牛酪蛋白全长cDNA克隆pBaS1c184X GLⅢ和COrI双酶切,回收基中的酪蛋白基因编码片段,与经BamHI,SmaI双酶切的植物表达载体pBI12.12分两步连接;第一步载体的BamHI末端与酪蛋白基因的BglⅡ粘性末端连接;第二步用T4DNA多聚酶补平酪蛋白基因的EcoTRⅠ末端,再与载体SmaⅠ进行平末端连接而环化。  相似文献

鹅源腺病毒全基因组DNA酶切片段的克隆   总被引:4,自引:0,他引:4  
利用1株鹅源腺病Y81G4株,经鹅胚增殖后收获尿囊液,用差速离心法纯化病毒子并提取病毒基因组DNA,病毒DNA经Hind Ⅲ酶切后共产生10个片段,分别回收各酶切片段,与经Hind Ⅲ单酶切的pUC18连续,获得8个不同的重组质粒。对于未克隆到的两末端片段,经碱处理去除末端蛋白后,以平端和HindⅢ粘端与经SmaⅠ和 HindⅢ双酶切的pUC18连接,获得了重组质粒pGAHC和pGAHI。克隆的各酶切片段,通过酶切电泳和Southern blotting结果鉴定,证明已分别克隆到该病毒基因组DNA HindⅢ酶切片段,且各酶切片段大小之和约为32.9kb.  相似文献

为研究牦牛BVDV E0蛋白的生物信息学及抗原性, 本研究对本实验室克隆并测序的牦牛病毒性腹泻病毒E0基因进行了生物信息学分析;同时用含有酶切位点、起始密码子、终止密码子的引物重新扩增E0基因, 并连接PMD18-T载体, 经BamHⅠ和HindⅢ双酶切后, 将获得的E0片段克隆至PET-28A原核表达载体, 构建重组质粒并转入JM109.酶切、质粒PCR鉴定正确后转入rosetta(DE3)菌进行诱导表达.经SDS-PAGE检测结果显示目的基因获得了较高的表达, 表达的融合蛋白约33ku, 经western-blotting检测表明目的蛋白具良好的有抗原性.  相似文献

本文利用DNA体外重组技术,将氧化亚铁硫杆菌染色体DNA的Hind Ⅲ酶切片段插入启动子探测质粒pSDSI(Ap~r,Tc~s,5.65kb)的Hind Ⅲ位点上,获得了一批具有启动子活性的染色体DNA片段.其中抗性最强的一株可在240μg/ml的四环素平板上生长,对此抗性最强的质粒pSDRF12进行了限制性酶切图谱分析,并利用其上的PstI和EcoRI位点分别酶切,得到了两个亚克隆pSDRF121和pSDRF122.新构建的两个亚克隆在其氧化亚铁硫杆菌启动子后只有一个Hind Ⅲ位点,可通过该位点插入外源目的基因,进一步观察其表达情况。  相似文献

利用大肠杆菌Klenow大片段聚合酶5'→3'聚合酶活性和T4DNA聚合酶3'→5'外切酶特性构建植物高效表达载体pBLT35Svp4-ST.用pUC18-435S中的T35S启动子取代植物表达载体pBLG-VP4-ST中的双35S启动子,通过Klenow大片段的聚合酶活性对植物表达载体pBLG-VP4-ST中的HindⅢ3'凹端补平,再利用T4DNA聚合酶外切酶活性将质粒pUC18-435S的四倍35S启动子即T35S中的SacⅠ3'凸端削平.在T4DNA连接酶作用下将一端为平端,另一端为BamHⅠ的粘性末端的四倍35S启动子定向连接到植物表达载体PBLG-VP4-ST中取代原有的双35S启动子,转化并筛选阳性克隆.结果经PCR和酶切鉴定证实,四倍35S启动子成功连接到植物表达载体PBLG-VP4-ST中.  相似文献

牛酪蛋白全长cDNA克隆pB_aS_1C184经Bgl Ⅰ和EcoR Ⅰ双酶切,回收其中的酪蛋白基因编码片段,与经BamH Ⅰ、SmaⅠ双酶切的植物表达载体pBI121.2分两步连接:第一步载体的BamHⅠ末端与酪蛋白基因的BglⅠ粘性末端连接;第二步用T_4DNA多聚酶补平酪蛋白基因的EcoR Ⅰ末端,再与载体SmaⅠ进行平末端连接而环化,转化JM109菌株,从得到的33个转化子中,通过~(32)P标记的酪蛋白cDNA基因为探针,点杂交筛选到一个阳性克隆。经酶切鉴定,确认酪蛋白cDNA基因编码区已正确插入到pBI121.2中。  相似文献

应用PCR从杜仲内生Pseudomonas koreensis JDM-2株基因组中扩增出ACC脱氨酶基因acdS,将其克隆到pMD18-T载体并段进行DNA测序,通过BamHⅠ和HindⅢ酶切从重组质粒pMD18ACDS中切下acdS基因序列,将其连接到pQE30质粒的BamHⅠ和HindⅢ位点,构建表达质粒pQE30ACDS,转化大肠杆菌BL21,利用比色法测定ACC脱氨酶活力.测序结果表明,JDM-2菌株acdS基因大小为1 017 bp,与已报道不同菌种acdS基因的同源性在86%~99%之间.SDS-PAGE结果显示在大肠杆菌BL21中表达了分子量为38 kDa的ACC脱氨酶.酶活性检测结果显示重组菌ACC脱氨酶的酶活力为0.214 U/mg.  相似文献

从人外周血细胞中提取mRNA,用RT-PCR法扩增含BamH与HindⅢ酶切位点VLA4基因序列,运用分子生物学方法将该序列克隆到pGEM中,构建成VLA4基因序列的真核表达载体并进行鉴定分析.对重组载体用双酶切系统酶切后进行电泳并测序,结果证明目的基因克隆及连接方向正确,测序结果正确.通过实验,成功地构建了含VLA4基因的真核表达载体pGEM-VLA4.  相似文献

目的克隆BMP7基因及构建BMP7基因真核表达载体,稳定转染于H22细胞系,制备荷瘤小鼠。方法应用RT-PCR技术从人胚肾细胞系293T细胞总RNA中成功扩增出1 293 bp包含BMP7基因编码区的cDNA序列,在上下游加上BamHI及HindⅢ双酶切位点后形成带酶切位点的BMP7基因,然后连入含BaraH I及HindⅢ双酶切位点的pEGFP-C1表达质粒上,构建BMP7基因的重组真核表达质粒pEGFP-C1-BMP7。脂质体介导方法转染细胞,RT-PCR、western-blot方法检测其mRNA及蛋白表达,皮下注射制备荷瘤小鼠。结果扩增的cDNA经测序比较与基因文库完全一致;BamHI及HindⅢ双酶切后,质粒形成约4.7 kb和约1.3 kb两条带,与理论计算值完全一致。注射转染BMP7基因后H22细胞小鼠瘤体明显增大。结论成功克隆BMP7基因及构建BMP7基因的真核表达载体。BMP7基因高表达可能是细胞癌变的原因之一。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
