自噬检测质粒pLVX-mRFP-EGFP-LC3的构建和表达鉴定

细胞自噬的机体重要的一种代谢过程,为了更好地研究不同条件下细胞自噬的水平,构建自噬检测质粒pLVX-mRFP-EGFP-LC3。使用Trizol法提取Hela细胞总RNA,PCR技术特异性扩增LC3基因,将目的片段插入pEGFP-N3载体中。再将mRFP片段插入到EGFP-LC3质粒中,利用PCR技术扩增带有不同酶切位点的mRFP-EGFP-LC3片段,插入到慢病毒载体pLVX-puro中。通过Fugene~?6将质粒瞬时转染到293T细胞中,使用荧光显微镜检测荧光蛋白的表达,使用Western blotting检测目的基因的表达。结果显示成功构建自噬检测质粒pLVX-mRFP-EGFP-LC3,荧光显微镜下可见红色和绿色荧光,Western blotting方法检测到LC3的表达。     

人PSF真核表达载体构建及其在CHO细胞中的表达

目的是构建人PSF基因真核表达载体pEGFP-N1-PSF,并在检测其在CHO细胞株中的表达情况。应用DNA重组技术和PCR方法从人宫颈癌Hela细胞中扩增PSF基因,插入pEGFP-N1真核表达载体,构建重组质粒pEGFP-N1-PSF并测序鉴定。将pEGFP-N1-PSF瞬时转染CHO细胞,通过Western blot和RT-PCR方法检测PSF的表达,荧光显微镜下观察绿色荧光蛋白表达。结果 CHO细胞转染pEGFP-N1-PSF真核表达载体后,RT-PCR和Western blot实验发现,在RNA和蛋白水平有PSF的表达,在荧光显微镜下可以观察到融合蛋白EGFP/PSF的表达。成功构建pEGFP-N1-PSF真核表达载体,证实其在CHO细胞中可以表达。     

柔嫩艾美耳球虫Serpin基因在293T细胞中的表达

为了检测柔嫩艾美耳球虫(Eimeria tenella,Et)丝氨酸蛋白酶抑制剂(Serine protease inhibitor,Serpin)基因(EtSerpin)在真核细胞293T中的表达情况,以Et孢子化卵囊cDNA为模板,经PCR扩增含完整Serpin开放阅读框的序列,将其克隆至pGEM-Teasy载体,构建pGEM-Teasy-EtSerpin质粒,双酶切回收目的片段后与相应酶切的真核表达载体pCAGGS连接,构建真核重组表达质粒pCAGGS-EtSerpin.该重组质粒经酶切和测序鉴定正确后转染293T细胞,用间接免疫荧光法和Western blot鉴定EtSerpin基因的表达情况,间接免疫荧光实验可以检测到红色荧光,Western blot结果显示出大小约45.5kDa的目的蛋白条带,结果表明构建的EtSerpin可以在293T细胞中获得表达.     

透明质酸合成酶2真核表达载体的构建及表达

构建了透明质酸合成酶2真核表达载体,并观察其在真核细胞中的表达。利用RT-PCR法从MG63细胞总RNA中扩增,获得人透明质酸合成酶2cDNA,并克隆至真核表达载体pEGFP-N3,将经双酶切鉴定和DNA测序证实的阳性重组子命名为pEGFP-N3-HAS2。脂质体法转染HEK293细胞,用倒置荧光显微镜检测、分析其在HEK293细胞表达及定位。双酶切鉴定和DNA测序证实成功构建pEGFP-N3-HAS2真核表达载体。转染HEK293细胞后可见HAS2-EGFP融合蛋白高表达并定位于浆膜。该载体的成功构建为进一步探讨透明质酸合成酶2在治疗骨性关节炎、修复关节软骨缺损中的应用价值奠定了基础。     

白藜芦醇对镉诱导HEK 293T细胞自噬与凋亡的影响

为了探究白藜芦醇与镉诱导人胚胎肾细胞293(HEK 293T)细胞自噬与凋亡之间的关系,本研究在已证明低浓度镉诱导细胞自噬,高浓度镉导致细胞凋亡的基础上,以HEK 293T细胞为研究对象,并分别与镉、白藜芦醇及各种抑制剂进行联合培养,利用免疫印记分析检测自噬标志蛋白LC3B-Ⅱ/Ⅰ、溶酶体标志蛋白cathepsin L...     

稳定表达EGFP-LC3蛋白的HeLa细胞系的构建

自噬是指细胞自身将细胞质蛋白或细胞器经溶酶体降解的一种过程.微管相关蛋白1轻链3(LC3)是自噬的标志蛋白,利用荧光显微镜观察EGFP-LC3荧光蛋白是否聚集成点状是检测自噬的方法之一.首先构建了表达EGFP-LC3蛋白的真核表达质粒,其具有荧光和药物筛选双标签.转染到HeLa细胞中,利用药物初步筛选后,再稀释到96孔板中筛选出有荧光的单细胞克隆,得到稳定表达EGFP-LC3蛋白的HeLa单克隆细胞系.该细胞系在饥饿诱导条件下EGFP-LC3蛋白可聚集成点状,验证了该细胞系能够用于指示自噬,为今后自噬研究奠定了基础.

人DLL1真核表达质粒的构建及过表达DLL1抑制人口腔鳞癌细胞SCC15的增殖

目的:构建人全长DLL1基因真核表达质粒及过表达DLL1对人口腔鳞癌细胞增殖的影响.方法:PCR扩增人全长DLL1基因,酶切和测序鉴定后克隆至真核表达载体pCMV-Tag4并构建真核重组质粒DLL1/pC MVTag4,瞬时转染HEK293T细胞并通过实时荧光定量PCR(Q-PCR)和Western blot鉴定DLL1的表达;进一步检测DLL1在SCC15等3株人口腔鳞癌细胞中的表达;DLL1/pCMV-Tag4瞬时转染SCC15细胞,Q-PCR和Western blot检测过表达及MTT检测DLL1过表达对细胞SCC15增殖的影响.结果:真核重组质粒DLL1/pCMV-Tag4在HEK293T细胞中成功表达;DLL1在口腔鳞癌细胞中的表达高于正常口腔细胞;DLL1/pCMV-Tag4在SCC15细胞中获得表达且过表达DLL1抑制SCC15细胞的增殖.结论:人全长DLL1分子在HEK293T及SCC15细胞中获得表达,过表达DLL1抑制口腔鳞癌细胞SCC15的增殖.     

构建重组质粒pEGFP-N1-ND CyclinB1及在胃癌细胞SGC-7901中的表达

目的:构建真核表达载体pEGFP-N1-ND CyclinB1,并观察其在人胃癌细胞SGC-7901中的表达及对SGC-7901活性的影响.方法:通过RT-PCR法扩增不可降解CyclinB1(ND CyclinB1),将其插入到pEGFP-N1载体的XhoⅠ和BamHⅠ酶切位点,构建真核表达载体pEGFP-N1-ND CyclinB1,并以PCR、双酶切、测序鉴定.通过脂质体法转染胃癌细胞SGC-7901,进行荧光检测、Western blotting分析和流式细胞仪分析.结果:PCR、酶切及测序证明重组质粒pEGFP-N1-ND CyclinB1构建成功.转染胃癌细胞SGC-7901 24 h后,荧光显微镜下观察到绿色荧光.Western blotting检测到CyclinB1的表达明显增加.流式细胞仪检测重组质粒细胞组凋亡指数高于对照组,差异有显著性.结论:成功构建pEGFP-N1-ND CyclinB1荧光真核表达载体,并可在SGC-7901细胞中有效表达.     

PEI介导pCEP4/EGFP瞬时转染293-F细胞条件优化

为构建真核表达载体pCEP4/EGFP,PEI介导瞬时转染293-F细胞,观察转染效率,优化转染条件.通过NotI和Hind III酶切位点将增强型绿色荧光蛋白基因(EGFP)插入真核表达载体p CEP4中,并转化Trans5α感受态细胞,氨苄青霉素筛选培养,挑取阳性克隆扩增后提取质粒,酶切鉴定正确后,PEI介导转染293-F细胞,流式细胞仪检测转染效率.实验结果表明,成功构建了真核表达载体pCEP4/EGFP,当N/P(PEI/DNA)为25,悬浮培养模式下转染293-F细胞,48 h转染效率为77.57%,重组载体p CEP4瞬时转染293-F细胞,可获得理想转染效果.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
