Rheumatoid arthritis (RA), which is marked by inflammatory synovitis, is a common, chronic autoimmune-disease, whose pathogenesis is complex and still unclear. In order to explore the effects of heat and hyposmotic stimuli on synoviocytes in rheumatoid arthritis, the changes of [Ca^2+]i induced by heat, hyposmotic and 4α-PDD stimuli were observed in synoviocytes. [Ca^2+]i elevation induced by heat 28℃, hyposmotic and 4α-PDD stimuli is found to be positively relative to increasing temperature, decreasing osmolality and rising concentration of 4α-PDD. Results show that there is reciprocity among these stimuli and desensitization, and that [Ca^2+]i elevation depends on Ca^2+ influx, but not necessarily links to Ca^2+ release from intracellular stores and voltage-dependent Ca^2+ channel in synoviocytes. The above characteristics of Ca^2+ influx are similar to those of TRPV4. A probable mechanism has been suggested that heat and hyposmotic stimulation might increase the level of [Ca^2+]i by activating the TRPV4-like channel and Ca^2+ influx in the synoviocytes.  相似文献

Actin is a protein abundant in cells. Recently, it has been proved to be universally existent in the nuclei of many cell types. Actin and actin-binding proteins, as well as aetin-related proteins, are necessary for the mediation of the conformation and function of nuclear actin, including the transformation of actin between unpolymerized and polymerized, chromatin remodeling, regulation of gene expression and RNA processing as well as RNA transportation. Fibroblast growth factor-2 (FGF-2) is a member of a large family of proteins that bind heparin and heparan sulfate and modulate the function of a wide range of cell types. It has been proved that FGF-2 stimulates the growth and development of new blood vessels (angiogenesis) that contribute to the pathogenesis of several diseases (i.e. cancer, atherosclerosis). However, many of the biological activities of FGF-2 have been found to depend on its receptor抯 intrinsic tyrosine kinase activity and second messengers such as the mitogen activated protein kinases. This review will focus on the mechanism of FGF-2/FGFR induced signaling pathway in tumor and human breast cancer.  相似文献

Both O3 and ClO2 have a high effect on inactivating-algae in source water with no forming THMs which do harm to human in producing drinking water, so they will be favorably substituted for Cl2. In order to make certain of the mechanism of inactivating algae with O3 and ClO2, the algal cell number change and its different characteristics of figures and structures in treated and untreated water have been studied by the microscopy and SEM and the mode of inactivating algae has been inferred. The results show that the mechanism of inactivating algae by O3 is not completely identical with that by ClO2. The actual reaction process and efficiency have been controlled by many factors, such as the different characteristics of oxidants and algal cells.  相似文献

An erythroid-specific nuclear matrix protein (termed ε-NMPk) in K562 cells, which can specifically bind to the positive stage-specific regulatory element (ε-PRE Ⅱ , - 446- - 419 bp) upstream of the human ε-globin gene, has been identified by using gel mobility shift assay. Meanwhile, Southwestern blotting assay showed that the nuclear matrix protein ε-NMPk in K562, cells may be composed of two polypeptides ( ~ 40 ku). In addition, it is observed in the gel mobility shift assay that the nuclear matrix proteins from K562, HEL and Raji cells can bind to the silencer DNA ( - 392- - 177 bp) in the 5'-flanking sequence of human ε-globin gene respectively. However, the shift band K detected in K562 cells is different from shift band H/R in HEL and Raji cells, suggesting that a common nuclear matrix protein may exist in HEL and Raji cells. Results show that the nuclear matrix protein may play an important role in the regulation of the human ε-globin gene expression.  相似文献

An irmovative mobile robot that has reconfigurable loeomotion chassis and reconfigurable bionic wheels has been developed to meet the needs of different payload and different terrain. Several prototypes have been achieved by the recortfiguration. By modeling relative comparison coefficients, these prototypes are analyzed in terms of geometrical parameter of trafficability, static stability and maneuverability. The effects of reconfiguration on these indices of robot performance can be compared, i.e. the variable height of chassis h has the biggest effect, the variable length of chassis 1 is the second, then is the camber angle β and the caster angle α. The optimized geometries of the complexes between HnY (n=2, 3; Y=O, S, N) and LiNH2 have been calculated at the B3LYP/6-311++G^** and MP2/6-311++G^** levels. Three stable complexes were obtained. Frequency analysis showed that the enlarged 2N-4Li presents the abnormal blue shift in three complexes. The calculated binding energy with basis set super-position error (BSSE) and zero-point vibrational energy (ZPE) corrections of complex Ⅰ-Ⅲ is -58.65, -31.66 and -69.59 kJ·mol^-1 (MP2), respectively. Natural bond orbital theory (NBO) analysis has been performed, and the results revealed that the H2O…LiNH2 (complex I) and H3N…LiNH2 (complex Ⅲ) are formed with coexisting σ-s and n-s type lithium bond interactions, complex Ⅱ is formed with ττ-s type lithium bond interaction between HnY (n=2,3; Y=O, N) and LiNH2, and H2S…LiNH2 (complex Ⅱ) is formed with n-s type lithium bond interaction between H2S and LiNH2. The effects of soluble and fibrillar Aβ1-40 on membrane permeability and intracellular free Ca^2 of nerve cells were investigated by the laser confocal microscopy. Results indicate that: i) Effects of soluble and fibrillar Aβ1-40 on cell membrane permeability are both concentration-dependent. Soluble Aβ1-40 increases membrane permeability only at concentration of 3μmol/L, while the toxic effect of fibrillar Aβ1-40 is much stronger, its evident effect begins from 1μmol/L. When its concentration rose to 3μmol/L, not only the membrane permeability increased, but also the nuclear membrane broke seriously, ii) Both soluble and fibrillar Aβ1-40 at high concentrations increased the intracellular free Ca^2 , and the increased amplitudes are concentration-dependent. However, the fibrillar one induces the increase of intracellular Ca^2 much quicker and synchronously.These results indicate that some correlation exists between the neurotoxicity of high concentration soluble and fibrillar Aβ1-40 and the change of physico-chemical properties and intracellular Ca ion imbalance.  相似文献

Since the man-machine interfaces (MMI) of a main control room provide the control platform of a nuclear power plant (NPP),the development of the design quality of MMIs plays a very important role in the operation of a NPP. With the development of digital technology, the development of the advanced main control rooms (AMCRs) has become an inexorable trend. Therefore, the positive and the negative effects of AMCRs on human factors engineering need to be evaluated. For this p~, a simulation system has been studied and developed to quantitatively evaluate a MMI design from the viewpoint of human factors. The simulation system takes advantage of computer simulation technology to simulate an operating process of an interaction between operators and a MMI design under an instruction of an operation procedure of the AMCR of a NPP. Meanwhile, the necessary data are recorded for evaluation. It integrates two editors and one simulator. In the paper, the simulation system is presented in detail. In this study, we report the experimental reali-zation of seven-qubit Deutsch-Jozsa (D-J) algorithm and controlled phase-shift gates with improved precision using liquid state nuclear magnetic resonance (NMR). Theexperimental results have shown that transformations Uf in the seven-qubit D-J algorithm have been implemented with different pulse sequences, and whether f is constant orbalanced is determined by using only a single function call(Uf). Furthermore, we propose an experimental method tomeasure and correct the error in the controlled phase-shift gate that is simple and feasible in experiments, and can have precise phase shifts. These may offer the possibility ofsurmounting the difficulties of low signal-to-noise ratio(SNR) in multi-qubit NMR quantum computers, morecomplicated experimental techniques, and the increase ofgate errors due to using a large number of imperfect selec-tive pulses. These are also applied to more complicated quantum algorithms with more qubits, such as quantumFourier transformation and Shor抯 algorithm.  相似文献
