Plurihormonality in the secretory granules of the normal human pituitary. An immunoelectron microscopic study

Normal human autopsy anterior pituitary tissue from 5 cases was embedded in LR White resin and immunolabelled using silver-enhanced 5-nm protein A gold probes. Follicle stimulating hormone (FSH), thyroid stimulating hormone (TSH), luteinizing hormone (LH), adrenocorticotrophic hormone (ACTH), growth hormone (GH) and prolactin (PRL) were immunolocalised to the level of secretory granule. A two-sided double-labelling method was used to cross-react two hormones at a time with respect to their corresponding antibodies. All possible combinations of the six pituitary hormones were tested. Plurihormonal granules were found that contained LH + FSH, LH + TSH, and FSH + TSH. Each hormone was also found in monohormonal granules. Granule diameter was significantly larger in the pluri as opposed to monohormonal granules.

Exocytosis of secretory granules — a probable mechanism for the release of neuromodulators in invertebrate neuropiles

Summary Presynaptic terminals typically contain secretory granules, usually 100–200 nm in diameter, in addition to the smaller synaptic vesicles. Evidence is presented that granule exocytosis is a widespread phenomenon in invertebrate neuropiles. Such secretory release is apparently associated with morphologically unspecialized regions of the plasmalemma, rather than synaptic thickenings.Acknowledgments. An electron microscopic study on the autonomic innervation of the rabbit parotid gland

Summary As visualized in the electron microscope, the parotid gland of the rabbit has a dual innervation. Both adrenergic and cholinergic nerves are equally distributed in the parenchyma and often run together within the same nerve bundle. Nerve terminals are observed not only subjacent to the basement membrane but interposed between the latter and the acinar cells, where they establish a close membrane to membrane contact with the latter.This investigation was supported by grants from Svenska Medicinska Sällskapet and the University of Umeå.     

The localization of3H-corticosterone in mast cell granules by electron microscopic autoradiography

Zusammenfassung Der Einbau von 1,2-³H-Corticosteron in die Mastzellen der lymphatischen Organe wurde bei Mäusen untersucht. Bei gleichzeitiger Anwendung von Autoradiographie und Elektronenmikroskopie konnte das Corticosteron in den Mastzellen nachgewiesen werden.

Riassunto Nel testicolo umano normale la fosforilasi (a) (attiva) é predominante nei confronti della forma (b) (inattiva), contrariamente a tutti gli altri tessuti. Tale comportamento é legato probabilmente al fatto che la fosforilasi (a), enzima del metabolismo del glicogeno, con la scissione di quest'ultimo fornisce l'energia necessaria alla sintesi degli acidi nucleici.     

Morphological characteristics of the secretory granules in pancreaticβ cells from species with identical primary structures of insulin

Zusammenfassung Die Primärstruktur des Insulins ist bei Hund und Schweinidentisch. Elektronenmikroskopisch wurde festgestellt, dass die sekretorischen Granulae der pankreatischen-Zellen bei beiden Arten eine unregelmässige Form haben. Eine quantitative Analyse ergab jedoch, dass die-Granulae des Hundes länger sind als die des Schweines.

Immunocytochemical anatomy of alpha granules in the human endocrine pancreas]

Using the sensitive protein A-gold technique for the demonstration of antigenic sites in thin section for electron microscopy it was found that in the alpha granules of human pancreas, glucagon immunoreactivity (specific, C-terminal) is restricted to the dense granule core while glicentin immunoreactivity predominates on the peripheral halo surrounding the dense core.     

Immunofluorescent localization of beta-lipotropin (beta-LPH) and beta-endorphins in normal and anencephalic human fetal pituitary gland]

The use of antibodies to alpha-(17-39) ACTH, beta-LPH, alpha- and beta-endorphins has enabled the detection of immunoreactive cells in human fetal pituitary glands as early as the eight week of gestation. The same cells reacted with the different antibodies. Immunoreactive cells were also observed in the pituitary of 6 human anencephalic fetuses.     

Immunohistochemical study of the pars intermediate of the mouse pituitary in different experimental conditions.

Alpha-MSH, beta-MSH and ACTH have been localized in the cells of hypophyseal intermediate lobe by fluorescence histoimmunological technics. Elaboration and excretion of these polypeptides are enhanced after dehydration or adrenalectomy. The most evident variations are seen with alpha-MSH and ACTH after dehydration, with beta-MSH after adrenalectomy.     

An improved method for the isolation of eosinophilic leukocytes from normal human blood

Summary A relatively short and simple method for the isolation of the eosinophils from normal human blood is reported. With a recovery of about 45–55%, cell preparations, showing a degree of purity of 90–98%, are obtained. The isolated cells are morphologically intact and viable, as assessed by the trypan blue exclusion test and by active phagocytosis.The authors are indebted to the Schweizerischer National-fonds zur Förderung der wissenschaftlichen Forschung for financial support.     

Lysosomal enzymes in the juxtaglomerular cell granules.

In addition to the already known acid phosphatase and beta-glucuronidase, 2 other lysosomal enzymes: aryl sulphatase and N-acetyl-beta-glucosaminidase were localized by histochemical methods in the renin-containing granules of the mouse juxtaglomerular cells.     

Sequence comparison of human pituitary growth hormone,human chorionic somatomammotropin and ovine pituitary lactogenic hormone

Zusammenfassung Ein Vergleich der Aminosäuresequenzen von menschlichen Wachstumshormonen und menschlichem Chorion-Somatomammotropin mit denjenigen von Prolactin zeigt, dass 26% des Rückstandes in allen drei Molekülen die gleiche Position hatten mit zusätzlichen 34% als möglichen Ersatz. Alle drei Hormone scheinen einen gleichen Ursprung zu haben.     

Myosin-like protein in the human parathyroid glands: An immunohistochemical study

Summary The immunofluorescent staining of the human parathyroid glands shows that the principal cells bind antimyosin-like antibodies. The presence of a myosin-like protein in the principal cells of the human parathyroid glands is presumably in rapport with parathormone secretion.     

Pigment granules in choroidal melanophores of the albino goldfish.

Pigment granules in choroidal melanophores of the albino goldfish contained fine particulate materials which were in various degrees aggregated in clumps. Tyrosinase was considered to be present in an inhibited state in these pigment granules.
