缺铁玉米根cDNA文库的构建及蛋白和cDNA差异比较

采用异硫氰酸胍酚氯仿法分离铁胁迫玉米根总ＲＮＡ,然后采用寡聚ｄＴ 纤维素层析柱法纯化Ｐｏｌｙ（Ａ） ＋ ＲＮＡ．用含有ＸｈｏＩ位点的ｌｉｎｋｅｒｐｒｉｍｅｒ 锚定合成ｃＤＮＡ 第一条链后,采用替代法合成第二条链,接着连接上ＥｃｏＲＩＡｄａｐｔｅｒ 以便定向重组到载体上．ｃＤＮＡ 经过ＸｈｏＩ消化和分级分离后,带有ＸｈｏＩ和ＥｃｏＲＩ粘性末端的ｃＤＮＡ 定向重组到λＺＡＰ表达载体的ＸｈｏＩ和ＥｃｏＲＩ位点上．经体外包装,重组的噬菌体转导宿主菌ＸＬ１Ｂｌｕｅ ＭＲＦ,最终获得滴度为４－５ ×１０５ ｐｆｕ／ｕｇ 的λＺＡＰ 表达ｃＤＮＡ 文库．通过差异筛选法和质膜双向电泳验证了缺铁和加铁条件下ｃＤＮＡ 和质膜蛋白的差异     

杂交捕获抑制聚腺苷二磷酸核糖聚合酶基因的体外翻译

体外转录载体ｐＳＰ７２—Ｃ１２和ｐＳＰ７２—Ｃ１．４分别经ＸｂａⅠ和ＸｈｏⅠ酶切线性化后，用ＳＰ６ＲＮＡ聚合酶与Ｔ７ＲＮＡ聚合酶进行体外转录得到编码人聚腺苷二磷酸核糖聚合酶基因的ｍＲＮＡ全序列（３．７ｋｂ）及与其５’端编码区的起始密码子ＡＵＧ上游区域互补的反义ＲＮＡ片段（１．４ｋｂ）。等摩尔数的正、反义ＲＮＡ通过酚相乳浊杂交技术进行分子杂交。在网织红细胞裂解物体外翻译系统中，正义ＲＮＡ能正常地翻译出蛋白质；而反义ＲＮＡ与正义ＲＮＡ杂交可阻断翻译的进行。

通过计算机分析转座子Ｔｎ９１７的全序列，详细阐述了其物理图谱、结构功能及其转录调节机制．Ｔｎ９１７的５个ＯＲＦｓ排列在同一条ＤＮＡ链上，且阅读方向都从左至右．ＯＲＦ１－３起始点的左侧翼排列有启动子序列和Ｓｈｉｎｅ－Ｄａｌｇａｒｎｏ序列．ＯＲＦ５（编码转座酶）和ＯＲＦ４（编码拆分酶）的转录方向是一致的，翻译也紧密偶联在一起．在ＯＲＦ３和ＯＲＦ４之间存在１个ｒｅｓ位点，与Ｔｎ３中的ｒｅｓ位点基本同源．翻译衰减的功能与ｒＲＮＡ甲基化酶（由ＯＲＦ２编码的、ｅｒｍ基因的产物）诱导有关，在这个结构基因的左侧翼有２００ｂｐ的前导区域编码一个具调控功能的３６个氨基酸组成的多肽（由ＯＲＦ１编码）．

本文用ＬｉＣｌ沉淀法提取了猪垂体总ＲＮＡ，经。ｏｌｉｇｏ（ｄＴ）－ｃｅｌｌｕｌｏｓｅ柱亲和层析，分离纯化了猪垂体Ｐｏｌｙ（Ａ）＋ＲＮＡ．用自制的兔网织红细胞体外翻译体系鉴定，表明该Ｐｏｌｙ／（Ａ）＋ＲＮＡ具有较高的翻译活性．ＳＤＳ－ＰＡＯＥ，放射自显影分析翻译产物提示，在制各的猪垂体Ｐｏｌｙ（Ａ）＋ＲＮＡ中，编码猪生长激素前体的ｍＲＮＡ占有较高的比例．     

在大肠杆菌中利用反义RNA抑制乙肝病毒（HBV）X基因的表达

稻瘟菌MgCYP51B基因稀有密码子和mRNA二级结构分析与表达优化

在大肠杆菌中异源表达稻瘟菌CYP51(P450-14DM, MgCYP51B)基因,通过对MgCYP51B进行跨膜区预测和同源模建分析,并截短稻瘟菌CYP51B基因,构建了9种不同的重组表达质粒以实现蛋白表达.结果表明：只有pET28-MgB-83在3种宿主菌中实现了表达,且在BL21(DE3)pLysS中表达量最大.通过对稀有密码子和mRNA翻译起始区二级结构进行分析,发现稀有密码子和mRNA翻译起始区二级结构稳定性对MgCYP51B蛋白的表达都有影响,适用于稀有密码子表达的Rossetta(DE3)并不适用于MgCYP51B基因的最优表达.同时只有翻译起始区二级结构自由能最低的pET28-MgB-83才能实现表达.本研究实现了稻瘟菌MgCYP51B基因在大肠杆菌中的异源表达,证实密码子的偏爱性和翻译起始区二级结构稳定性影响MgCYP51B蛋白表达.

This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
