Summary Changes of extracellular concentration of brain potassium [K+]e were studied in lightly anesthetized unrestrained rats with ion-selective K+-microelectrodes introduced into the cerebral cortex with a head-mounted microdrive system. Nociceptive stimuli elicited EEG arousal lasting for 47 sec on the average which was accompanied by an increase of [K+]e from 3.0 mM to 3.31 ± 0.04 mM.Acknowledgment. Release of endogenous somatostatin (SRIF) from the rat cerebral cortical slices incubated in Krebs-bicarbonate buffer was increased from the basal rate of 3.4 +/- 0.6% of the total SRIF content in 15 min at [K+]o = 5.6 mM, to 13.1 +/- 1.6% upon raising the [K+]o to 56.6 mM. The high-K+ evoked SRIF release was absent when Ca++ in the medium was replaced by Mn++. The isolated synaptosomes from rat cerebral cortex contain 13.2 +/- 3.1 ng SRIF/mg protein compared to 0.33 +/- 0.01 ng/mg protein in the cortical tissue as a whole, suggesting that nerve terminals are the main source of the peptide released upon membrane depolarization.  相似文献

Effects of extracellular magnesium ions ([Mg2+]o ) on intracellular free Mg2+ ([Mg2+]i ) and its subcellular distribution in single fission yeast cells, Schizosaccharomyces pombe, were studied with digital-imaging microscopy and an Mg2+ fluorescent probe (mag-fura-2). Using 0.44 mM [Mg2+]o , [Mg2+]i in yeast cells was 0.91±0.08 mM. Elevation of [Mg2+]o to 1.97 mM induced rapid (within 5 min) increments in [Mg2+]i (2.18±0.11 mM). Lowering [Mg2+]o to 0.06 mM, however, exerted no significant effects on [Mg2+]i (0.93±0.14 mM), at least for periods of up to 30 min. Irrespective of the [Mg2+]o used, the subcellular distribution of [Mg2+]i remained hetero geneous, i.e. where the sub-plasma membrane region >cytoplasm >nucleus. [Mg2+] in all three subcellular compartments increased significantly, two- to threefold, concomitant with [Mg2+]i when placed in 1.97 mM [Mg2+]o . We conclude that [Mg2+]i in fission yeast is maintained at a physiologic level when [Mg2+]o is low, but intracellular free Mg2+ rapidly rises when [Mg2+]o is elevated. Like most eukaryotic cells, yeast may have a Mg2+ transport system(s) which functions to maintain gradients of Mg2+ from the outside to inside the cell and among its subcellular compartments. Riassunto Si è studiata l'attività fosfoproteinfosfatasica in omogenati totali di uova ed embrioni diParacentrotus lividus e diArbacia lixula. La reazione ha un optimum a pH acido, è stimolata da K+ e Na+ ed è inibita da Ca++ e Mg++. L'attività enzimatica aumenta tra la fecondazione e lo stadio a 64 blastomeri; in seguito rimane costante fino a blastula con mesenchima; quindi generalmente decresce.

Summary The activity of melezitase and maltase was optimal at pH 6.2 and temperature 35–40°C and inhibited by the end-products. Summary The influence of electrolyte composition and glucose concentration of a cryprotective medium on the survival of auricle fragments from adult rat hearts after storage at –196°C was investigated. Using a K+-, Mg++-, Ca++-rich solution with increased glucose concentration, a high rate of surviving fragments was found after cryopreservation.  相似文献

Summary In isolated mitochondria of heart muscle from rabbits and oxen there is, under suitable conditions, an accumulation of Ca++, which is significantly enhanced by elevating the K+/Na+ quotient of the incubation medium. K-strophanthine (10–5–10–7) does not influence the accumulation of Ca++ by the mitochondria of heart muscle. Therefore the intracellular increase in exchangeable Ca++ observed after digitalis-glycosides could be explained by a decrease of the intracellular K+/Na+ quotient, which is caused by inhibition of the membrane ATPase and diminishes the capacity for Ca++ accumulation in mitochondria.  相似文献

Summary Respiratory ionic content (Na+, K+, Ca++) of hepatic tissue and ionic excretion measurements were performed in the TenchTinea tinea kept in diluted sea water. After bilateral section of parasympathetic system, the changes of Na/K value are less than in fresh water. The survival and hepatic O2 uptake were the same. It is suggested than after both vagi section sea water Ca++ allowed a best osmoregulation in this species.

Résumé L'augmentation de la concentration en potassium (K+) à 25 meq/l et au delà, du liquide dans lequel des hypophyses de rat sont incubées in vitro stimule la sécrétion de l'hormone thyréotrope (TSH). La présence de l'ion calcium (Ca++) est nécessaire à cet effet de K+; l'incubation dans un milieu sans Ca++ empèche l'effet (sécrétion de TSH) de K+, qui est rétabli quand on réintroduit Ca++ dans le liquide d'incubation. La stimulation de la sécrétion de TSH due à K+ est inhibée par la thyroxine. Les effets de K+ sur la sécrétion de TSH sont identiques à ceux de TRF (TSH-releasing factor) hypothalamique, en ce qui concerne la présence de Ca++ et l'inhibition par la thyroxine.

Summary Intracellular potentials in the cells from 17.5-day old rat visceral yolk sacs were measured by a glass microelectrode. When penetrated from the maternal side, the cells have potentials of about 50.2±1.9 mV (inside negative) which were reduced by increasing the external K+ concentration and increased by removing Na+ ions from the bathing fluid. Triaminopyrimidine (TAP) which inhibited Na+ transport caused a dose-dependent depolarization of the cell membrane. The depolarization was dependent on the presence of extracellular Ca2+ ions. It is proposed that TAP may inhibit Na+ transport by increasing the intracellular concentration of calcium ions.This work was supported by the University of Hong Kong (grant number 335. 034.5105).Acknowledgment. The mechanisms of HCO 3 and Cl transport across basolateral membranes from rat ileum were investigated in isolated vesicles by means of uptake experiments. Neither Cl/HCO 3 exchanger nor Na+–(HCO 3 )n cotransport seem to be present in ileal basolateral membranes. Moreover Cl uptake is unaffected bycis Na+ and/or K+ gradients, indicating the absence of Na+–Cl, K+–Cl and Na+–K+–2Cl symport activity. An electrically conductive pathway seems to be responsible for both HCO 3 and Cl fluxes. Evidence is also given for the presence of a Na+/H+ exchanger at the basolateral pole of ileal enterocytes.  相似文献

Summary Based on polarographic viz. spectrophotometric determinations of the stabilities of several Cu++- and Co+++ hydroxyquinolin complexes it has been shown that a correlation exists between the quotient of complex-stabilitiesK Co +++ /K Cu ++ and antitubercular activity (cf. table and graph). This supports the hypothesis that the 8-hydroxyquinolin derivatives and analogues act by competitive antagonism; the Cu++-hydroxy-quinolates are able to replace Co in a prosthetic group of Mycobacterium-enzyme with Cu.

Summary The cytoplasma fraction of the bovine choroid plexus epithelial cells was found to contain a considerable ATPase activity. The influence of Na+, K+, Li+, Rb+, Cs+, Co++, Mn++, Zn++ and Fe+++ on the activity of the Mg++-dependent enzyme has been studied. The monovalent cations do not influence the enzymic activity, whereas the effect of the bi- and trivalent cations is characterized by an inhibition of the ATPase.  相似文献

Summary Release of Ca++, Mg++ and K+ by the carboxylic ionophore X-14547 A was studied in the mitochondrial membrane. A comparison was made with A.23187 (Calcimycin) and X.537 A (Lasalocid A) under the same experimental conditions. Summary Thiamine deficiency caused a marked decrease of intestinal alkaline phosphatase (al-Pase) activity, but had no effect on the Ca++-ATPase activity and Ca++-absorption in rats. The al-Pase activity was significantly decreased 1 h after oral administration of ethanol at 0.5 and 2.5 g/kg. In contrast, Mg++-, Ca++- and (Na++K+)-ATPase activities did not change after the administration of ethanol. These findings show that the al-Pase activity, unlike the Ca++-ATPase activity, is not related to Ca++-absorption. A possible role of al-Pase activity in the active transport of thiamine in the intestine was discussed.  相似文献
