Summary The amout of bound calcium and force were measured in chemically skinned rat myocardial fibers during isometric contraction at different concentrations of free Ca2+. The data obtained suggest that calcium binding by cardiac myofibrils is cooperative and probably depends on mechanical tension.The author expresses his gratitude to Dr V. I. Summary In the presence of calmodulin and phosphate and an ATP-regenerating system, Triton-treated skinned fibers of theTaenia coli could be made to contract and relax by step changes of Ca++ within about 30 sec. In the absence of phosphate, relaxation was slower, and during this slow relaxation tension was not maintained actively. The passive tension could be abolished by phosphate (3–6 mM). Phosphate had little effect on contractile tension but decreased the speed of contraction.Supported by the Deutsche Forschungsgemeinschaft. Summary ATPase activity and force generation have been measured simultaneously in isolated, demembranated muscle fibers of the Pacific blue marlin (Makaira nigricans) between 0 and 30°C. Tension generation is relatively independent of temperature above 15°C and falls with a Q10 of <1.5 on decreasing the temperature to 0°C. In contrast, the Q10 for ATPase activity is 2.2 over the range 0–30°C. The results are interpreted in terms of the cross bridge theory of contraction.  相似文献

With the aid of thymidine-H3 autoradiography gliogenesis in the rat brain was seen to start during embryonic stages, which might continue into the postnatal stages of development. Summary Calmodulin was immunocytochemically localized in the brush borders of rat intestinal epithelial cells from the tip to the base of the villi, from day 18 of fetal life up to the adult stage. The early (14th day) fetal cells, like the adult crypt cells, were not immunoreactive, although their calmodulin content was equal to that of the mature cells from the tips of the villi.  相似文献

Summary Evidence from light and electron microscopic studies indicates that differentiation of myocardial cells in moth embryos begins at 6 days post-oviposition and is completed on eclosion. Fibrillogenesis, and development of cytoplasmic organelles and membranes are described. The heart is fully differentiated and functional at eclosion.Acknowledgments. Summary The contraction induced by a Ca2+-independent myosin light chain kinase (MLCK-) was characterized in terms of isometric force (Fo), immediate elastic recoil (SE), unloaded shortening velocity (Vus), shortening under a constant load and ATPase activity of chemically skinned smooth muscle preparations. These parameters were compared to those measured in a Ca2+-induced contraction to assess the nature of cross bridge interaction in the MLCK-induced contraction. Fo developed in chicken gizzard fibers as well as SE were similar in contractions elicited by either agent. Vus in the contraction induced by MLCK-(0.36 mg/ml) was similar though averaged 39.3±8.9% less than Vus induced by Ca2+ (1.6x10–6M) in the control fibers. Addition of Ca2+ (1.6x10–6M) to a contraction induced by MLCK-resulted in small increases in both Fo and Vus. Shortening under a constant load was similar for both types of contractions. The contraction induced by MLCK-was accompanied by an increased rate of ATP hydrolysis. The MLCK-induced contraction is thus kinetically similar though not identical to a contraction induced by Ca2+. We conclude that with respect to actin-myosin interaction, MLCK- and Ca2+-induced contractions are similar.  相似文献

Summary Diacylglycerol (DG) and triacylglycerol (TG) levels in rat lung tissue were determined from day 17 of gestation to day 10 post partum and studied in parallel with ultrastructural differentiation. The DG level, although rather low at all measured stages, rose significantly between days 17 and 19 and at birth. TG level increased steadily during the whole studied period and especially between days 17 and 19 and at birth. In DG as well as in TG, saturated fatty acids were predominant. The rising of TG levels paralleled the appearance and accumulation of lipid vacuoles in mesodermal cells lying in contact with type II cells. The possible role of these cells is discussed.  相似文献

Mit Beginn des Sehens erhöht sich der gesamte Nukleinsäuregehalt des Organismus; vorübergehend zeigt sich auch ein prozentualer Anstieg. Summary The effects of various concentrations of ionized Ca were examined on the activity of rat brain tryptophan hydroxylase previously treated with EGTA. Within the range of ionized Ca-concentrations thought, to be physiologically important (10–8 to 10–5 M), no significant activation of the enzyme occurred, although activation was observed at higher concentrations of the metal.  相似文献
