从泡菜中分离得到一株芽孢杆菌,经16S RNA及生理生化鉴定为凝结芽孢杆菌(BCS 13002).文中研究了碳源、中和剂对菌株生产L-乳酸的影响,并在5 L发酵罐中分别以葡萄糖及玉米淀粉为碳源进行分批补料发酵和同步糖化发酵(SSF)的研究.结果表明,葡萄糖是最合适的碳源,使用木糖和玉米淀粉作为碳源均不利于L-乳酸的产生;使用NaOH作为中和剂的效果优于Ca(OH)_2和CaCO_3.在发酵罐中以葡萄糖为碳源进行分批补料发酵,得到L-乳酸含量为115.86 g/L,糖酸转化率约为82.31%,并证明MnSO_4·H_2O和MgCl_2·6H_2O在发酵中有特殊的作用.以玉米淀粉为原料同步糖化发酵,得到L-乳酸123.3 g/L,糖酸转化率约为70.03%,产品中L-乳酸的光学纯度均达到99.8%以上.  相似文献

米根霉As3.819是一株L-乳酸的高产菌株,且能够分泌淀粉酶直接利用淀粉发酵生产乳酸。文章研究了不同培养基主要成分下米根霉菌株As3.819生产L-乳酸和淀粉酶情况,并对发酵液中淀粉酶水解产物进行分析。研究结果表明,在甘薯淀粉8%、硫酸铵为0.4%时L-乳酸和淀粉酶的产量相对较高,且在培养基中加入麸皮后能增加淀粉酶的分泌,种子培养基中碳源为麦芽糖时也能起到很好的效果。  相似文献

采用兽瘟链球菌 (Streptococcuszooepidemicus)进行发酵生产透明质酸 (hyaluronicacid ,HA) .摇瓶研究表明 :葡萄糖的补加量和补加方式对HA发酵有着较大的影响 ,采用 4 0g·L- 1的初始葡萄糖浓度及维持培养过程 10g·L- 1的糖浓度 ,可使HA的产量达到 4 .0g·L- 1,分子量在 2 .0× 10 6 Da以上 .  相似文献

L-乳酸因其为生产绿色化学材料聚L-乳酸的原料而倍受重视.文中以米根霉As3.6462为菌种,玉米淀粉为原料,研究了用吸附固定米根霉发酵L等L酸.结果表明,相对聚乙烯醇、棉布,丝瓜布有更好的固定化效果.实验条件下,载体边长在8—10mm,载体用量为15mL/50mL种子培养基,玉米淀粉发酵L-乳酸产量为72—83g/L,连续发酵4批后产量基本稳定.丝瓜布固定化米根霉发酵L看L酸有良好的工业的应用前景.  相似文献

采用紫外线和Co60照射联合诱变鼠李糖乳杆菌(Lactobacillus rhamnosus)JCM1553菌株,选育得到1株L-乳酸高产突变株SCT-10-10-60.经77代传代培养证实该菌株L-乳酸发酵遗传稳定.在37℃,200rpm下,该菌株摇瓶发酵葡萄糖60h的发酵液乳酸浓度达到最大,为195.67g/L,发酵糖酸转化率达到95.33%,比出发菌株最大乳酸产量、发酵速率和糖酸转化率分别提高了16.24%、50.13%和17.81%.相同条件下该菌株发酵木薯淀粉84h的乳酸浓度达到最大值,为203.33g/L,糖酸转化率达到78.85%,比出发菌株最大乳酸产量和发酵速率均提高了29.49%,糖酸转化率则提高24.53%.该菌株发酵产物的L-乳酸含量高达96.75%,与出发菌株(96.97%)无统计学显著差异(P>0.05).该高产菌株乳酸发酵性能提高的主要原因是菌体增长速率加快.其L-乳酸发酵性能显著优于现有的生产菌株,具有较高的潜在工业价值.  相似文献

研究了"宁紫1号"、"宁选1号"甘薯在贮藏100 d期间的贮藏效果,以期得出较为适宜的贮藏参数,并分析甘薯的营养和生理变化规律.以250 mg·L-1施保克溶液浸泡处理甘薯,置于7℃贮藏,以室温贮藏的甘薯为对照,定期进行理化指标检测.结果表明:在贮藏期内,7℃贮藏结合施保克处理可有效降低甘薯的霉腐率,减少可溶性蛋白质、可溶性总糖、总黄酮的损失.此外,两种甘薯的α-淀粉酶活性在贮藏中均高于β-淀粉酶活性.7℃贮藏结合施保克处理可提高甘薯的贮藏品质,可以在甘薯实际贮藏中应用.  相似文献

运用正交试验设计理论,对高产L-乳酸突变株NAF-032的最佳摇瓶发酵条件进行了研究,确定了最佳的发酵培养基和发酵条件.最佳发酵培养基为(g/L):葡萄糖160,氯化铵2,KH2PO4 0.3,MgSO4·7H2O 0.25,ZnSO4·7H2O 0.08.最佳培养条件为:34℃,摇床转速200 r/min,250 mL三角瓶装培养液50 mL,一次性添加CaCO380 g/L用于调节pH值.在最适发酵条件下,米根霉NAF-032的摇瓶发酵产L-乳酸产量达123.3 g/L.  相似文献

运用BP神经网络对鸟苷发酵培养基组成进行建模以及鸟苷产量预测,在此基础上利用遗传算法对鸟苷发酵培养基组成进行全局性寻优,得到最佳鸟苷发酵培养基组成:葡萄糖120 g·L-1,豆饼水解液50 g·L-1,酵母粉16 g·L-1,(NH4)2SO415 g·L-1,味精10 g·L-1,CaCO320 g·L-1(分消),CaCl22 g·L-1,硫酸镁3 g·L-1,生物素0.9 mg·L-1,硫酸锰4 mg·L-1,磷酸二氢钾4 g·L-1。结果表明,BP神经网络与遗传算法耦联法能够有效地优化发酵培养基配比。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
