Normal human autopsy anterior pituitary tissue from 5 cases was embedded in LR White resin and immunolabelled using silver-enhanced 5-nm protein A gold probes. Follicle stimulating hormone (FSH), thyroid stimulating hormone (TSH), luteinizing hormone (LH), adrenocorticotrophic hormone (ACTH), growth hormone (GH) and prolactin (PRL) were immunolocalised to the level of secretory granule. A two-sided double-labelling method was used to cross-react two hormones at a time with respect to their corresponding antibodies. All possible combinations of the six pituitary hormones were tested. Plurihormonal granules were found that contained LH + FSH, LH + TSH, and FSH + TSH. Each hormone was also found in monohormonal granules. Granule diameter was significantly larger in the pluri as opposed to monohormonal granules.  相似文献

Summary Presynaptic terminals typically contain secretory granules, usually 100–200 nm in diameter, in addition to the smaller synaptic vesicles. Evidence is presented that granule exocytosis is a widespread phenomenon in invertebrate neuropiles. Such secretory release is apparently associated with morphologically unspecialized regions of the plasmalemma, rather than synaptic thickenings.Acknowledgments. Summary As visualized in the electron microscope, the parotid gland of the rabbit has a dual innervation. Both adrenergic and cholinergic nerves are equally distributed in the parenchyma and often run together within the same nerve bundle. Nerve terminals are observed not only subjacent to the basement membrane but interposed between the latter and the acinar cells, where they establish a close membrane to membrane contact with the latter.This investigation was supported by grants from Svenska Medicinska Sällskapet and the University of Umeå.  相似文献

Zusammenfassung Die Primärstruktur des Insulins ist bei Hund und Schweinidentisch. Elektronenmikroskopisch wurde festgestellt, dass die sekretorischen Granulae der pankreatischen-Zellen bei beiden Arten eine unregelmässige Form haben. Eine quantitative Analyse ergab jedoch, dass die-Granulae des Hundes länger sind als die des Schweines.

Using the sensitive protein A-gold technique for the demonstration of antigenic sites in thin section for electron microscopy it was found that in the alpha granules of human pancreas, glucagon immunoreactivity (specific, C-terminal) is restricted to the dense granule core while glicentin immunoreactivity predominates on the peripheral halo surrounding the dense core.  相似文献

The use of antibodies to alpha-(17-39) ACTH, beta-LPH, alpha- and beta-endorphins has enabled the detection of immunoreactive cells in human fetal pituitary glands as early as the eight week of gestation. The same cells reacted with the different antibodies. Immunoreactive cells were also observed in the pituitary of 6 human anencephalic fetuses.  相似文献

Alpha-MSH, beta-MSH and ACTH have been localized in the cells of hypophyseal intermediate lobe by fluorescence histoimmunological technics. Elaboration and excretion of these polypeptides are enhanced after dehydration or adrenalectomy. The most evident variations are seen with alpha-MSH and ACTH after dehydration, with beta-MSH after adrenalectomy.  相似文献

Summary A relatively short and simple method for the isolation of the eosinophils from normal human blood is reported. With a recovery of about 45–55%, cell preparations, showing a degree of purity of 90–98%, are obtained. Zusammenfassung Ein Vergleich der Aminosäuresequenzen von menschlichen Wachstumshormonen und menschlichem Chorion-Somatomammotropin mit denjenigen von Prolactin zeigt, dass 26% des Rückstandes in allen drei Molekülen die gleiche Position hatten mit zusätzlichen 34% als möglichen Ersatz.
