A new selenium source, Nano red elemental selenium (Nano-Se) was used to study the effect on the GPx activity of broiler chick
kidney cells (BCKC)in vitro, Sodium selenite (Na2SeO3) and seleno-1-methionine (Se-Met) were used as the controls. The results showed that the effects of three kinds of Se forms
on the GPx activity of BCKC were accordant (p>0.05) compared with each other at 0.01. 0.05 and 0.10 μmol/L Se concentrations treatments. In the range of 0.00–0.10 μmol/L
Se concentrations, the GPx activity increased with elevation of Se concentrations in medium. For the three kinds of Se forms,
the GPx activity reached the climax at 0.10 μmol/L Se concentration. At 0.20 and 0.30 μmol/L Se concentrations, the influnces
of three kinds of Se forms were not accordant with one another. For Nano-Se, the GPx activity at 0.20 and 0.30 μmoi/L Se concentrations
remained the same as that at 0.10 μmol/L Se concentration treatment. For Se-Met, the GPx activity at 0.20 μmol/L Se concentration
treatment remained the same with 0.10 μmol/L treatment; the GPx activity at 0.30 μmol/L Se concentration treatment was declined
significantly (p<0.05) compared with 0.10 or 0.20 μmol/L treatment. For Na2SeO3, the GPx activity falled gradually with Se concentration increasing from 0.10 μmol/L to 0.30 μmol/L, and at 0.30 μmol/L Se
concentration treatment, the GPx activity was less than the original of BCKC. The results implicated, on the GPx activity
of BCKCin vitro, the ranking of width range of the most suitable Se concentration for nutrition curve of the three Se formes is Nano-Se>Se-Met>Na2SeO3.

The effect of iron deficiency on heterocyst differentiation and some physiological properties of the filamentous cyanobacteriumAnabaena sp. PCC 7120 was investigated. Under moderate iron limitation conditions, achieved by addition of iron chelator 2, 2′-Dipyridyl
(<80 μmol/L) led to delayed heterocyst differentiation, no heterocyst differentiation was observed under severe iron limitation
conditions, when the concentration of 2,2′-Dipyridyl in the medium was more than 100 μmol/L. it seemed that there are certain
iron-regulated genes or operons whose function is to control heterocyst development. In addition, iron deficiency impaired
the growth. Low iron cells had a decrease in the quantities of pigment content (chlorophyll and phycocyanin content), the
whole cellin vivo absorbance spectra confirmed the decrease, the protein electrophoretic profiles revealed that iron-deficient cells had less
protein bands, with the increase of 2,2′-Dipyridyl, the protein bands was more and more less. And differently, iron deficiency
also caused an increase of ROS (Reactive Oxygen Species) and SOD activity, it suggests that iron deficiency led to oxidative
stress, which generally occured under high-iron conditions.

Effect of quercetin on HeLa cell system of cervical cancer was studied by methods of MTT and Annexin V-FITC/PI. The results show that quercetin has functions of inhibiting breeding of HeLa cells and inducing apoptosis of the cells. The total apoptosis rate is positively proportional to reaction duration and concentration of quercetin used. The maximum apoptosis rate being (88.76±2.35)% was obtained when the concentration was 50.0 μmol/L and the cells were treated with quercetin for 72 hours. Based on establishing a model of tumor of cervical cancer transplanted into nude mice, quercetin of different concentrations was injected into abdominal cavity of nude mice and situation of tumor growth was reviewed. The result showed that with quercetin concent'ration increasing from 0 to 100.0 μmol/L, the transplantation volume and weight of the tumors decreased from (279.59±70.58) mm^3 and (0.145±0.019) g to (128.72±36.12) mm^3 and (0.089± 0.019) g respectively, while apoptosis rate of the transplanted tumor increased from (9.63±1.85)% to (34,98±0.47)%, which proved that quercetin inhibited increment of volume and weight of transplanted tumor in nude mice bodies. 相似文献

The reaction of 5-(4-hydroxyphenyl)-10,15,20-triphenyl porphyrin with 2,6-dibromomethylpyridine and 4,4‘-dicarboxyl-2,2‘-bipyridine respectively gave 2,6-bis-[5,10,15-triphenyl-20-( 4-phenoxymethyl )-porphyrin-yl]-pyridine(3) and 4, 4‘-di-[5,10,15-triphenyl-20-( p-phenoxycarbonyl)-porphyrin-yl]-2,2‘-bipyridine (4). 5-[4-(4‘- Bromobutoxy) phenyl]-10, 15, 20-triphenyl porphyrin reacted with 2,6-dihydroxymethyl pyridine to give 2,6-bis-[5,10,15-triphenyl- 20-(4-(p-phenoxy)-butoxymethyl)-porphyrin-yl]-pyridine(5). Cellular immune response is a major barrier to xenotransplantation, and cell adhesion is the first step in intercellular recognition.
Flow-cytometric adhesion assay has been used to investigate the differential adhesions of monocyte (Mo), natural killer cell
(NK) and T lymphocyte (T) present within human peripheral blood mononuclear cells (PBMC) to porcine aortic endothelial cells
(PAEC), and to demonstrate the effect of human interferon-γ (hIFN-γ) or/and tumor necrosis factor-α (hTNF-α) pretreatment
of PAEC on their adhesiveness for different PBMC subsets. The preferential sequence for PBMC subset binding to resting PAEC
is Mo, NK and T cells, among which T cells show the slightest adherence; hTNF-α can act across the species, and augment Mo,
NK and T cell adhesion ratios by 40%, 110% and 3 times, respectively. These results confirm at the cell level that host Mo
and NK cells are major participants in the cellular xenograft rejection, thereby, providing a prerequisite for further studying
the human Mo/NK-PAEC interactive mechanisms. 相似文献

To reveal the relationship between mitogen-activated protein kinase (MAPK) and tau phosphorylation, we used different concentration of PD98059, an inhibitor of MEK (MAPK kinase), to treat mice neuroblastma (N2a) cell line for 6 h. It showed that the activity of MAPK decreased in a dose-dependent manner. But Western blot and immunofluorescence revealed that just when the cells were treated with 16μmol/L PD98059, tau was hyperphosphorylated at Ser396/404 and Ser199/202 sites. We obtained the conclusion that overinhibited MAPK induced tau hyperphosphorylation at Ser396/404 and Ser199/202 sites. 相似文献

An α-amylase encoding gene was amplified by polymerase chain reaction fromSaccharomycopsis fibuligera and inserted into a shuttle vector YEp352, together with the yeast phosphoglycerate kinase 1 promoter and α-factor signal
gene. The recombinant expression plasmid pLA8α was transformed into an industrial strain ofSaccharomyces cerevisiae Sc-11. The activity of the α-amylase produced by the transformant Sc-11-pLA8α was 6.3 U/mL and the starch utilization rate
in YPS medium was 42%. The purified amylase was analyzed by SDS-PAGE, showing a molecular weight of 55×103 protein band. Furthermore, the residual sugar, ethanol and some volatile compounds in the fermented worts under simulating
brewing conditions were determined by chromatographic analyses. The fermentation characteristics of Sc-11-pLA8α were similar
to that of Sc-11 and only minor changes in the concentration of flavor compounds could be observed.

Expression and cellular localization of orphan receptor TR2 mRNA in relation to germ cell apoptosis in cryptorchid testes
of rat and rhesus monkey have been studied by usingin situ hybridization andin situ 3′-end labeling of DNA fragments (TUNEL). The results show that: (i) TR2 mRNA is specifically expressed in the germ cells,
mainly in the spermatocytes, round and elongated spermatids. The expression level of TR2 mRNA varies with the seminiferous
cycle, (ii) In the rat cryptorchid testes on days 3 and 5 after the surgery, the germ cells began to undergo apoptosis with
no evident decrease in TR2 mRNA level. On day 7.5, however, most germ cells underwent apoptosis, while the expression level
of TR2 mRNA declined markedly, and TR2 mRNA was rarely expressed on day 10 thereafter, (iii) On days 15 and 20 of the cryptorchid
testes of rhesus monkey, TR2 mRNA was only expressed in a few of primary spermatocytes and the mRNA was almost undetectable
on days 30, 45, 60. These results suggest that TR2 mRNA probably plays an important role in spermatogenesis and germ cell

Twenty Yongzheng copper coins were fast dat-ing by X-ray energy dispersive spectrometer with scanning electron microscope (SEM-EDS) according to official ingredient. The 20 coins were classified in two groups according to the results. Group 1 represents the coins minted before AD 1 728 and group 2 represents the coins minted after AD 1 728. At concentration range (0.00%-5.13%), the distribution of lead is not homogeneous in the coins, and the largest diameter of lead inclusion is less than 15 μm. An expanded granular sludge bed (EGSB) reactor was adopted to study the influence factors and rule of enhancing granular sludge concentration and performance. The experiment was performed at 33 ℃, pH 6.0-8.0 with continuous flow by adding proper quantity of nutritional trace elements. The results show that SLR was the key of steady operation of EGSB reactor. The increment of the granular sludge was influenced by volume loading rate (VLR), liquid up-flow velocity and sludge loading rate (SLR). Concentration of granular sludge increased rapidly when liquid up-flow velocity was over 0.94 m · h^-1 with SLR being at 1.0-2.0 d ^-1. With the propriety parameters: liquid up-flow velocity 2.52 m · h^-1, SLR 1.0-2.2 d^-1 and VLR 8.2-13.1 kg · m ^3 · d^-1, 23 days' continuous operation resulted in an increment by over 80% of granular sludge concentration in the EGSB reactor, plus good granular sludge property. 相似文献

Novel adsorbent, Fe(III)-loaded ligand exchange cotton cellulose adsorbent [Fe(III)LECCA], was used to investigate the adsorption
performances and mechanism of fluoride removal from aqueous solutions. The adsorbent was found to adsorb fluoride rapidly
and effectively. The fluoride removal was influenced by pH. Adsorption model followed first-order reaction at different temperature,
theapparent adsorption activated energyE, was 6.37 kJ·mol−1, and adsorption enthalpy ΔH was 5.35 kJ·mol−1. The adsorption capacity of fluoride on adsorbent was 3.2 mmol·g−1 (dry weight). The maximal integer coordination ratio of fluoride with Fe(III) LECCA was 3∶1. The ligand exchange mechanism
of adsorption was elucidated through chemical methods and IR spectral analysis.

The slightly polluted wastewater from oil refinery contains some COD, oil pollutants and suspended solids (SS). A small-scale fixed film biological aerated filter (BAF) process was used to treat the wastewater. The influences of hydraulic retention time (HRT), air/water volume flow ratio and backwashing cycle on treatment efficiencies were investigated. The wastewater was treated by the BAF process under optimal conditions: the HRT of 1.0 h, the air/water volume flow ratio of about 5 : 1 and the backwashing cycle of every 4-7 days. The results showed that the average removal efficiency of COD, oil pollutants and SS was 84.5%, 94.0% and 83.4%, respectively. And the average effluent concentration of COD, oil pollutants and SS was 12.5, 0.27, 14.5 mg·L^-1, respectively. The experimental results demonstrated that the BAF process is a suitable and highly efficient method to treat the wastewater. 相似文献

The characterization of the algaeAnabaena cylindrica solution with Fe (III) was investigated using fluorescence emission and synchronous-scan spectroscopy. The ranges of concentrations
of algae and Fe (III) in aqueous solutions were 5.0×108 2.5×108 cell/L and 1060 μmol/L. respectively. The effective characterization method used was synchronous-scan fluorescence spectroscopy
(SFS). The wavelength difference (Δλ) of 90 nm, was maintained between excitation wavelength (λen) and emission wavelength
(λen). The peak was observed at about (λex) (λem) 326 nm for synchronous scan fluorescence spectroscopy. The fluorescence
quenching in system of algae Fe(III)-HA was studied using synchronous-scan spectroscopy for the first time, Fe(III) was clearly
the effective quencher. The relationship betweenI I (quenching efficiency) andc (concentration of Fe (III) added) was, a linear correlation for the algae solution with Fe(III). Also, Aldrich humie acid
(HA) was found to be an effective quencher.
