Amnesic effects of intravenous diazepam and lorazepam.

The amnesic effects of 2 benzodiazepine drugs, diazepam (Valium) and lorazepam (Ativan), have been investigated. Some of the effects were similar to those of certain clinical amnesic syndromes. The effects were more extensive than previous work has indicated.

Summary Muscimol induced in rats a recall deficit which possibly results from a dissociation of learning. In one out of the 2 experimental conditions studied, a cross dissociation of learning was found between diazepam and muscimol.This work was supported by a grant of INSERM (ATP 39-76-71).

Zusammenfassung Die Glucuronide des Oxazepams und Lorazepams wurden aus dem Harn von Zwergschweinen, denen die entsprechenden Benzodiazepine verabreicht wurden, isoliert. Die Metaboliten wurden durch Analyse der Spaltprodukte gekennzeichnet. Massenspektrometrie der Trimethylsilylderavite war für die Struktur derO-Glucuronide beweisend.     

Circadian variation of diazepam acute toxicity in mice

Summary The LD₅₀ of i.p. injected diazepam was determined every 4 h over a 24-h period in albino mice adapted to a 12-h dark/12-h light programmed illumination cycle. Results show that diazepam is more toxic during the light phase of the cycle than during the dark phase and demonstrate circadian variation in the toxicity of the compound in mice.

Facilitation of synaptic transmission by diazepam in a perfused frog cerebellum

Summary The action of diazepam on the parallel fibres-Purkinje cell synapse was studied in a perfused frog cerebellum. Diazepam facilitates the excitatory input to Purkinje cells and hence increases the inhibition produced by Purkinje cells activity.Supported by a grant from the Israel Center of Psychobiology.This work is part of M. Inhibition of lymphocyte blastogenesis by diazepam, meprobamate, and other psychotropic drugs]

Imipramine, diazepam, chlordiazepoxide, and meprobamate inhibit incorporation of 3H-thymidine in human lymphocytes in culture. For the first three drugs, 50% inhibition occurs at a concentration of 5 X 10(-5) M, and for meprobamate at a concentration of 10(-3 M. Ethanol has no action up to 10(-1) M.     

Enhancement by pyridoxine of the action of diazepam on spinal presynaptic inhibition

Summary In spinal unanesthetized cats, pyridoxine pretreatment significantly enhanced the diazepam-induced increase in dorsal root potentials and dorsal root reflexes.Acknowledgments. Effect of diazepam (Valium®) on chronic stress-induced hypertension in the rat

Summary Chronic treatment with diazepam was effective in preventing chronic stress-induced hypertension in rats. It also prevented the stressful stimuli from maintaining a hypertensive level in animals previously made hypertensive by chronic stress.     

Effect of vagal denervation on insulin release after oral and intravenous glucose

Zusammenfassung Perorale Glykosebelastung ergab bei vagotomierten Ratten Erhöhung des Blutglykosespiegels. Nach intravenöser Glykosezufuhr wurden niedrigere Werte von immunoreaktivem Insulin im Serum gefunden. Ebenso war der insulinogene Index bedeutend niedriger sowohl nach peroraler als auch nach intravenöser Glykosezufuhr. Vagotomie dürfte somit die glykosebedingte Insulinfreisetzung reduzieren.
