用水稻着丝粒重复序列RCS1为探针,与3072个克隆进行菌落杂交,得到了32个阳性克隆,用RCS1与拟斯卑尔脱山羊草着丝粒重复序列Tcs250为探针进一步筛选,在32个RCS1相关的阳性克隆中任选10个克隆进行点杂交,分别有6个和5个阳性克隆.为了克隆RCS1相关片段,依据RCS1的序列设计了三对引物,将引物3从上述阳性克隆中扩增的一个543bp的片段克隆测序,发现与水稻RCS1部分片段达到约83%的同源,与大麦的反转座子(Ty3/gypsy)部分序列同源性达到了92%,与节节麦中着丝粒的整合酶基因部分序列同源性达到了96%,命名为TBRCS1.TBRCS1可能是野生一粒小麦着丝粒区的组成部分.  相似文献

二棱和四棱皮大麦农艺性状对产量的通径分析与比较研究   总被引:1,自引:0,他引:1  
对二棱带皮型和四棱带皮型大麦的7个农艺性状与单株产量分别进行了通径分析和比较研究,结果表明:在二棱大麦中, 各性状对单株产量的直接正效应从大到小的顺序依次为:单株穗数、千粒重、芒长和穗长等,而在四棱大麦中,则为:单株穗数、穗粒数、穗粒重和千粒等,可见两种类型中,单株穗数对单株产量的贡献均居首位,因此,两种类型的大麦要提高单株产量均应以增加单株有效穗数为主,二棱皮大麦要兼顾高千粒重、长芒和长穗为配合选择指标,四棱皮大麦则应有两种选择模式;饲用大麦追求高产,应注重“穗重型”,而啤酒大麦应以大粒高产为主要育种目标。  相似文献

以侵染苜蓿的苜蓿花叶病毒中国分离株(Alfalfa mosaic virus Chinese isolate,A1MV—Ch)RNA2为模板,利用人工合成的特异性引物,进行反转录及PCR扩增,分两段分别扩增了复制酶基因的5′端1.32kb和3′端及其非编码区的1.23kb cDNA序列.用限制性内切酶切割PCR产物后,与pUCl9重组,转化大肠杆菌DH5α,筛选重组质粒,用限制性内切酶分析及PCR鉴定,得到分别含有5′端序列和3′端序列的重组质粒。已由上述两种重组质粒构建了含有完整复制酶基因的重组质粒.进行了全序列测定,并与国外报道的A1MV-425株系的相应序列相比较,其复制酶基因编码区核苷酸序列同源性达97.8%,推测的氨基酸序列的同源性达97.6%,3′端非编码区核苷酸序列同源性为98.2%.并将全长复制酶基因与植物表达载体pROKⅡ重组,得植物转化载体pAlMV—FL.  相似文献

河套蜜瓜ACC氧化酶基因cDNA部分片段的克隆和序列分析   总被引:1,自引:1,他引:1  
以河套蜜瓜(Cucumis melo L.ev Hetao)成熟果实为材料,用硫氰酸胍法提取总RNA,以oligo(dT)15为引物,反转录合成cDNA,利用一对甜瓜ACC氧化酶(1-aminocyclopropane-1-earboxylate oxidase,ACO)基因cDNA的特异性引物进行聚合酶链式反应(PCR)扩增。获得了预期大小的cDNA片段.将此片段克隆于pUCl9质粒中,进行DNA序列分析.序列分析结果表明该片段为545bp,编码甜瓜ACO的第126至第306个氨基酸.该河套蜜瓜ACO基因cDNA序列与Andes甜瓜、Cantaloup charentais甜瓜和哈密瓜的相应序列进行比较,其核苷酸序列与Andes甜瓜的同源性为99.4%;与Cantaloup charentais甜瓜和哈密瓜的同源性均为99.6%。与其对应的氨基酸序列的同源性均为99.4%.  相似文献

从河南省西峡晚白垩世一枚恐龙蛋化石内腔絮状内含物及外壳提取DNA样品,并用一段特异引物与6碱基随机引物进行PCR扩增、基因克隆和序列分析。结果表明用这对引物从蛋腔絮状内含物样品中扩增到一段150bp的DNA片段,推测氨基酸序列与EMBL库中已知蛋白质氨基酸序列进行了序列同源性分析,结果表明所克隆到的这段基因与非洲爪蟾表皮钙粘着蛋白(cad-herin)前体及脑细胞粘着蛋白、牛及人胎盘胞钙粘着蛋白在氨基酸水平有显著的序列同源性,分别为40.6%,37.5%,34.4%和36.4%。  相似文献

 以成功克隆大蕉几丁质酶基因(MpChi-1)时设计的一对引物,采用RT-PCR技术从广亲和水稻品种(Oryza.satival L.Cpslo17)中扩增到一水稻几丁质酶的全长cDNA,长1 115 bp,命名为Oschi;此序列已在GenBank中注册,登录号为EU045451;将Oschi基因核苷酸测序结果在NCBI服务器上用BLAST搜索软件进行同源性基因检索;并将Oschi的cDNA序列及氨基酸序列与大蕉及其它单子叶植物的相应序列进行多重序列排列比较并构建系统树;结果显示此基因具有编码Ⅰ类几丁质酶的基本结构;与大蕉核苷酸序列相比同源性为99%,氨基酸序列同源性为99%,与预测登记的最相近的水稻[Oryza sativa(japonica cultivar-group)]核苷酸序列相比同源性为98%, 氨基酸序列同源性为98%;广亲和水稻Oschi基因与大蕉MpChi-1基因的序列及蛋白结构有较高的一致性。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
