生物素-抗生物素系统斑点免疫结合试验检测棘球蚴病患者的免疫应答

建立了生物素-抗生物素系统斑点免疫结合试验(BAS-DIBA),并以此法研究了棘球蚴病患者对细粒棘球蚴(Echinococcus granulosus)抗原的免疫应答.79例棘球蚴病患者血清、69例非棘球蚴病患者血清和119例健康人血清的BAS-DIBA试验结果为:两试验检测抗棘球蚴抗原抗体的敏感性分别为97.45%和92.41%;特异性分别为98.40%和98.94%;在91.14%的棘球蚴病患者血清中,BAS-DIBA试验较DIBA法敏感;两法同为阳性的棘球蚴病患者血清中,BAS-DIBA检出的抗体效价的几何均数为DIBA法的5.48倍;4例DIBA试验阴性的棘球蚴病患者血清在BAS-DIBA法中为阳性.证实所建立的BAS-DIBA法是检测棘球蚴病患者免疫应答敏感性高、特异性强的方法,特别是对于低抗体水平的棘球蚴病患者可能具有更高的应用价值.     

棘球蚴抗原纯化对免疫球蛋白IgG免疫应答的影响

以纯化的棘球蚴抗原和未经纯化的粗抗原用于棘球蚴感染者血清中抗棘球蚴抗原的特异性免疫球蛋白IgG水平的检测试验,结果表明,当血清稀释度为1∶200时,使用纯化抗原可以获得较高的特异性 (98.9%),并可保持在使用粗抗原及血清稀释度为1∶400时获得的较高的敏感性（93.7%）,提示在棘球蚴感染者血清中抗棘球蚴抗原特异性免疫球蛋白IgG水平的检测中,使用纯化棘球蚴抗原和粗抗原均可获得较高的敏感性(93.7%)和较高的特异性 (98.4%,98.9%),使用纯化抗原时的特异性略高,但在统计学上并无显著差异.     

细粒棘球蚴重组蛋白14-3-3和Eg10免疫差异的初步研究

表达纯化细粒棘球蚴重组蛋白14-3-3(rEg14-3-3)和Eg10(rEg10),分别免疫小鼠,8w后进行细粒棘球蚴原头蚴攻击感染.计算免疫保护力,检测脾组织中CD4+T、CD8+T细胞亚群以及T细胞增殖情况.实验结果表明,rEg14-3-3免疫组小鼠具有85.3%抵抗细粒棘球绦虫感染的能力,而rEg10免疫组小鼠与对照组小鼠相比无免疫保护力;与PBS对照组相比,只有rEg 14-3-3免疫组的CD4+T细胞亚群的分布有明显差异,且rEg 14-3-3抗原能诱导淋巴细胞显著增殖.因此rEgl4-3-3蛋白能抑制细粒棘球蚴的生长,诱导小鼠产生高水平的免疫保护力,可作为疫苗候选分子.     

果洛州人群棘球蚴病患病现况调查

目的：掌握果洛藏族自治州人群棘球蚴病的患病状况,为制定该地区本病控制策略提供科学依据。方法：应用B超扫描、血清免疫学检测结合患者的症状和体征综合确定人群棘球蚴病的患病状况。结果：果洛州人群棘球蚴病患病的患病率为9.45%,其中女性显著高于男性,分别为11.89%和7.23%,差异有统计学意义;每个年龄段均可见有棘球蚴病的发生,并随着年龄的增长,患病率逐渐升高,60岁以上组达到最高。不同职业的人群中,牧民和僧人的患病率最高,分别为15.36%和11.70%,差异有统计学意义。结论：果洛藏族自治州棘球蚴病流行严重、疫情活跃,加大该病在本地区的防治力度具有战略性意义。     

MMP-2、TIMP-2在小鼠原发性肝泡球蚴病中的表达及意义

为初步研究基质金属蛋白酶-2(MMP-2)及其抑制剂(TIMP-2)在小鼠原发性肝泡球蚴病血清中的表达及意义。将60只12周龄健康雌性昆明小鼠分为空白组(20只)和肝泡状棘球蚴组(40只)。建模100d后分别提取2组小鼠血清,用酶联免疫吸附试验(ELISA)检测2组小鼠血清中MMP-2和TIMP-2的浓度。结果显示:肝泡状棘球蚴组小鼠血清MMP-2、TIMP-2浓度均高于空白组(P<0.05),且MMP-2与TIMP的比例明显高于空白组。由此可知,小鼠血清MMP-2浓度的升高及MMP-2与TIMP-2比例的失衡可能和肝脏泡球蚴组织的侵润生长存在相关性。     

生物素链霉亲和素联合免疫层析用于快速检测NT-proBNP

为提高常规免疫层析技术的检测灵敏度,引入生物素-链霉亲和素系统,以荧光微球作为标记物,建立了一种新型快速定量检测NT-pro BNP的方法.试验结果表明:新型试剂盒在0.02~30μg·L~(-1)范围能对NT-proBNP实现快速检测.在全血检测中,新型试纸条所测NT-pro BNP浓度与雷度快速免疫分析仪定量检测结果具有很好的一致性.新型免疫层析试纸条大大提高了快速检测的准确性,可广泛用于床旁检验(POCTs)等临床诊断阶段.     

斑点免疫结合法检测3种植物病毒

研究了改进的斑点免疫结合法在检测芜菁花叶病毒、烟草花叶病毒和小西葫芦黄化花叶病毒中的应用.无论是使用全抗血清还是纯化的免疫球蛋白,检测感染组织粗汁液中的病毒或纯化的病毒,均获得了满意的结果.同时进行的斑点免疫结合试验、酶联免疫吸附试验和免疫吸附电子显微术等的比较结果表明:无论是检测纯化病毒还是感染组织粗汁液中的病毒,斑点免疫结合法的检测敏感性均优于后2种方法,病毒的可测感度芜菁花叶病毒为0.65ng,烟草花叶病毒为0.39 ng,小西葫芦黄化花叶病毒为0.45 ng;应用碱性磷酸酶标记抗体及羟基吲哚磷酸盐和氮兰四唑为底物较为合适,这种底物可在室温下长期保存,并且反应产物为不褪色的紫色,易于观察和保存.     

免疫磁珠的制备及其在捕获乳腺癌患者白细胞中的应用

应用生物素-亲和素系统制备免疫磁珠,考察了不同条件对制备免疫磁珠的影响.结果表明:当反应温度为4℃、亲和素浓度为0.2mg/mL、PBS pH=5.8、反应时间为2h时,磁性微球的亲和素载量为25~55mg/g;BNHS与抗体质量比1∶7时,生物素与抗体的结合率最高;亲和素与生物素化抗体摩尔比在1∶3时,亲和素与生物素的偶联率最高.抗CD45免疫磁性微球的白细胞去除率高达78%,磁珠回收率达98%,可用于进一步捕获乳腺癌外周血循环肿瘤细胞.     

氯霉素残留胶体金免疫层析快速检测法的研制

建立一种快速、简易的检测样品中氯霉素残留含量的胶体金免疫层析法.将抗氯霉素单克隆抗体-胶体金复合物包被在胶体金结合垫上,并将人工合成的氯霉素抗原包被在硝酸-醋酸纤维素薄膜表面作为检测线(T线),其与待测样品中氯霉素竞争结合胶体金标记的氯霉素单克隆抗体,并能以颜色直观显示检测的结果.用GICA与ELISA试剂盒对比检测了...     

AZD2014对体外细粒棘球绦虫原头蚴活性影响的研究

目的探讨双TOR抑制剂AZD2014在体外对细粒棘球绦虫原头蚴活性的影响。方法细粒棘球绦虫原头蚴分成6组进行体外培养,其中四组加入AZD2014使其浓度分别为200、100、50、25μmol/L;继续培养10 d,期间利用伊-红染色方法在倒置显微镜下观察原头蚴的活力,实验重复3次后,绘制原头蚴活力曲线;Western-blot检测100μmol/L的AZD2014培养原头蚴4 d后的p-Akt1、p-4EBP1、p-S6K1的表达量;Caspase-3试剂盒检测100μmol/L的AZD2014培养原头蚴4 d后的Caspase-3酶活性。结果 100μmol/L的AZD2014作用4 d后,原头蚴活性开始下降,存活率为(75.93%±7.28%);体外培养10 d后,100μmol/L组原头蚴活力仅为(24.03%±1.01%),200μmol/L组原头蚴活力为(2.40%±1.14%);100μmol/L的AZD2014培养4 d后,原头蚴蛋白因子p-Akt1、p-4EBP1、p-S6K1表达量明显下降;在100μmol/L的AZD2014培养原头蚴4 d后,其Caspase-3酶活性下降。结论 AZD2014在体外具有抗细粒棘球蚴原头蚴的作用,理论上,可做为临床上抗包虫的新型药物。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. What＇s more, the linguistic phenomena in a speaking community can be understood more thoroughly.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
