Neuropeptide Y inhibits corticosterone secretion by isolated rat adrenocortical cells

Summary Studies with isolated rat adrenocortical cells have shown that neuropeptide Y (NPY) inhibits both basal and ACTH-stimulated corticosterone secretion. These results suggest the regulatory role of NPY in corticosterone secretion from the adrenal gland, especially during stress.Supported in part by a grant CPBP 06.03.3.16.

Preparation of isolated cells from rat heart

Variations of testosterone uptake by gonadotropic cells of the adenohypophysis of the rat

Testosterone (T) could be detected by immunocytochemistry after cryoultramicrotomy and T is likely bound on a binding site of a high affinity. Castration and adrenalectomy involve a swift disappearance of T in the gonadotropic cells. Synthesis of new binding sites was induced by daily T injection in the castrated and surrenalectomized Rat from 90 days. This synthesis is inhibited by cycloheximide. These data suggest that binding sites are proteins. The captation of T by the gonadotropic cells was diminished similarly at the diminution of T, afterwards, T captation was stabilized at a weak level which became stable for several weeks. It seems that immunocytochemistry permits the study of the variation of T captation by Rat pituitary gonadotropic cells.

Effect of glucagon on ethanol oxidation in isolated rat liver cells

Summary Addition of glucagon 5 min after ethanol was found to stimulate the rate of ethanol oxidation in hepatocytes isolated from starved rats. This stimulation is of the same order of magnitude as that mediated by asparagine. The glucagon effect is suppressed by antiproteolytic agents such as insulin or NH₄Cl. Comparison of the mode of action of oxytocin and lysine-vasopressin on the isolated rat uterus

Zusammenfassung Ein der Rezeptorentheorie zugrunde liegender Vergleich der Dosis-Wirkungs-Beziehung zwischen Oxytocin und Lysine-vasopressin an der isolierten Rattengebärmutter hat gezeigt, dass während der Evolution die Oktapeptide der Neurohypophyse sowohl ihre Affinität für den Rezeptor als auch ihre maximale Aktivität bei Absättigung des Rezeptors, «intrinsic activity», verändert haben. Weiterhin wurde gefunden, dass Magnesium-Ionen in einer Konzentration von 0.5 mM nicht nur, wie bisher angenommen, die Affinität der Hormone, sondern auch ihre maximale Aktivität bei Absättigung des Rezeptors beeinflussen.     

The nature of DNA synthesis by isolated nuclei from cells of a rat tumour

Summary The DNA synthetic activity of nuclei isolated from a solid rat tumour was determined. The nuclei had DNA synthetic properties similar to nuclei from other sources but the time course of the reactions was different.     

In vivo effect of inhibin on FSH uptake by rat testis

Summary Simultaneous administration of inhibin and¹²⁵I-hFSH significantly decreased the uptake of¹²⁵I-hFSH by the immature rat testis. The data suggest that inhibin may modulate FSH action at the gonadal level.     

Effect of gentamycin on insulin release and45Ca net uptake by isolated islets

Summary In isolated islets, gentamycin reduced both the⁴⁵Ca net uptake and insulin release induced by glucose, but failed to inhibit the insulin secretion provoked by the combination of Ba²⁺ and theophylline. This indicates that the inhibitory effect of gentamycin is due to a reduction of Ca²⁺ entry into B-cells instead of to a harmful effect upon the integrity of the effector system, responsible for the extrusion of the insulin containing granules.Supported by a Grant (No. 79/1872) from the São Paulo State Research Foundation (FAPESP), Brazil.     

The action of the peptidoleukotriene LTD4 on intracellular calcium in rat mesangial cells

The dose-dependent effect of CGP 45715A on the LTD₄-induced Ca²⁺ response of glomerular mesangial cells has been studied. Our results demonstrate that the LTD₄-dependent increase in the cytosolic Ca²⁺ concentration primarily involves an InsP₃-mediated release of Ca²⁺ from intracellular storage sites and to a minor extent an enhanced influx of Ca²⁺ through receptor-operated Ca²⁺ channels located in the plasma membrane. The action of CGP 45715A on the Ca²⁺ response is an inhibitory one and is convincingly explained by a displacement of LTD₄ from its receptor site(s). The contractile effect of LTD₄ on pulmonary smooth muscle is proposed to be mainly caused by a receptor-mediated hydrolysis of phosphatidylinositol-4,5-bisphosphate.     

Effect of acetone on VLDL secretion by the isolated rat liver

Summary High acetone levels occur in uncontrolled diabetes and after isopropanol administration to rats. In both conditions, very low density lipoproteins (VLDL) secretion is depressed. Acetone, however, failed to affect the VLDL secretion rate by the isolated perfused rat liver, suggesting that this metabolite is not involved in impaired VLDL production in diabetes and after isopropanol administration.
