非综合征母系遗传耳聋的分子遗传学研究

对一个母系遗传的“线粒体耳聋”家系,应用ＰＣＲ,ＰＣＲ－ＳＳＣＰ,ＰＣＲ－ＲＦＬＰ,ＰＣＲ产物克隆测序技术对该家系ｍｔＤＮＡ进行了分析,发现该家系全部患者都有ｍｔＤＮＡ１２ＳｒＲＮＡ基因１５５５位点Ａ１５５５Ｇ突变,另外,除此突变位点外,我们还在部分患者中发现了一新的突变位点：ｍｔＤＮＡ１２ＳｒＲＮＡ基因１４３８位点Ａ→Ｇ改变,说明Ａ１５５５Ｇ突变可能是导致该家系耳聋的主要因素之一．     

白暨豚

世界上有众多的大江大河，分布于其中的淡水鲸类动物仅有4种，它们是南美洲亚马逊河和奥利诺科河水系的亚河豚（Inia geoffrensis）、印度河的印河豚（Platanista minor）、恒河水系中的恒河豚（P．gangetica），以及中国长江中下游的白暨豚（Lipotes vexillifer）。     

日本绒螯线粒体DNA序列研究Ⅱ.1.16rRNA

参考果蝇、卤虫与锯缘青蟹的线粒体ＤＮＡ １６ＳｒＲＮＡ基因序列进行了日本绒螯蟹相同基因片段的引物设计、ＰＣＲ扩增及序列测定,得到５６７ｂｐ的咸基序列,其Ａ、Ｔ、Ｇ、Ｃ含量分别为１９４ｂｐ（３４．２２％）、２１６ｂｐ（３８．１０％）、９８ｂｐ（１７．２８％）５９ｂｐ（１０．４１％）,与果蝇、卤虫及锯缘青蟹类的１６ＳｒＲＮＡ基因片序列含量相似。     

线粒体DNA在龟鳖目中的进化速率与古生物分化事件

基于龟鳖目的化石资料与线粒体ＤＮＡ序列数据的综合分析，估测了龟鳖类线粒体ＤＮＡ的进化速率，并采用相对速率ｓｗｃｗｉｆ （ｒｅｌａｔｉｖｅｒａｔｅｔｅｓｔ）分析了研究基因（１２ＳｒＲＮＡ和细色素ｂ）的进化速率的稳定性；结果显示，１２ＳｒＲＮＡ基因的进化速率在不同谱系中近乎恒定，而细胞色素ｂ基因的进化速率测未能通过相对速率检验。根据１２ＳｒＲＮＡ基因的颠换进化速率，分别用内推法和外推法推测曲颈龟亚目和     

电镜原位杂交对淡水三角头涡虫中期染色体RNA的研究

本文用ＲＮａｓｅ－ｇｏｌｄ标记法定位了淡水三角头涡虫中期染色体内ＲＮＡ，发现ＲＮＡ在染色单体切面内部及边缘均有分布。此外，以玉米１８ｓｒＲＮＡ的ｃＤＮＡ和水稻５ｓｒＲＮＡ的ＤＮＡ为探针，经电镜原位杂交后观察了涡虫１８ｓｒＲＮＡ（也包括４５ｓｐｒｅ－ｒＲＮＡ）及５ｓｒＲＮＡ在中期染色单体切面上的分布特点，证明涡虫染色单体边缘及内部ＲＮＡ的两个来源是核仁核糖体ＲＮＡ和核基质内的５ｓｒＲＮＡ。     

河田鸡线粒体DNA物理图谱

运用差速离心法、蛋白酶Ｋ及ＲＮａｓｅ消化法制备并纯化河田鸡（ＧａｌｕｓｇａｌｕｓｄｏｍｅｓｔｉｃｕｓＨｅｔｉａｎｂｒｅｅｄ）的线粒体ＤＮＡ（ｍｔＤＮＡ）．用１１种限制酶对其进行单酶切分析,结果表明,ＡｖａＩ、ＢａｍＨＩ、ＢｇｌＩ、ＤｒａＩ、ＥｃｏＲＩ、ＨｐａＩ、ＰｖｕⅡ、ＳａｃＩ、ＳａｌＩ和ＳｔｕＩ在河田鸡ｍｔＤＮＡ上分别有５、２、４、５、１、３、４、３、３和＞９个酶切位点,Ｓｃａｌ在不同个体河田鸡ｍｔＤＮＡ上检测出两种限制性格局,分别有２和３个酶切位点．此外,还用ＢａｍＨＩ、ＢｇｌＩ、ＤｒａＩ、ＥｃｏＲＩ、ＨｐａＩ、ＰｖｕⅡ、ＳａｃＩ、ＳａｌＩ和ＳｃａＩ进行双酶切,构建了ｍｔＤＮＡ的物理图谱     

肠炎沙门氏菌2种rDNA 16s—23s基因间隔区序列分析

采用凝胶分离ＰＣＲ产物的方法，对肠炎沙门氏菌中种ｒＤＮＡ１６ｓ－２３ｓ基因间隔区进行克隆和序列分析。肠为沙门氏菌小ｒＤＮＡ１６ｓ－２３ｓ基因间隔区，全长３５４个碱基对，包含１个谷氨酸ｔＲＮＡ基因。大ｒＤＮＡ１６ｓ－２３ｓ基因间隔区，全长５１８个碱基对，其中包含异亮氨酸和丙氨酸２个ｔＲＮＡ基因。它们分别与大肠杆菌的２个ｒＲＮＡ操纵子ｒｒｎＥ和ｒｒｎＨ有较高的序列相似性。肠炎沙门氏菌大ｒＤＮＡ１６ｓ－     

C1 SnoRNA，水稻中一种新的核仁小分子RNA

通过计算机分析国际分子生物学数据库和ｃＤＮＡ序列分析等方法，在水稻（Ｏｒｙｚａｓａｔｉｖａ）中发现了一种新的核仁小分子ＲＮＡ：Ｃ１ＳｎｏＲＮＡ．该ＲＮＡ具有ｂｏｘＣ，ｂｏｘＤ和末端碱基配对区等典型的核仁小分子ＲＮＡ结构特征；并有１４个核苷酸与水稻１８ｓｒＲＮＡ中一段保守序列互补．推测Ｃ１ＳｎｏＲＮＡ可能在１８ｓｒＲＮＡ甲基化过程中起重要作用．编码Ｃ１ＳｎｏＲＮＡ的基因位于水稻Ｈｓｐ７０基因的第一个内含子中．这是在水稻中发现的第一个由基因内含子编码的ＳｎｏＲＮＡ．

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
