Summary A new method is described for isolating hepatocytes by an enzymatic procedure: 0.05% bacterial collagenase in calcium-free Hanks' solution is injected into the liver tissue which is removed aseptically. Summary A routine method for the preparation of pigment granules from insect eyes is reported. When viewed using an electron microscope, the isolated granules appear to be free from different structural elements, and there is very little contamination by membranous material. Summary An improved and short method is described for the isolation of intact eosinophilic leukocytes from horse blood with high yield (1–1.5 g/20 1). Viability and purity of the preparations were verified by light and electron microscopy and by the trypan blue exclusion test. Summary The behavior of Protozoa can be studied by using the ethogram as a conceptual tool capable of giving an almost complete picture of the motor biology of these microorganisms. A new semi-automatic, computerized method for drawing ethograms is described here: it allows a time-saving of about 70%, in comparison with the similar manual procedure. Microorganism movements are photographed by a Pentax LX camera from the screen of a TV monitor, connected to the stereomicroscope by a TV camera, and resolved into single images using a stroboscopic apparatus. The pictorial data are introduced into the computer by means of a digitizing tablet, and the track analysis is performed semi-automatically. The measurements recorded are then processed using a commercial statistics package in order to obtain a general view of the quantitative parameters of each ethogram. 相似文献

Summary A relatively short and simple method for the isolation of the eosinophils from normal human blood is reported. With a recovery of about 45–55%, cell preparations, showing a degree of purity of 90–98%, are obtained. The antileprosy drug dapsone is unable to penetrate intact Mycobacterium leprae in vitro, as determined by its effect on o-diphenoloxidase in the bacilli. When combined with the peptide polylysine, the sulfone drug passes through the bacterial cell membranes, and penetrates the enzyme protein, resulting in a 100% inhibition of its activity. 相似文献
