原发性血小板减少性紫癜患者胸腺近期输出功能状态

目的:了解原发性血小板减少性紫瘢(ITP)患者胸腺近期输出功能的状态,评价其T细胞潜能.方法:采用实时定量PCR(TaqMan)方法检测15例ITP患者外周血单个核细胞(PBMCs)中T细胞受体重排删除DNA环(TRECs)的含量,17例正常人作为对照.结果:正常人外周血中每1 000个PBMCs中TRECs拷贝数为(3.76±3.42),ITP患者TRECs水平变化较大[每1000个PBMCs中TRECs拷贝数为(2.60±2.99)],4例病人未能检测到TRECs,病人平均TRECs水平与正常人的差异没有显著性(P＞0.05),女性病人TRECs水平略高于男性(P=0.499),但无统计学意义.结论:多数ITP患者外周血初始T细胞水平和胸腺近期输出功能保持正常水平.     

SLE患者外周血γδT细胞系的建立及表型和功能的研究

目的:建立体外扩增系统性红斑狼疮(SLE)患者外周血γδT细胞的方法,并初步分析其表型和功能,探索γδT细胞在SLE发病中的作用。方法:采用单克隆抗体固相法,对15例SLE患者和8例正常人外周血γδT细胞进行体外扩增建系,以流式细胞仪检测γδT细胞表型,并用MTT法观察γδT细胞对Daud i细胞的细胞毒作用。结果:建立了SLE患者外周血γδT细胞系,其平均纯度为(58.1±11.2)%,较正常对照组(80.3±9.2)%偏低(P<0.05);其细胞表型为:Vδ1(34.4±24.5)%、Vδ2(61.9±28.6)%、Vδ3(16.1±10.6)%、Vγ9(76.4±11.8)%,其中Vδ2表达较对照组降低,而Vδ1和Vδ3表达增加(P均<0.05);其细胞毒作用在二组间无明显差别。结论:SLE患者外周血γδT细胞的Vδ、Vγ基因的取用表达存在差异,提示其在SLE的发病中可能起一定作用。     

ITP中T细胞活化信号分子c-Cbl和Cbl-b的表达变化

目的:分析免疫性血小板减少症(ITP)中T细胞活化信号通路相关分子Cbl-b和c-Cbl基因的表达情况.方法:利用SYBR Green实时荧光定量聚合酶链式反应(RT-PCR)方法检测18例ITP患者外周血单个核细胞(PBMC)中Cbl-b和c-Cbl基因的表达水平,20例健康成人作为对照.结果:ITP组Cbl-b的表达水平(中位数为0.091%)与健康成人组Cbl-b的表达水平(中位数为0.366%)相比,明显降低(P0.001);而c-Cbl基因在ITP组中的表达水平(中位数为0.372%)与健康成人组的表达水平(中位数为0.298%)相比没有统计学差异,但c-Cbl基因在ITP女性患者样本中的表达水平(中位数为0.236%)明显低于男性患者样本c-Cbl的表达水平(中位数为0.479%)(P=0.039).结论:ITP患者外周血中Cbl-b表达下调,可能是其T细胞免疫异常活化的原因之一.     

流式细胞术检测拉米夫定对慢性乙型肝炎患者T细胞亚群的影响

目的研究拉米夫定对慢性乙型肝炎（CHB）患者细胞免疫功能的影响.方法采用流式细胞术检测50例CHB患者在服用拉米夫定前后外周血T细胞亚群,同时检测45例健康体检者外周血T细胞亚群作为对照.结果CHB患者在使用拉米夫定后,CD4 T细胞百分比较使用前明显升高（P〈0.05）,CD8 T细胞百分比明显下降（P〈0.05）,CD4/CD8明显升高（P〈0.05）.结论拉米夫定可改善CHB患者的细胞免疫功能.

RACK1基因在急性冠脉综合征行PCI术患者外周血中的表达及意义

选取在我院行PCI术的急性冠脉综合征患者66例,用实时荧光定量PCR法分别测定PCI术前及术后3个月时患者外周血单个核细胞中RACK1基因的表达。40名健康者作为对照组。测定结果是:(1)急性冠脉综合征组患者外周血单个核细胞中RACK1 mRNA表达水平明显高于对照组(17.56±4.98 vs 3.11±1.24,P0.05);(2)急性冠脉综合征患者行PCI术3月后,外周血单个核细胞中RACK1 mRNA表达水平较术前明显降低(10.84±3.71 vs 17.56±4.98,P0.05),但仍高于健康者(10.84±3.71 vs 3.11±1.24,P0.05)。RACK1在急性冠脉综合征患者外周血中表达上调,其表达水平可能和病情进展相关。     

辅助性T细胞22在类风湿关节炎外周血的表达和意义

目的:探讨类风湿关节炎(RA)患者外周血中辅助性T细胞22(Th22)/白细胞介素-22(IL-22)的表达情况及其与临床指标和症状之间的相关性.方法:流式细胞术检测RA患者(50例)、骨关节炎(OA)患者(15例)、健康对照组(15例)外周血中Th22细胞所占比例,ELISA法检测血清中IL-22水平.采用t检验和Pearson相关分析法进行统计学分析.结果:RA患者外周血中Th22细胞比例[(1.98±0.71)%]高于OA组[(1.21±0.52)%](t=2.290,P=0.021)及健康对照组[(1.18±0.45)%](t=2.524,P=0.015).RA患者血清IL-22水平[(53.746±8.327)ng/L]高于OA患者[(37.918±5.687)ng/L](t=2.560,P=0.014)及健康对照组[(34.638±5.231)ng/L](t=2.768,P=0.009).RA患者外周血Th22细胞比例与血清IL-22水平、血沉(ESR)、类风湿关节炎患者病情评价(DAS28)评分呈正相关关系(r分别为0.42、0.38、0.32,P0.05).RA患者抗合并间质性肺疾病(CCP)阳性组Th22细胞比例高于抗CCP阴性组,差异有统计学意义(t=2.785,P=0.008).抗CCP阴性RA组Th22细胞比例与病程呈负相关(r=-0.59,P=0.04).RA有骨侵蚀组Th22细胞比例高于无骨侵蚀组,差异有统计学意义(t=2.542,P=0.014).RA-ILD组Th22细胞比例高于RA患者未合并间质性肺疾病(RA-NILD)组,差异有统计学意义(t=2.513,P=0.017).结论:RA患者外周血Th22细胞及IL-22表达增高,Th22细胞比例与IL-22水平及疾病活动度(ESR、DAS28评分)正相关,同时与骨侵蚀和ILD的发生有相关性,提示Th22细胞参与RA发病机制,有可能成为RA治疗的新靶点.Th22细胞在抗CCP阴性RA中的表达情况有助于RA的早期诊断.     

不同全麻方法对2型糖尿病患者T细胞亚群的影响

目的:BIS联合AAI监测下研究不同麻醉方法对2型糖尿病手术患者T细胞亚群的影响.方法:选择36例择期手术行子宫肌瘤切除术患者,随机分为两组,A组:全凭静脉全麻组;B组:静吸复合全麻组;每组18例,分别于麻醉前(T0)、麻醉后1h(T1)、术后1d(T 2)和7d(T3)抽取静脉血,用流式细胞仪测定T细胞亚群和NK细胞的数量.结果:两组T1、T2CD3+、CD4+、CD4+/CD8+、NK细胞均有所下降,与麻醉前相比,A组P0.05;术后1d下降较为显著,与麻醉前相比,A组P0.01,B组P0.05,组间比较亦有显著性差异(P0.05);术后7d各组数据与术前水平差别不大.结论:接受腹腔镜手术的2型糖尿病患者,患在麻醉后及术后的细胞免疫功能均有不同程度抑制,而全凭静脉麻醉抑制程度更为明显.     

淋巴瘤白血病患者TCR Vβ亚家族T细胞的分布及其克隆性

目的:了解B细胞淋巴瘤白血病(LL)患者TCR Vβ亚家族T细胞的分布及其克隆性.方法:利用RT-PCR分别扩增3例LL患者外周血单个核细胞的 TC R Vβ 24个亚家族基因的CDR3,以了解患者各Vβ亚家族的利用情况,然后将阳性的PCR产物进一步经荧光素标记和基因扫描分析产物的CDR3长度,了解T细胞克隆性.结果:患者中仅存在2～6个Vβ亚家族T细胞.基因扫描分析显示,2例患者外周血中的Vβ1 、Vβ 3或Vβ4亚家族出现克隆性增殖T细胞. 结论:LL患者外周血存在倾斜性分布和克隆性增殖的Vβ亚家族T细胞,这可能是机体T细胞受白血病细胞相关抗原的刺激作用而引起机体产生相应的特异性抗白血病的免疫反应.     

女性系统性红斑狼疮患者外周血Tim-1表达水平及其与疾病活动的相关性

目的:检测T细胞免疫球蛋白粘蛋白分子-1在女性系统性红斑狼疮(SLE)患者外周血的表达情况,并探讨与SLE疾病活动的相关性.方法:采用酶联免疫吸附测定(ELISA)方法检测SLE治疗前后和健康对照组外周血中T细胞免疫球蛋白粘蛋白分子-1的表达量,SLE活动性以系统性红斑狼疮病变活动指数(SLEDAI)表示,并进行相关性分析以明确T细胞免疫球蛋白粘蛋白分子-1与系统性红斑狼疮患者疾病活动性的关系.结果:治疗前SLE患者外周血中T细胞免疫球蛋白粘蛋白分子-1的表达水平明显高于对照组和治疗后SLE患者外周血中的表达水平,有统计学差异(P0.05),且其表达水平与SLEDAI评分呈正相关(r=0.639,P0.01).结论:T细胞免疫球蛋白粘蛋白分子-1的表达水平与SLE的发病有关,且可反映SLE患者的疾病活动状态.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
