Effects of bunaphtine on45Ca movements in rat aortic smooth muscle

Summary The effect of bunaphtine (BNA, 5×10^–5 M) on La³⁺-resistant⁴⁵Ca content and⁴⁵Ca efflux was studied on rat aortic smooth muscle. BNA decreased both control and norepinephrine-stimulated, La³⁺-resistant⁴⁵Ca content and increased the⁴⁵Ca efflux. These effects could explain the inhibition of the contractile responses induced by BNA.

The effect of bunaphtine (BNA, 5 X 10(-5)M) on La3+ -resistant 45Ca content and 45Ca efflux was studied on rat aortic smooth muscle. BNA decreased both control and norepinephrine-stimulated La3+-resistant 45Ca content and increased the 45Ca efflux. These effects could explain the inhibition of the contractile responses induced by BNA.

Zusammenfassung Der Ca-Gehalt von isolierter Längs-muskulatur des Meerschweinchens hängt stark von der Ca-Konzentration der Tyrode-Lösung ab (untersuchter Bereich 0–2,7 mM). Nach schrittweiser Änderung der extracellulären Ca-Konzentration erreicht der zelluläre Ca-Gehalt nach etwa 20 Minuten seinen neuen Gleichgewichtswert.     

Effects of disodium EDTA on the cardiovascular responses to prostaglandin E1

Zusammenfassung Die Wirkung des Dinatrium-EDTA auf die Kreislaufreaktionen des Prostaglandin E₁ (PGE₁) wurde an narkotisierten Hunden untersucht. Das Ausmass der positiv chronotropen und inotropen Einflüsse des PGE₁ war während der Infusion von EDTA bedeutend geringer als das des PGE₁ vor der EDTA-Gabe. Die Gegenwart oder das Einströmen von Kalziumionen scheint in der pharmakologischen Wirkung des PGE₁ eine Rolle zu spielen.     

Differential effects of ethanol on prostaglandin responses of arterial and venous smooth muscle

Summary The present results, using isolated rat aortic strips and portal vein segments, demonstrate that ethanol, depending upon concentration, can either enhance or attenuate the contractile actions of PGF₂ on at least 2 different types of vascular smooth muscle. At the very least, the present findings question the use of ethanol as a solvent when investigating the contractile actions of PG molecules on smooth muscles.Supported by NIH grant No. HL-18015 and NIMH grant No. MH-26236. A comparative study of the intracellular Ca++ movements in white and red muscle

Riassunto Dopo iniezione di⁴⁵Ca⁺⁺ in conigli, radioattività appare nei mitocondri e nel reticolo sarcoplasmico del massetere dell'adduttore magno. La quantità di radioattività, e l'attività specifica del Ca⁺⁺, sono molto più elevate nei mitocondri che nel reticolo sarcoplasmico del massetere. Sono dello stesso ordine di grandezza nei due organelli dell'adduttore magno. I risultati suggeriscono un ruolo prominente dei mitocondri nei movimenti intracellulari del Ca⁺⁺ nel muscolo rosso, ed un analogo prominente ruolo del reticolo sarcoplasmico nel muscolo bianco.

Calcium and the contractile response to prostaglandin in the smooth muscle of guinea-pig stomach

Summary In the longitudinal smooth muscle of guinea-pig stomach, verapamil (10^–5 M) which showed marked suppression of high K-induced contractures, did not suppress the contractile response to PGE₁ (1.5×10^–9 to 10^–6 M) markedly. These results suggest that the contractile mechanism of PGE₁ in guinea-pig stomach may mainly depend on a release of bound Ca in the cell and partly depend on a Ca influx from the extracellular origin.

Riassunto La prostaglandina E₁ (PGE₁) ha un duplice effetto sulla sintesi epatica del colesterolo: a basse concentrazioni determina un incremento della incorporazione di acetato in colesterolo, mentre a concentrazini superiori a 50 nmoli essa determina una riduzione di questo paramentro.

Ca2+ uptake and distribution in alloxan-diabetic rat arterial and venous smooth muscle

Summary This report demonstrates that in experimental diabetes mellitus (DM) calcium uptake and its distribution is altered in rat aortic but not in portal venous smooth muscle. Results are interpreted as consequences of increased calcium binding by aortic smooth muscle in experimental DM, which could account for the hyporeactivity of alloxan diabetic rat aorta reported previously.Supported by NIH grant No.HL-18015 and NIDA grant No. DA-02339.

Response of briefly glycerinated smooth muscle to Ca2+ and Mg2+

Summary Smooth muscle, treated with 50% glycerol solution at 27°C for 20 min, contracted on the application of Ca²⁺ or Mg²⁺. The briefly glycerinated smooth muscle can be used as a model system of smooth muscle contraction.
