基于多面体寡聚硅倍半氧烷-甲基丙烯酸羟乙酯poly(POSS-MA-co-HEMA)有机硅胶杂化整体柱,建立了典型苷类药物(连翘苷、豆腐果苷、紫丁香苷、天麻素)的亲水作用毛细管电色谱检测技术。考察了苷类药物的亲水作用,优化了流动相乙腈比例、缓冲液浓度、缓冲溶液pH、分离电压以及反压阀压力的影响,在最优条件(流动相为2 mmol/L磷酸三乙胺缓冲液(乙腈含量90%,pH=7.0),分离电压-8 kV、柱压力750 psi),4种苷类药物在10 min内得到了基线分离。连翘苷、豆腐果苷、紫丁香苷和天麻素线性范围分别为10~200 μg/mL、25~200 μg/mL、15~200 μg/mL和25~200 μg/mL, 检测限为1.5~8.0 μg/mL。应用于血清模拟样品分析,平均回收率为95.6%~99.1%,相对标准偏差小于3.3%。  相似文献

基于N-氨乙基-3-氨丙基甲基二甲氧基硅烷(AEPTS)键合开管柱,研究遗忘性贝类毒素(软骨藻酸,DA)的电色谱分离行为,建立了DA的毛细管开管柱电色谱(OTCEC)快速分析技术.考察并优化了缓冲液类型、缓冲液浓度、p H值、分离电压以及进样时间等参数影响,在最优条件下(20 mmol·L-1Na H2PO4-Na2HPO4缓冲液,p H=4.00,电动进样25 s,进样和分离电压为-18 k V),DA标准溶液在0.12~5.00μg·m L-1范围内线性良好,检测限(S/N=3)为0.03μg·m L-1.应用于扇贝中DA的快速分析,扇贝贝肉提取液无需净化即可直接进样,扇贝中DA线性浓度范围为0.50~10.00μg·m L-1,检测限(S/N=3)为0.25μg·m L-1(相当于贝肉中DA含量1.0μg·g-1),扇贝样品加标回收率为80.5%~83.1%,RSD8.5%.  相似文献

目的:建立HPLC同时测定清心明目上清片中四种成分(栀子苷、连翘苷、黄芩苷和盐酸小檗碱)含量的方法.方法:色谱柱.AglientTC-C18(4.6mm×250mm,5μm),流动相为乙腈(A)-0.2%磷酸溶液(B),梯度洗脱程序依次为.0~10 min 18%B,10~12 min 20%~30%B,12~14 min30%~16%B,14~20 min 16%~28%B,20~30 min 28%~65%B,30~40 min 65%~80%B,流速0.9 mL·min-1,柱温为25℃,检测波长分别为238 nm(栀子苷和连翘苷)、275nm(黄芩苷和盐酸小檗碱).结果:栀子苷、连翘苷、黄芩苷和盐酸小檗碱分别在0.031 1~0.622 2μg(r=0.999 6),0.533 3~8.000μg(r=0.999 6),0.080 0~1.600 0μg(r=0.999 5),0.115 6~1.155 6μg(r=0.999 4)范围内线性关系良好,平均加样回收率分别为98.4%、98.3%、97.1%、98.2%.结论:本法操作简便、精密度高、重现性好,适用于同时测定清心明目上清片中栀子苷、连翘苷、黄芩苷和盐酸小檗碱的含量.  相似文献

建立HPLC法同时测定感冒退热颗粒中含表告依春和连翘苷的量,以有效控制其质量.测定条件如下:Ultimate XB-C18(5μm,4.6 mm×250 mm)为色谱柱,乙腈-0.02%磷酸水溶液(23∶77)为流动相,检测波长235 nm,流速:1.0 mL/min,柱温为35℃.结果表明,表告依春在0.010 63~0.05315μg范围内呈线性关系(R=0.999 9),加样回收率为98.50%,RSD值为0.51%(n=9);连翘苷在0.099 81~0.499 05μg范围内呈线性关系(R=0.999 9),加样回收率99.93%,RSD值为0.61%(n=9).不同生产厂家的感冒退热颗粒中含连翘苷量相差不大,但含表告依春量差异较大.本实验建立的方法简便、稳定、准确可靠,可用于感冒退热颗粒的质量控制.  相似文献

该方法建立清热止带颗粒含量测定方法。本文选用了高效液相色谱法(HPLC),Agilent C18分析柱(4.6×250 mm,5μm);流动相:乙腈-0.05 mol·L-1三乙胺溶液(用磷酸调p H至3.0)(25∶75);检测波长:345 nm;流速:1.0 ml.min-1。盐酸小檗碱在0.408~1.836μg之间线性关系良好,r=0.999 5(n=6),平均回收率为97.7%,RSD=1.61%(n=6)。该方法简便、灵敏度高、重现性好,可作为该制剂的质量控制方法的参考依据。  相似文献

为建立一个稳定可靠分离度好的方法测定红景天中含红景天苷的量.采用高效液相色谱法,色谱柱为BDS HYPERSIL-C18(4.6 mm×250 mm,5μm);以乙腈-0.03%磷酸为流动相进行梯度洗脱;流速1.0 m L/min;检测波长275 nm;柱温30℃.结果表明,红景天苷的进样量在1.25~10μg范围内有良好的线性关系,平均回收率为98.16%,RSD为0.89%.该方法准确、可靠、分离度好,可用于红景天的质量评价.  相似文献

通过对中药山茱萸提取物的毛细管区带电泳(CZE)和毛细管胶束电动色谱(MECC)分离条件的优化研究,建立了获得其化学特征指纹图谱的方法.山茱萸醇提物化学特征指纹图谱的CZE优化条件是:未涂层弹性融硅毛细管为54cm(内径为50μm,有效分离长度为50cm);50mmol/L硼砂缓冲液,pH值为9 5;压力进样为34 5kPa·s(5p.s.i.·s),操作电压为22kV( )→(-),柱上200nm紫外检测,30℃柱温.优化的MECC条件:30mmol/LSDS,50mmol/L硼砂缓冲液,pH值为9.5,其他条件与CZE的相同.该方法具有高效、快速、样品用量少和分析成本低等特点.  相似文献

测定喜树果中喜树碱和10-羟基喜树碱的含量,采用高效液相色谱法,色谱柱为DiamonsilC18(5μm,200mm×4.5mm),乙腈:水(25:75)为流动相,检测波长254nm,流速1.0mL·min-1。结果:喜树碱在(0.0644~32.2)μg·mL-1、羟基喜树碱在(0.25~25)μg·mL-1范围内线性关系良好。说明方法分离效果好、专属性强、分析速度快、灵敏度高,可作为喜树果药材含量测定的方法。  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
