目的:分析增强型绿色荧光蛋白(EGFP)与蛋白激酶C-θ(PKCθ)的融合蛋白(PKCθ-EGFP)在HeLa细胞中的表达、亚细胞定位及其对细胞形态的影响。方法:用脂质转染胺(Lipofec-tAMINE 2000)为载体分别以pPKCθ-EGFP和pEGFP-N1转染HeLa细胞,流式细胞仪分析其表达率,激光共聚焦显微镜分析其亚细胞定位,同时分析二丁酸佛波醇酯(PDB)和小檗碱对其亚细胞定位和细胞形态的影响。结果:转染pEGFP-N1或pPKCθ-EGFP的HeLa细胞48 h后,表达绿色荧光的细胞百分率分别为(70.9±4.4)%和(45.8±2.3)%(P<0.05)。表达的PKCθ-EGFP融合蛋白均匀分布于细胞内,与EGFP蛋白在细胞内的分布相似。PDB刺激30 min后,表达PKCθ-EGFP的HeLa细胞的形态发生显著变化,即由原来的梭形变成圆形,荧光强度增强。但表达EGFP的HeLa细胞的形态及荧光分布均不受影响。小檗碱对PDB诱导的HeLa细胞的形态和EGFP-PKCθ融合蛋白的分布无明显影响。结论:表达PKCθ-EGFP融合蛋白的HeLa细胞在PDB作用下细胞形态发生显著变化。  相似文献

目的是构建人PSF基因真核表达载体pEGFP-N1-PSF,并在检测其在CHO细胞株中的表达情况。应用DNA重组技术和PCR方法从人宫颈癌Hela细胞中扩增PSF基因,插入pEGFP-N1真核表达载体,构建重组质粒pEGFP-N1-PSF并测序鉴定。将pEGFP-N1-PSF瞬时转染CHO细胞,通过Western blot和RT-PCR方法检测PSF的表达,荧光显微镜下观察绿色荧光蛋白表达。结果 CHO细胞转染pEGFP-N1-PSF真核表达载体后,RT-PCR和Western blot实验发现,在RNA和蛋白水平有PSF的表达,在荧光显微镜下可以观察到融合蛋白EGFP/PSF的表达。成功构建pEGFP-N1-PSF真核表达载体,证实其在CHO细胞中可以表达。  相似文献

构建GGT1重组真核表达载体,观察GGT1在COS7细胞中的定位.利用PCR技术扩增GGT1全长基因,经HindⅢ和EcoR Ⅰ酶切后连接入pEGFP-N1真核表达载体.将构建完成的重组表达质粒转染入COS7细胞,利用激光共聚焦显微镜观察GGT1在细胞中的定位.构建载体经双酶切和序列测定证实重组载体包含有正确的GGT1编码序列.激光共聚焦显微镜观察到,整个细胞内弥散绿色荧光,绿色荧光分布于COS7细胞浆中,提示GGT1定位于细胞浆中.成功构建人pEGFP-N1-GGT1真核表达载体,检测到GGT1定位于哺乳细胞的细胞浆中,为GGT1的功能研究提供了线索.  相似文献

【目的】构建pEGFP-N1-LC3真核表达载体,并将其转染进人胚肾细胞293T,通过Earle's盐平衡液(Balanced salt solution,EBSS)饥饿诱导观察细胞自噬现象。【方法】RT-PCR扩增LC3基因,并将其插入pEGFP-N1真核表达载体构建重组质粒。将重组质粒转染至人胚肾细胞293T,显微镜观察、Western blot检测GFP-LC3融合蛋白表达情况。对转染细胞进行Earle's盐平衡液饥饿诱导,通过Western blot验证自噬指标,激光共聚焦显微镜观察自噬泡形成。【结果】成功构建pEGFP-N1-LC3真核表达载体,并在293T细胞中成功表达目的蛋白,在进行Earle's盐平衡液饥饿诱导后观察到自噬泡的形成,Western blot检测到LC3-Ⅰ向LC3-Ⅱ的转化。【结论】本实验为研究自噬在癌细胞病理进程中的机制提供了实验素材。  相似文献

应用PCR技术从p EGFP-N1载体扩增获取CMV启动子和EGFP基因,利用限制性内切酶BglⅡ/NotⅠ以及NotⅠ/Eco RⅠ将其插入p T/HB,构成p T-CMV-EGFP重组转座子,p T-CMV-EGFP与SB11转座酶形成"睡美人"转座系统.采用脂质体与荧光表达载体p EGFP-N1不同配比转染小鼠成纤维细胞系3T3,p TCMV-EGFP与SB11梯度比例转染小鼠肝癌细胞系H22后,经流式细胞术比较阳性细胞率,当条件为脂质体与总DNA质量比为4∶1;转座酶与转座子间质量比为1∶4转染时,细胞阳性率最高(H22悬浮细胞可达42.23%),转染结果较稳定(SD=1.25).通过此优化体系转染小鼠骨髓来源的树突状细胞DCs,经激光共聚焦显微镜显示,能够获得被EGFP修饰的DCs,并通过流式细胞术检测被标记有EGFP的特异DCs阳性率可达20%以上.表明本研究已成功建立了一个可用于原代DCs转染的方法.  相似文献

为构建含绿色荧光蛋白(EGFP)和人胰岛素原基因的逆转录病毒表达载体,并检测其在人肝癌细胞HepG2中的表达,将IRES-EGFP片段克隆到含调控元件的人胰岛素原基因逆转录病毒表达载体(pLXSN-GI-Ins)中,构建得到表达质粒pLXSN-GI-Ins-EGFP.经脂质体介导转染HepG2细胞后,各孔分别加入含有30.0 mmol/L葡萄糖的培养液继续培养24 h,在荧光显微镜下观察EGFP基因的表达,检测细胞上清液中的胰岛素值.数据显示,成功构建逆转录病毒表达质粒pLXSN-GI-Ins-EGFP.转染HepG2细胞后48 h,表达EGFP基因的细胞数目占总细胞数目的比值为(38.0±5.0)%.结果表明,构建了含EGFP和调控元件的人胰岛素原基因逆转录病毒表达载体,并且能够在HepG2细胞中成功表达.  相似文献

为构建人LMP3-EGFP融合蛋白真核表达载体,并检测其在HEK293细胞中的表达及定位,通过基因重组的方法构建LMP3/pEGFP-N3重组真核表达载体,并通过酶切和基因测序鉴定。脂质体法转染HEK293细胞,用倒置荧光显微镜检测、分析其在HEK293细胞表达及定位。经酶切和基因测序鉴定LMP3/pEGFP-N3重组真核表达载体构建成功。转染HEK293细胞后,荧光显微镜检测显示融合蛋白仅在细胞浆中表达。说明基因重组技术可成功构建LMP3/pEGFP-N3重组体真核表达载体,重组表达的融合蛋白仅存在于细胞浆内。  相似文献

目的:为研究细胞周期蛋白在肿瘤形成过程的分子机制,构建带FLAG标签的细胞周期蛋白E的真核表达载体,并检测其在瞬时转染HeLa细胞株中的蛋白表达。方法:通过RT-PCR扩增cyclinE基因编码cDNA,并将扩增的cDNA片段插入p3XFLAG-CMVTM-14真核表达载体,重组子经酶切分析和测序鉴定后,用脂质体介导的基因瞬时转染法,将重组正确的表达载体转染HeLa细胞,用Western-blot技术检测其在HeLa细胞中融合蛋白的表达。结果:经酶切鉴定和测序分析证实人cyclin E的真核表达载体构建成功,并能在瞬时转染的HeLa细胞中表达。结论:成功构建了人cyclin E的真核表达载体,为进一步研究细胞周期蛋白的功能奠定了基础。  相似文献

目的:构建pEGFP-C1-MCH真核表达载体,并将其转染入HEK293细胞中,筛选阳性细胞克隆,为研究MCH基因在能量代谢中的功能及机制提供细胞模型.方法:提取脑组织总RNA,反转录为cDNA,参照Genbank中提供的序列设计引物扩增MCH基因全长.再将该基因全长cDNA克隆至质粒pEGFP-C1,经菌落PCR筛选及双酶切和DNA测序鉴定,成功构建了含有目的基因MCH的重组质粒pEGFP-C1-MCH.并利用脂质体2000介导其转染HEK293细胞,用荧光显微镜和RT-PCR检测EGFP和MCH在细胞中的表达.结果:克隆的pEGFP-C1-MCH质粒序列中的MCH与Gen Bank相符;细胞转染72 h后,转染成功的细胞在荧光显微镜下表达较强的绿色荧光,MCH基因稳定表达.结论:pEGFP-C1-MCH真核表达载体的构建及其在HEK293细胞中的稳定表达,为研究MCH基因在能量代谢中的功能及作用机制提供了实验模型.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
