To quantitatively determine tylosin and tilmicosin in edible animal tissues,a time-resolved fluoroimmunoassay(TRFIA) has been developed and validated.For this purpose,desmycosin-O-carboxymethoxylamine-BSA was fixed onto microtiter plates,standards and samples were loaded and,finally,diluted europium-labeled anti-tylosin antibodies were added.Results show that the limit of detection for tylosin was 0.03 ng mL-1 and that for tilmicosin was 0.05 ng mL-1.The recoveries were 73.6% to 120.5%,with coefficients of variation below 15.6% in various biological matrices spiked with tylosin and tilmicosin at concentrations of 50-200 ngg-1.There was good correlation(R2>0.99) between the TRFIA,an enzyme-linked immunosorbent assay and high performance liquid chromatography data.In conclusion,the new TRFIA is applicable to the detection of tylosin and tilmicosin and is an effective and economical method that will enable high-throughput sample screening.The method is expected to be widely applicable.  相似文献

The cyanobacterial communities in the surface and bottom waters of Sanya Bay were investigated on April 24 and 25,2010.Flow cytometry showed that the total cyanobacterial abundance in the surface and bottom layers ranged from 0.7×10 4 to 2.38×10 4 cells mL-1 and from 1×10 4 to 1.8×10 4 cells mL-1,respectively.Cyanobacterial diversity was analyzed using a molecular fingerprinting technique called denaturing gradient gel electrophoresis(DGGE),followed by DNA sequencing.The results were then interpreted through multivariate statistical analysis.Differences in the compositions of cyanobacterial communities were observed in the surface and bottom waters at the same station,with some bands obtained from both the surface and bottom layers,whereas some bands were present only in one layer.The predominant cyanobacterial species of the excised DGGE bands were related to Synechococcus or Synechococcus-like species(56.2%).Other phylogenetic groups identified included Chroococcidiopsis(6.3%),Cyanobium(6.3%) and some unclassified cyanobacteria(31.2%).A redundancy analysis(RDA) was conducted to reveal the relationships between the cyanobacterial community composition and environmental factors.Analysis results showed that the spatial variations in the cyanobacterial community composition in surface waters was significantly related to chlorophyll a(Chla),the biochemical oxygen demand(BOD),nitrate and phosphate(P<0.05).Meanwhile,the spatial variations in the bottom waters was significantly affected by nitrate,nitrite,and phosphate(P<0.05).Environmental parameters could explain 99.3% and 58.3% of the variations in the surface and bottom layers,respectively.  相似文献

【目的】研究斜阳岛附近海域柳珊瑚Anthogorgia caerulea可培养共生放线菌多样性及其发酵液代谢产物的生物毒活性。【方法】采用纯培养法和基于16SrRNA基因序列的系统发育分析对从广西斜阳岛附近海域采集的柳珊瑚Anthogorgia caeru[ea可培养共生放线菌多样性进行研究,利用卤虫致死法测试其生物毒活性。【结果】从柳珊瑚Anthogorgia caeru[ea中分离获得相关可培养放线菌23株,采用16SrRNA基因序列的系统发育分析后发现,这些菌株属于2个亚纲9个科9个属23种。绝大部分菌株都具有一定的生物毒活性,其中10株菌株有较强生物毒活性,l株有显著毒活性。【结论】广西斜阳岛附近海域柳珊瑚Anthogorgia caeru[ea中存在较为丰富的放线菌多样性,部分菌株具有较强的生物毒活性。  相似文献

A cost-effective electrocatalyst with high activity and stability was developed. The Fe-Nx and pyridinic-N active sites were embedded in nitrogen-doped mesoporous carbon nanomaterial by carbonization at high temperature. The electrocatalyst exhibited excellent electrochemical performance for the oxygen reduction reaction, with high onset potential and half-wave potential values (Eonset = 1.10 ?V and E1/2 ?= ?0.944 ?V) than 20 ?wt % Pt/C catalyst (1.04 and 0.910 ?V). The mass activity of the Schiff base network (SNW) based SNW-Fe/N/C@800° electrocatalyst (0.64 ?mA ?mg?1 @ 1 ?V) reached about half of the commercial Pt/C electrocatalyst (1.35 ?mA ?mg?1 @ 1 ?V). The electrocatalyst followed the 4-electron transfer mechanism due to very low hydrogen peroxide yield (H2O2 ?< ?1.5%) was obtained. In addition, this electrocatalyst with dual active sites showed high stability during cyclic voltammetry and chronoamperometry measurements. More importantly, the electrocatalyst also demonstrated the power density of 266 ?mW ?cm?2 in the alkaline anions exchange membrane fuel cell (AEMFC) test, indicating its prospective application for fuel cells.  相似文献

In this study, the biological and chemical diversity of 8 symbiotic marine fungal strains, with strong cytotoxicity against brine shrimp larvae, were investigated by nucleotide sequencing, morphology and cluster analysis of HPTLC fingerprint. These strains were identified by ITS rDNA sequencing, phylogenic analysis, and morphology to be Hypocrea lixii, Chaetomium globosum, Aspergillus fumigatus, Asp. clavatus and Alternaria sp. Their differences in secondary metabolites were shown by cluster analysis of digitalized colors of HPTLC spots, a newly developed method, which produced a similar dendrogram with that of ITS cluster analysis. Furthermore, this method can fully display intraspecific differences and even the remarkable difference in Aspergillus strains which goes beyond the boundary between genera. Their biological-chemical diversity may be the basis of their potent cytotoxicity and implies their potential in producing diversified antitumor or pesticidal constituents.  相似文献

In this paper,we report that the thermoelectric performance of n-type Pb Se could be improved through synergistically optimizing electrical and thermal transport properties via Sb doping and Mg alloying.The carrier concentration was firstly optimized through Sb doping,resulting in a maximum power factor of~15.4μW cm~(-1)K~(-2)and maximum ZT of~0.9 at 873 K in Pb_(0.99)Sb_(0.01)Se.Then,Mg was selected for alloying in Pb sites to produce point defects,which can largely intensify the phonon scattering and lower thermal conductivity.After Mg alloying,the thermal conductivity at 300 K(873 K)was significantly suppressed from~4.6 Wm~(-1)K~(-1)(1.5 Wm~(-1)K~(-1))for Pb_(0.99)Sb_(0.01)Se to~2.9 Wm~(-1)K~(-1)(1.1 Wm~(-1)K~(-1))for Pb_(0.99)Sb_(0.01)Se-6%Mg Se.Through combining Sb doping and Mg alloying,a maximum ZT of~1.1 was achieved at 873 K for Pb_(0.99)Sb_(0.01)Se-6%Mg Se,and the average ZT(ZT_(ave))was increased by 28.6%from~0.42 for Pb_(0.99)Sb_(0.01)Se to~0.54 for Pb_(0.99)Sb_(0.01)Se-6%Mg Se.The results indicate that Pb Se is a robust candidate for medium-temperature thermoelectric applications.  相似文献

Objective: To investigate the mechanism of carbapenem resistance and the occurrence of plasmid-mediated quinolone resistance determinants qnr and aac(6)-Ib-cr in a clinical isolate of Enterobacter cloacae. Methods: An ertapenem-resistant E. cloacae ZY106, which was isolated from liquor puris of a female gastric cancer patient in a Chinese hospital, was investigated. Antibiotic susceptibilities were determined by agar dilution method. Conjugation experiments, isoelectric focusing, polymerase chain reaction (PCR), and DNA sequence analyses of plasmid-mediated carbapenemases and quinolone resistance determinants were preformed to confirm the genotype. Outer membrane proteins (OMPs) were examined by urea-sodium dodecyl sulfatepolyacrylamide gel electrophoresis (Urea-SDS-PAGE). Results: Minimum inhibitory concentrations (MICs) of imipenem, meropenem, and ertapenem for ZY106 were 2, 4, and 16 μg/ml, respectively. Conjugation studies with Escherichia coli resulted in the transfer of significantly reduced carbapenem susceptibility. ZY106 produced IMP-1 metallo-β-lactamase and CTX-M-3 extended-spectrum β-lactamase, and E. coli transconjugant produced IMP-1. Plasmid-mediated quinolone resistance determinant qnrS1 was detected in ZY106. Transfer of the qnrS1-encoding-plasmid into E. coli by conjugation resulted in intermediate resistance to ciprofloxacin in E. coli transconjugant. Urea-SDS-PAGE analysis of OMPs showed that ZY106 lacked an OMP of approximately 38 kDa. Conclusion: It is the first IMP-1-producing Enterobacteriaceae in China and the first report of a clinical isolate that harbors both blaIMP and qnrS genes as well. The blaIMP-1, blaCTX-M-3, and qnrS1 are encoded at three different plasmids. IMP-1 combined with the loss of an OMP possibly resulted in ertapenem resistance and reduced imipenem and meropenem susceptibility in E. cloacae.  相似文献

Naringin is a predominant flavanone in grapefruit and shows a variety of biological effects such as antioxidative, anticancer, anti-inflammatory and antibacterial activity. However, its application in pharmaceutical field is limited by low water solubility, poor bioavailability and instability. To overcome the problem, naringin has been encapsulated in poly(lactic-co-glycolic acid) (PLGA) polymer by emulsion-diffusion-evaporation method in this work. Moreover, naringin loaded PLGA nanospheres were characterized by scanning electron microscope (SEM), dynamic light scatter method (DLS), fourier transform infrared (FTIR) spectra, UV–vis spectra and fluorescence spectra of DNA-EB competition displacement. The mean diameter of PLGA nanospheres and NRG/PLGA nanospheres was 123 ?± ?25 ?nm and 137 ?± ?30 ?nm, respectively. The drug encapsulation efficiency was 86.4% while the drug loading rate was 22.3%. The fluorescence spectra of the competitive DNA-binding experiments revealed that the functional activity of naringin was retained after loaded in PLGA. It is revealed that the initial burst effect happened in the initial 24 ?h and followed by sustained release lasting for 10 days. Moreover, the nanospheres exhibited strong antibacterial activity, and 99.9% of E. coli and S. aureus were killed when treated with naringin loaded PLGA nanospheres at the concentration of 0.2 ?mg ?mL?1 within 24 ?h. Furthermore, the viable cells remained only 48% when the concentration of NRG/PLGA nanospheres was 32 ?μg ?mL?1 and NRG/PLGA nanospheres was important for inhibition of cancer cells. It is concluded that the stable naringin loaded PLGA nanospheres could have potential application in food industry and nanomedicine field.  相似文献

Serving as the only defensive line between pathogens and human body, personal protective equipment (PPE) is increasingly attractive among researchers because of their strong antibacterial and temperature control abilities. However, efficient antibacterial properties and regenerative thermal control simultaneously remain unexplored in PPE. Here, one-dimensional halloysite nanotubes (HNTs) modified by acid etch method, are used to synthesis a multifunctional material of PEG/Cu2O@A-HNTs via in-situ reduction and physical adsorption which serves the above two purposes. PEG/Cu2O@A-HNTs showed rapid bacterial inactivation and achieved 96.3% bacteriostatic rate against E. coli in only 20 ?min. Meanwhile a broad and complete inactivation spectrum included both E. coli and S. aureus following a series of antibacterial mechanisms. Moreover, adsorption of 70 ?wt% PEG by acid etch HNTs is attributed to the nearly 3 times increased specific surface area compared with native HNTs. This enabled PEG/Cu2O@A-HNTs to attain a phase change enthalpy of 108.4 ?J/g. In addition, using PEG/Cu2O@A-HNTs as additives, antibacterial and phase change fiber (APCF) were melt-spun. Their efficiency factor against E. coli and S. aureus was above 99.99%, and retained a temperature control ability for 180s and 272s compared with PA6 fiber in hot and frigid environments respectively.  相似文献

A rapidly growing number of successful genome sequencing projects in plant pathogenic fungi greatly increase the demands for tools and methodologies to study fungal pathogenicity at genomic scale. Magnaporthe oryzae is an economically important plant pathogenic fungus whose genome is fully sequenced. Recently we have reported the development and application of functional genomics platform technologies in M. oryzae. This model approach would have many practical ramifications in design and implementation of upcoming functional genomics studies of filamentous fungi aimed at understanding fungal pathogenicity.  相似文献

Magnaporthe oryzae has been used as a primary model organism for investigating fungus-plant interaction. Many researches focused on molecular mechanisms of appressorium formation to restrain this fungal pathogen. Autophagy is a very high conserved process in eukaryotic cells. Recently, autophagy has been considered as a key process in development and differentiation in M. oryzae. The distribution of a CP-asymmetric quantityA in the decay channelJ/ψπ + π ? π 0 is investigated. This dimensionless quantityA is constructed from the momenta of the π-meson final states, namely A=P x π + P y π ? -P x π ? P y π ?, whereP h,P x h andP y h are the module of the momentum of a hadron h, the x- and y-components of that momentum respectively, theJ/ψ particle is produced by e+ e? collision, and the direction of the momentum of e+ is taken to be the positive direction ofz axis. There would exists the violation under the combined transformation of charge conjugation and space reflection (CP violation) when the average ofA among a lot of events, 〈A〉, be examined to be nonzero clearly from data. In this way, 748 events are selected from the BES experimental data, and analyzed. The corresponding averageA-value is measured to be 〈A〉 = 0.010 39 ± 0.014 61 ± 0.015 2. Fasciclin family proteins have been identified as cell adhesion molecules in various organisms. In this study, a novel Magnaporthe oryzae fasciclin-like protein encoding gene, named MoFLP1, was isolated from a subtractive suppressive cDNA library and functionally analyzed. Sequence analysis showed that the MoFLP1 gene contains an open reading frame (ORF) of 1050 nucleotides encoding 349 amino acids with a calculated molecular weight of 35.85 kDa and a pI of 7.76. The deduced MoFLP1 protein contains a 17-amino acid secretion signal sequence and an 18-amino acid sequence with the characteristics of a glycosylphosphotidylinositol (GPI) anchor additional signal at its N- and C-terminuses, respectively. Potential N-glycosylation sites and domains involving cell adhesion were also identified in MoFLP1. Sequence analysis and subcellular localization by the expression of MoFLP1-GFP fusion construct in M. oryzae indicated that the MoFLP1 protein is probably localized on the vacuole membrane. Two MoFLP1 null mutants generated by targeted gene disruption exhibited marked reduction of conidiation, conidial adhesion, appressorium turgor, and pathogenicity. Our results indicate that fasciclin proteins play important roles in fungal development and pathogenicity in M. oryzae.  相似文献

An interspecies conservedPlasmodium asparagine rich antigen, designated as ARK26, was isolated by immunoscreeningP. falciparum genomic DNA expression library with mouse convalescent anti-P. yeolii serum. Partial DNA sequence analysis reveals that ARK26 contains clusters of asparagines and no randomly repeated amino acid sequence motifs are observed. A 65×103 GST fusion protein is expressed by recombinant plasmid PGEX-5X-1 (ARK26) inE. coli C strain ABLE-K. Computer programs predict that two asparagine rich regions are among the possible antigenic epitopes of p37 encoded by ARK26. Interestingly, the sequence of ARK26 displays significant similarity to yeast and several other species’ mitochondrial genes, and its possible function is discussed. 为了探讨在土池中高密度养殖日本对虾(Penaeus japonicus Bate)的可能性,于1997年8月28日至1998年1月4日在广西沿海土池中进行秋冬日本对虾养殖试验,试验共设5口虾塘,每口池塘面积0.667hm2 ,底质酸性砂质,池水深可达1.8米,养殖水靠水泵抽取,沿进水渠道引入虾塘,1号-5号虾塘的养殖密度分别为62.4万尾/公顷,96.0万尾/公顷,96.0万尾/公顷,88.8万尾/公顷,96.8万尾/公顷,结果表明,在高密度的养殖条件下,日本对虾的生长速度随温度的下降和对虾个体的增大而减慢,与自然条件下生长的日本对虾相比较,高密度养殖试验中的日本对虾的生长缓慢得多,说明在类似的条件下进行的高密度日本对虾养殖,很难使日本对虾在常规养殖时间内达到较好的商品规格。  相似文献

Web-like ZnO nanostructures have been successfully synthesized using the potassium nitrate route at various temperatures to simplify conventional preparation methods. The structures and morphologies of the as-prepared products were characterized by X-ray powder diffraction (XRD) and field-emission scanning electron microscopy (FE-SEM). The results showed that the reaction temperature was an important parameter, and that there was a feedback effect between nano-structure and growth parameters, combined with in situ micro-calorimetry, the reaction rate constants of the three systems were found to have been: 2.43×10-6, 2.70×10-8 and 3.12×10-7s-1 respectively. Furthermore, based on the relationship governing the potential differences between nanoand bulk ZnO, thermodynamic functions of nano-ZnO such as standard molar entropy (Sm,ZnO(nano)), standard molar Gibbs free energy of formation (△rGm,ZnO(nano)), and standard molar enthalpy of formation (△rHm,ZnO(nano)) have been calculated by the electrochemical method.  相似文献
