抗菌肽AD基因的改造及在毕赤酵母中的表达

采用PCR技术改造抗菌肽AD基因，将其C末端改造为Asn编码，改造后的抗菌肽Cecropin AD基因克隆到pPICZα－A载体上，构建成酵母重组分泌型表达载体，转化毕赤酵母（Pichia pastoris)受体菌GS115中，采用zerocin抗性标记筛选重组转化子，经摇瓶发酵，浓缩发酵液进行酸性聚丙烯酰胺凝胶电泳分析和抑菌活性检测。结果表明，改造后的Cecropin AD基因在毕赤酵母中获得了表达，表达产物经α－Factor信号肽引导分泌到胞外，具有较强的杀菌活性。     

重组人角质细胞生长因子(KGF-2)在毕赤酵母中的克隆与表达

利用RT－PCR技术，从体外培养的人胚胎成纤维细胞中扩增出人角质细胞生长因子（KGF－2）的cDNA及基因序列，并将其基因序列克隆入质粒pGEM-T Easy中，经测序与文献报道序列一致，将KGF－2基因切出后定向插入酵母分泌性表达质粒pPICZαA中，转化毕赤酵母菌株GS115，获得的重组体经甲醇诱导可表达出人角质细胞生长因子KGF－2，表达的KGF－2经Ni^2 柱亲和层析纯化后，有很好的生物学活性。     

纳豆激酶基因在巴斯德毕赤酵母中的表达

构建pPICZαA-NK重组质粒，并转化入E.coli TOP-10中，得到转纳豆激酶基因工程菌，提取重组质粒经单酶切，双酶切，PCR分析及序列测定，证明克隆到载体pPICZαA上的外源基因即为纳豆激酶基因，将重组质粒pPICZαA-NK线性化后，分别转化毕赤酵母GS115与KM71，在含Zeocin^TM的选择性YEPD平板筛选到重组酵母，经检测重组酵母发酵上清液中具纳豆激酶溶纤活性，SDS-PAGE电泳结果表明外源蛋白的表达量占菌体蛋白的10%左右。     

乙肝表面抗原在分泌型毕赤酵母中的表达及纯化

目的对分泌型毕赤酵母中表达乙肝表面抗原并进行亲和层析纯化。方法从已确诊的乙肝患者阳性血清中提取乙肝病毒DNA,PCR扩增乙肝病毒表面抗原编码基因S,将其插入含有仅分泌信号肽序列和6个组氨酸纯化标签的毕赤酵母表达载体pPICZα,成功构建了重组表达载体pPICZα—HBVs。电转化酵母菌株GS115,得到重组乙肝表面抗原S的酵母表达菌株。结果诱导培养基BMMY中甲醇终浓度为1％,诱导表达48h重组蛋白表达量及抗原性达到最高。非变性条件下亲和层析纯化后,薄层扫描分析得到纯度超过95％,表达量约为2mg／L的重组蛋白。结论Western-blot和ELISA分析表明,所得产物为乙肝表面抗原S蛋白,其纯度和产量足以免疫小鼠制备单克隆抗体。     

重组毕赤酵母体系生成PIUGT1蛋白适宜条件研究

通过单因素试验研究pH和温度对重组毕赤酵母GS115/pPIZαA - PlUGT1生长以及甲醇体积分数、pH和温度对诱导表达PlUGT1蛋白的影响.结果表明,pH6.5和30℃是培养重组毕赤酵母GS115/pPIZαA - PlUGT1最佳的生长条件,重组毕赤酵母GS115/pPlZαA - PlUGT1诱导表达蛋白...     

家蝇防御素defensin cDNA的克隆及在毕赤酵母中的表达

提取家蝇总RNA,RT-PCR扩增编码家蝇防御素defensin的cDNA,克隆并测定了其序列,将该cDNA序列与酵母表达载体pPICZαA重组,构建酵母分泌型表达载体pPICZα-de,电激法转化毕赤酵母(Pichia pasto-ris)受体菌GS115。经表型筛选和PCR鉴定证明目的片段已稳定整合到酵母染色体基因组中。在含不同浓度的Zeoc in平板上筛选到高拷贝整合的转化子,阳性克隆经甲醇诱导表达,用Tric ine-SDS-PAGE确定了表达产物的正确性,琼脂孔穴平板扩散法、比浊法测定了表达产物的活性,对金黄色葡萄球菌Staphylococcus aureus具有一定的杀菌活性。     

结合利用pGAPZαA和pPICZαA构建目的蛋白的多拷贝载体并在毕赤酵母中表达

结合利用载体pGAPZαA和pPICZαA构建目的蛋白的可线性化多拷贝表达载体,并转化毕赤酵母表达目的蛋白.首先将目的蛋白基因构建到组成型表达载体pGAPZαA中,然后依据载体自身特性,用同尾酶BglⅡ和Bam HⅠ切取其表达盒并连接到甲醇诱导型载体pPICZαA中的Bam HⅠ位点处,经多次重复后获得目的蛋白多拷贝表达载体,线性化后电转化毕赤酵母表达目的蛋白.灵芝免疫调节蛋白LZ8是一个已成功在毕赤酵母中表达的蛋白,作为检验该方法的样本其基因被首先构建到载体pGAPZαA中得到质粒pGAPZαA-LZ8,切取其表达盒构建到载体pPICZαA中,经过4次重复得到了多拷贝表达载体pPICZαA-[GAPZαA-LZ8]4,然后利用pPICZαA特有的限制性内切酶位点SacⅠ进行线性化,最终电转化到毕赤酵母GS115中进行组成型表达蛋白LZ8,其表达量达到pGAPZαA-LZ8转化菌株表达量的2~3倍.经检验,结合利用载体pGAPZαA和pPICZαA可以构建目的蛋白的多拷贝表达载体,并且可以对载体进行线性化而不影响转化酵母时的转化效率.     

HIV-1外膜蛋白gp120在酵母Pichia pastoris中的表达

目的 用巴斯德毕赤酵母系统表达HIV外膜糖蛋白gp120.方法 将从HIV-1型国际标准株pHXB2-gp160的基因序列中扩增出的gp 120分子的长片段基因(1 419 bp)和短片段基因(417bp)分别克隆入真核表达载体pPICZαA与pPICZB中,以电穿孔法转化酵母GS 115.用YPDS-zeo平板筛选重组子、PCR方法检测整合到酵母菌GS 115基因组中的gp 120片段基因,经甲醇诱导表达后,SDS-PAGE和免疫印迹分析表达产物.结果 诱导后gp 120短片段多肽在GS 115中少量表达,在诱导表达24 h重组蛋白表达量及抗原性达到最高,表达产物被降解,具有良好的抗原特异性.诱导后gp 120长片段多肽未被GS 115所表达.结论 在巴斯德毕赤酵母中成功表达gp 120分子长片段需要优化gp 120基因,为制备HIV-1的诊断抗原和基因工程重组疫苗打下基础.     

汉坦病毒囊膜糖蛋白G2在毕赤酵母GS115中的表达及鉴定

构建了编码汉坦病毒囊膜糖蛋白G2基因的重组质粒,在毕赤酵母中表达,为汉坦病毒基因工程疫苗的研究提供实验基础。利用PCR法从含汉滩病毒76-118株M基因的M56质粒中扩增编码糖蛋白G2的基因片段,克隆入酵母分泌表达载体pPICZαA,构建重组质粒pPICZαA-G2。酶切鉴定挑取的阳性克隆,转化入GS115工程菌,在含100μg/mLZeocin的YPD培养基上筛选。挑选单菌落,PCR鉴定阳性克隆,用0.5%甲醇诱导表达,并利用SDS-PAGE及Western-Blot鉴定表达产物。序列分析表明所获得的基因片段与编码汉滩病毒76-118株囊膜糖蛋白G2的基因一致;100μg/mLZeocinYPD培养基上筛选出含pPICZαA-G2转化子,PCR鉴定为阳性克隆;SDS-PAGE可见约70kDa处有目的蛋白表达条带,经Western-Blot证实该条带为汉滩病毒囊膜糖蛋白G2。成功地构建了重组酵母表达载体pPICZαA-G2,并在毕赤酵母中初步表达成功,为今后汉坦病毒囊膜糖蛋白G2表达纯化以及基因工程疫苗的制备奠定了一定基础。     

兔IL-15蛋白在毕赤酵母中的表达及其免疫学活性

用特异性引物扩增出兔IL-15基因片段,用KpnI和NotI双酶切后将其定向克隆到pPICZαA中,构建出重组质粒pPICZαAIL-15。将pPICZαAIL-15用SacI内切酶线性化后,电转化感受态GS115酵母细胞,用PCR法筛选阳性重组子。用1.5%的甲醇诱导重组酵母菌,取培养物上清进行重组蛋白的检测。结果SDS-PAGE电泳可在重组酵母菌培养物上清中检测到分子量为18.6kDa大小的重组蛋白。凝胶薄层扫描分析表明,3株重组酵母菌在1.5%甲醇诱导144h后,重组IL-15表达量约占培养物上清总蛋白量的6.1%~8.6%。将重组IL-15用Ni离子亲和层析纯化后,在MTT试验中表现出明显的促进淋巴细胞的增殖反应,证实表达的重组IL-15具有天然蛋白的免疫学活性。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
