Zusammenfassung Die Bilirubinmengen in den Geweben ikterischerGunn-Ratten werden zusammen mit den in den subzellulären Anterilen vorliegenden Bilirubinmengen ikterischerGunn-Rattenlebern bestimmt. Die Bilirubinbestimmung erfolgte nach i.v. Infusion von Bilirubin in die Rattenleber: es findet sich vorwiegend im Leberzellensaft der ikterischenGunn-Ratte, verteilt sich jedoch nach Bilirubininfusion gleichmässig über alle Leberanteile. Die teils bekannten, teils bestätigten Unterschiede können durch Plasma-Eiweiss-Bindung des Bilirubins erklärt werden.

Zusammenfassung Ratten wurden während 10 Wochen mit durch Pantothensäure angereicherte Nahrung oder mit Pantothensäure-Mangeldiät gefüttert. Im Anschluss daran wurden die durch Differentialzentrifugation gewonnenen Fraktionen von Leberhomogenat auf ihren Protein- und Panthothensäuregehalt untersucht. Die Resultate deuten darauf hin, dass Panthothensäure die Synthese von Leberproteinen, insbesondere in der Überstands-Fraktion, stimuliert.

Summary Rat liver contains a particulate (membrane-bound) glycosyl transferase, concentrated in Golgi apparatus fractions, which catalyzes the synthesis of a trisialoganglioside from the ganglioside precursor disialohematoside (GD3). Sialic acid was not incorporated into exogenous GD1a from CMP-N-acetylneuraminic acid suggesting that GD1a is an endproduct of the monosialoganglioside pathway. Thus, the disialoganglioside pathway may be a primary source of trisialoganglioside and higher ganglioside homologs in adult rat liver. 相似文献

Summary 1-Nitropyrene, a mutagenic compound found in diesel exhaust and photocopy toners, is reduced anaerobically by rat liver fractions to 1-aminopyrene. This reaction is stimulated in the microsomal fraction by NADPH and in the cytosol by FMN. Riassunto É stata studiata la distribuzione dello89Sr++ nelle frazioni subcellulari di fegato di ratto. Nei primi secondi dopo l'iniezione, la maggior parte dello89Sr++ si trova nei mitocondri. In alcuni minuti, la radioattività diminuisce nei mitocondri ed aumenta nel reticolo endoplasmico e nella fase solubile. Lo89Sr++ viene probabilmente trasferito da una struttura cellulare all'altra; i mitocondri ed il reticolo endoplasmico svolgono ruoli distinti nella deposizione dello Sr++ nella cellula epatica.

The activity of polyamine oxidase (PAO) in rat tissues, and its subcellular distribution, were assayed using a simple polarographic method. The highest PAO activity was measured in the liver and the lowest in the skeletal muscle. In liver, kidney and uterus the highest specific PAO activity was found in the light mitochondrial fraction. PAO was not found in the microsomal fraction except in the kidney. 相似文献

A significant decrease in transglutaminase (TGase) activity was observed in the cytosol and nuclear fractions of carbon tetrachloride-damaged rat liver. The degree of decrease in TGase activity in the cytosol fraction was closely related to the serum transaminase level. Gel filtration studies revealed that TGase activity in 80 kDa fractions significantly decreased, but that in 160 kDa fractions slightly increased after carbon tetrachloride treatment. 相似文献

Summary 3 molecular forms (P1, P2 and P3) of acid phosphatase (E.C.3.1.3.2) have been detected in chicken liver homogenate. The different intracellular localization of these molecules has been demonstrated by cellular fractionation and electrophoretic analysis. P1 and P2 phosphatases are both present in the particulate fraction. P3 is present in a pure form in the soluble fraction. The difference between the enzyme molecules present in the particulate fraction and that in the soluble one is confirmed by the different activation-inhibition effect of various ions and substances on the enzymatic activity of subcellular fractions. 相似文献

We determined characteristics of rat liver mitochondrial fractions, resolved at 1000 (M1), 3000 (M3), and 10,000 g (M10) after 2 and 10 days cold exposure. In all groups, the M1 fraction exhibited the highest oxidative capacity, oxidative damage, H2O2 production rate, and susceptibility to stress conditions, and the lowest antioxidant levels. Cold exposure increased cytochrome oxidase activity in all fractions and succinate-supported O2 consumption in the M1 and M10 fractions during state 3 and state 4 respiration, respectively. With succinate, the H2O2 release rate increased in all fractions during state 4 and state 3 respiration, whereas with pyruvate/malate, it increased only during state 4 respiration. Increases in tissue mitochondrial proteins caused a faster H2O2 flow from the mitochondrial to cytosolic compartment, which was limited by the reduction in the M1 fraction. Despite increased liposoluble antioxidant levels, cold also caused enhanced oxidative damage and susceptibility to oxidative challenge and Ca2+-induced swelling in all fractions. These changes leading to elimination of H2O2-overproducing mitochondria avoided excessive tissue damage. We propose that triiodothyronine, whose levels increase in the cold environment, brings about the biochemical changes producing oxidative damage and those limiting its extent.Received 16 July 2004; received after revision 27 September 2004; accepted 18 October 2004 相似文献

Tissue-type transglutaminase (TGase) was purified from rat liver, and the effects of nucleotides on its activity were examined. The enzyme activity is inhibited by ATP in a concentration-dependent way, with complete inhibition by 3 mM ATP. Partially-purified TGase from human brain was inhibited by ATP in a manner similar to that observed with the rat liver enzyme. This suggests that the inhibition is a common phenomenon for tissue-type TGase in all species and tissues. The inhibition is reversible since full activity is restored by lowering the ATP concentration. CTP has a TGase-inhibitory potency equivalent to that of ATP, whereas GTP and UTP possess about 50% of the inhibitory activity of ATP. ADP inhibits TGase activity to the same extent as ATP, but AMP causes much less inhibition, and there is no inhibition by adenosine or adenine. The inhibition by ATP is insensitive to ionic strength and is non-competitive with the substrate putrescine. Since ATP levels in cells are of mM order, these results suggest that TGase activity is controlled by ATP in vivo. 相似文献

Mg2+-dependent and HCO-3-stimulated ATPase activity was highest in the brush border (microvilli) of rat duodenal mucosa compared with that of the other gastrointestinal mucosa. This ATPase may be useful to neutralize the gastric acid in the duodenal lumen. Carbonic anhydrase seems to accomplish a subsidiary role in the above reaction. 相似文献

The distribution of two lysosomal markers (beta-acetylglucosaminidase and acid phosphatase) between liver fractions was studied in the newborn Rat. The results indicate that after birth the hydrolase-bearing particles increased in size and had a lower density than the primary lysosomes.
