３８８白血病细胞观察动物生存期,且用γ射线测定法评价１２５Ｉ－纤维蛋白原与肿瘤细胞的结合能力．结果显示Ｈｏｍ１．５～４．５μｇ／Ｌ能诱导ＨＬ－６０细胞分化,Ｈｏｍ０．１μｍｏｌ／Ｌ２４ｈ对Ｆｒｉｅｎｄ白血病细胞Ｎａ＋－Ｋ＋－ＡＴＰ酶活性抑制,对Ｍｇ＋＋－ＡＴＰ酶活性无影响,Ｈｏｍ０．５～１．５ｍｇ·ｋｇ－１／ｄ剂量下能抑制Ｌｅｗｉｓ癌肺转移和Ｐ３８８白血病细胞淋巴道转移,以及Ｈｏｍ２～１０ｍｇ／Ｌ抑制纤维蛋白原与癌细胞的结合．提示Ｈｏｍ能通过以上途径诱导肿瘤细胞分化和抗肿瘤转移     

MTT法测定高三尖杉酯碱对人肝癌HepG2抑制作用

采用MTT法检测高三尖杉酯碱对HepG2细胞体外培养的抑制作用.将常规用96孔板培养的HepG2细胞作为空白组,其余分11个剂量的给药组,相应处理24、48、72 h.与空白组对照后,HepG2细胞的增殖被各给药组不同程度的抑制,并随着给药剂量的增加其抑制率也增加.高三尖杉酯碱在体外可有效的抑制人肝癌细胞的活性和细胞增殖.     

中国鲎鲎素T-1抗人早幼粒白血病HL-60细胞活性研究

利用从中国鲎血细胞中提取的鲎素Ｔ－１作抗肿瘤活性实验．结果表明,鲎素Ｔ－１在体外对人早幼粒白血病ＨＬ－６０细胞的增殖有明显抑制作用．急性毒理实验显示,鲎素Ｔ－１对正常活体毒性较小,可用于活体实验．荷瘤裸鼠实验统计,鲎素Ｔ－１对肿瘤抑制率达４２．３％．

粉防己碱促进增生性瘢痕成纤维细胞凋亡的实验研究

目的观察粉防己碱对培养的人增生性瘢痕成纤维细胞凋亡的影响。方法取材自人体增生性瘢痕,提取出成纤维细胞进行培养,设实验组和对照组,实验组加入粉防己碱,用Tunnel法检测细胞的凋亡,比较两组间的细胞凋亡率。结果使用粉防己碱后,成纤维细胞的凋亡率显著增加,与对照组比较,差异有非常显著意义(P<0.01)。结论通过促进瘢痕组织成纤维细胞的凋亡,粉防己碱可能具有防治瘢痕的作用,值得进一步探讨。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
