In Ornithogalum virens, following Giemsa staining, the mitotic chromosomes revealed distinct C-banding pattern. Interphase nucleus also showed 12 chromocentres corresponding to the number of C-bands. Based on the measurements of C-bands, about 13.5% of the genome is heterochromatic.  相似文献

Summary Various alterations in Giemsa banding technique have been introduced to observe their influence on band formations in M-chromosome of Vicia faba root tip dividing cells. With the introduction of some minor alterations in the technique, revelation of a large number of classes of constitutive heterochromatin has been made possible. Apparently such Giemsa banding pattern is comparable to the ones observed in a routine way amongst mammalian chromosomes.Acknowledgments. Summary Vicia faba (broad bean) root-tip chromosomes were subjected to the SSC-Giemsa and trypsin-Giemsa regimes. Phase-lucent cross-bands seen after fixation/SSC treatment subsequently stain positively with Giemsa. Sequential analysis of the trypsin-Giemsa regime shows, in contrast, that bands are manifest through selective removal of material from interband regions.  相似文献

Summary The uptake of Ag-ions by isolated nucleoli of rat liver cells was studied. Nucleolar proteins separated by electrophoresis were examined for selective silver-staining. A mechanism for a preferential Ag-staining of the nucleoli is discussed.We thank Mrs L. Schwinn for her valuable technical assistance, and Mrs K. Jain for presenting us DNA. Zusammenfassung Das dem Centromer der menschlichen Chromosomen (ebenfalls bei andern Arten) anliegende Gebiet erscheint hell, klar und rund. Dies wurde in einigen Zellen bei fast jedem Centromer, bei andern Zellen nur selten oder überhaupt nicht beobachtet. Eine ähnliche Erscheinung trat auch in Form von Streifen auf, die sich manchmal vom Centromer aus zu anliegenden Chromosomen erstreckten oder sich ins Cytoplasma ausbreiteten. Die mögliche Deutung dieser Beobachtung wurde besprochen.
