2-Carboxyethylgermanium sesquioxide (Ge-132), a synthesized organogermanium compound with immunomodulaing activities, was shown to be an inducer of anti-suppressor T cells in normal mice. The suppressor cell activity of T6S cells, a clone of burn-induced CD8+ IL-4-producing suppressor T cells, was clearly inhibited when a mixed lymphocyte-tumor cell reaction of the clone was conducted with splenic mononuclear cells from mice treated orally with a 100 mg/kg dose of Ge-132. The activity of anti-suppressor cells was demonstrated in spleens of mice 2 days after treatment with Ge-132 and reached its peak on day 3. The anti-suppressor cells induced by the compound were of a contrasuppressor T cell-linage, because they were characterized as CD4+ CD28+ TCR/+ Vicia villosa lectin-adherent T cells. These cells produced IFN- but did not produce IL-2, IL-4, IL-6 or IL-10 in their culture fluids. CD4+ anti-suppressor T cells induced by Ge-132 may be different from other subsets of CD4+ T cells because Th1 and Th2 cells generated in our laboratory did not adhere toVicia villosa lectin-coated petri dishes, and each produced specific cytokines. Th1 cells produced IFN- and IL-2 while Th2 cells produce IL-4 and IL-10 in vitro. These results suggest that Ge-132 may be useful as an inducer of contrasuppressor T cells in immunocompromised individuals bearing suppressor T cells. To eliminate suppressor T cells from immunocompromised hosts may result in improved resistance from various opportunistic infections.  相似文献

Summary Administration of LH-RH to adult male Skipper frogs resulted in marked hypertrophy and degranulation of basophils-2 (B2) in the pars distalis of the pituitary and a significant increase in their nuclear and cellular area. Concomitantly, there is a significant increase in the relative weight of the testes, in the number of cell nests containing secondary spermatogonia and primary spermatocytes, and in the nuclear diameter of the Leydig cells. There is also an increase in the 5-3-hydroxysteroid dehydrogenase and glucose-6-phosphate dehydrogenase activities in the Leydig cells. The results indicate that the B2 cells are gonadotrops and the hormone(s) secreted by B2 cells regulate the spermatogenetic and steroidogenic activity of the testis inR. cyanophlyctis.The free gift of LH-RH from NIH is gratefully acknowledged. Summary In transfilter associations ofPortulaca grandiflora callus tissues withRhizobium sp. cowpea 32H1 the nitrogenium fixed by the bacteria was channelled into the normal pathway of nitrogen metabolism of higher plants. In associations kept in an15N2 containing atmosphere 10% of the15N taken up into the plant cells was incorporated intoPortulaca proteins. The results demonstrate that cells of nonleguminous plants are profiting from the quasi-symbiotic situation.  相似文献

Summary A transfilter culture of the non-leguminous plantPortulaca grandiflora var. JR andRhizobium sp. cowpea 32H1 was established. Using15N2-analysis we demonstrated that15N-containing substances produced by the bacteria passed through the membrane and15N was enriched in the plant cells.Acknowledgment. To study the origins of biologically active substances in marine sponges, a carotenoid produced by a marine bacterium,Pseudomonas sp. strain number KK10206C, which was associated with a marine sponge,Halichondra okadai, was investigated. A visible absorption spectrum-guided isolation procedure led to the isolation of a novel C50-carotenoid, okadaxanthin. Its structure, 2,2-bis(4-hydroxy-2-methyl-2-butenyl)-,-carotene, was elucidated mainly by spectroscopic methods. Okadaxanthin turned out to be a potent singlet oxygen quencher, approximately 10 times as strong as -tocopherol.  相似文献

Summary Numerous intramitochondrial pararystalline inclusion bodies (ICIB) were observed in the germinal plasm of a mid-blastula, and in primordial germ cells (PGCs) after their migration to the germinal ridges, inRana ridibunda, R. lessonae andR. esculenta. In oogonia the number of ICIB decreases rapidly. Single ICIB are observed in the germ cells up to the leptotene stage; they have never been observed in pachytene oocytes. In diplotene oocytes that have reached a diameter of about 100 m ICIB are visible again, and their number increases concomitantly with oocyte growth.  相似文献

Summary The karyotype of the pampas fox has 2n=74 and a NF=76. Except forChrysocyon brachyurus, 2n=74 is a common diploid number for the South American Canidae. The surfaces of mammalian cells are covered by a variety of carbohydrates linked to proteins and lipids. N-glycans are commonly found carbohydrates in plasma membrane proteins. The structure and biosynthetic pathway of N-glycans have been analyzed extensively. However, functional analysis of cell surface N-glycans is just under way with recent studies of targeted disruption of genes involved in N-glycan synthesis. This review briefly introduces the potential role of processing -mannosidases in N-glycan biosynthesis and recent findings derived from the -mannosidase IIx (MX) gene knockout mouse, which shows male infertility. Thus, the MX gene knockout experiment unveiled a novel function of specific N-glycan, which is N-acetylglucosamine-terminated and fucosylated triantennary structure, in the adhesion between germ cells and Sertoli cells. Analysis of the MX gene knockout mouse is a good example of a multidisciplinary approach leading to a novel discovery in the emerging field of glycobiology.Received 29 November 2002; received after revision 30 December 2002; accepted 20 January 2003  相似文献

Summary The photoproduction of ammonia by cells of the heterocystous cyanobacteriumCyanospira rippkae in the presence of the glutamine synthetase inhibitor, L-methionine D,L-sulfoximine (MSX), was investigated. The time course of changes in protein, pigment and carbohydrate concentrations and the C2H2-reducing activity of nitrogenase in MSX treated and untreated filament suspensions was also determined. The results show that nearly 40 h after MSX addition the cells are able to recover from the nitrogen starvation induced by the inhibitor by themselves, without the removal of MSX or the addition of nitrogenous compounds. Biliproteins, mobilized as a consequence of MSX addition, seem to play a key role in the process of cell recovery. These findings were exploited in a semicontinuous ammonia producing process with cells immobilized in a dialysis tube photobioreactor.  相似文献

Summary In leaning stems, reaction wood can be formed as a response to gravitation stimulus. In seedlings ofAesculus hippocastanum, reaction wood formation could likewise be stimulated by centrifugation. This proved the differentiation of wood cells to be affected by mass acceleration. The reaction wood formation is related quantitatively to mass accelerationb and timet, the formation of reaction wood being described by quantitative stimuli law as . Summary The -amylase of mycelial cells ofAspergillus oryzae exists in a particular form in 8000 g pellet. The lysosomal localization of acid phosphatase is confirmed by electron microscopy. Summary Two new butenolides, seiridin andiso-seiridin, were isolated from culture filtrates ofSeiridium cardinale, the pathogen of cypress canker, a destructive disease ofCupressus and relatedConiferae These metabolites were characterized as 3-methyl-4-(2-hydroxyheptyl)-2(5H)-furanone and its 4-(3-hydroxyheptyl) isomer, respectively. Chlorotic, and necrotic symptoms were produced on leaves of either host or non-host test plants by absorption of 0.3 mg/ml solutions of either compound. The sensitivity ofDeleya halophila to oxidative stress caused by hydrogen peroxide (H2O2) was found to vary, depending on the NaCl concentration of the growth medium. Pretreatment of the bacteria at a low concentration of H2O2 (50 M) protected the cells against the lethal effects of higher levels (1–2 mM) of H2O2. Exposure ofD. halophila cells to 50 M H2O2 resulted in the induction of several proteins (hydrogen peroxide-inducible proteins, hips). However, the kinetics of induction, the extent of induction and the number of hips appear to be influenced by the salt concentration of the growth medium. Five of the hips exhibited apparent molecular masses identical to those of five heat shock proteins (hsps).  相似文献

Thrombospondin (TSP) is a multifunctional glycoprotein which is synthesised by several cell types including osteoblasts, and incorporated into the extracellular matrix (ECM) of these cells. The function and regulation of TSP in bone is not clear. In this study, using a long term culture model of human osteoblast-like cells, we examined the distribution of TSP in the ECM and its modulation by added estradiol. In this model the osteoblast-like cells form a regular multilayer which continues to increase in depth up to 50 days post confluence. In the ECM of these cultures and in 19-week fetal bone, the bone markers osteocalcin and alkaline phosphatase were diffusely distributed in the matrix. In contrast, labelling for TSP was concentrated, confined to the banded collagen and its immediately adjacent ECM. This pattern of labelling resembled that of the growth factors transforming growth factor-I (TGF), and insulin-like growth factor-I (IGF-I), with which TSP label co-localised. Labelling intensities were comparable between fetal bone and the in vitro material for TSP, TGF and IGF-I. TSP label was present by 10 days post confluence, reached a maximum by 20 days, and declined slowly thereafter, a time course which was similar to that of IGF-I. Incubation of osteoblast-like cell cultures with 17 estradiol resulted in an increase in multilayer depth and a maximal 3-fold increase in TSP labeling at 30 days as well as approximately 2-fold increases for TGF and IGF-I. The dose-response relationship for these responses to estradiol treatment was biphasic with maximal increases at 10–10 M–10–11 M of added estradiol. Treatment with 17 estradiol produced labelling intensities that were not significantly different from controls. Studies with other cell types have suggested that TSP may be involved in modulation of growth factor activity. The similarities between TSP, TGF and IGF-I, in terms of their distribution and regulation by 17 estradiol treatment, may indicate a role for TSP in modulating bone cell proliferation and function through interaction with local growth factors.  相似文献

Cyclins are regulatory proteins that interact with cyclin-dependent kinases (CDKs) to control progression through the cell cycle. In Arabidopsis thaliana, 34 cyclin genes have been described, grouped into five different types (A, B, D, H, and T). A novel class of seven cyclins was isolated and characterized in Arabidopsis, designated P-type cyclins (CYCPs). They all share a conserved central region of 100 amino acids (cyclin box) displaying homology to the corresponding region of the PHO80 cyclin from Saccharomyces cerevisiae and the related G1 cyclins from Trypanosoma cruzi and T. brucei. The CYCP4;2 gene was able to partially re-establish the phosphate-dependent expression of the PHO5 gene in a pho80 mutant strain of yeast. The CYCPs interact preferentially with CDKA;1 in vivo and in vitro as shown by yeast two-hybrid analysis and co-immunoprecipitation experiments. P-type cyclins were mostly expressed in proliferating cells, albeit also in differentiating and mature tissues.
