Summary The fungicide dithane Z-78 (zinc ethylene bisdithiocarbamate) has been found to be an inhibitor of growth and heterocyst differentiation inNostoc muscorum. Its inhibitory effect has been reversed by exogenous glucose. A spontaneous mutant resitant to a toxic concentration of the fungicide grows in the presence of dithane at a dose normally applied in fields and does not require an exogenous carbon source for its growth.Thanks are due to Council of Scientific and Industrial Research, Govt. of India, New Delhi-110001, for providing financial assistance to A.V. in the form of a Post-Doctoral Research Fellowship.  相似文献

Exogen cell fusion induced in vitro by a wild strain of Visna virus of Sheep is compared with two mutant strains isolated from the precedent. One of them produces large plaques in vitro (strain LPF), and the other produces small plaques (strain SPF). These strains behave in different ways according to infection multiplicity, type of cells, temperature and timing of fusing activity. Taking the wild strain K 796 as a base of reference, the strain SPF seems to have a higher rate fusing activity and the strain LPF lower rate of fusing activity.  相似文献

Résumé Par passage en série sur des cellules en culture et en présence de 5-iodo-2-desoxyuridine (IUdR) on a obtenu une souche de virus herpétique résistant à l'IUdR. Cette souche est très peu virulente chez la souris et le lapin, mais est capable de protéger ces animaux contre le virus pathogène d'origine.

Summary Several N-1-alkyl-, 3-, and 4-carbamidopyridinium halides were synthesized and determined to be inhibitors of mitochondrial oxidative phosphorylation. L-Glutamate respiration was most depressed by N-1-dodecylpyridinium bromide whereas succinate respiration was most depressed by N-1-dodecylisonicotinamide bromide. Combination of inhibitors with mitochondrial sites may involve lipophilic interactions as modified by steric restrictions.  相似文献

BHK21 cells transformed by wild type or Ts3 mutant polyoma virus contain an inhibitor of polyoma virus replication when grown at permissive (36 degrees C) as well as non-permissive temperature (39 +/- 0.5 degrees C). Cells transformed by the Tsa mutant contain the inhibitor at the permissive but not at the non-permissive temperature. The inhibitor reappears in the latter cells however, upon shift from the non-permissive to the permissive temperature. If a reversible protein inhibitor (methionyl-adenylate, reversible inhibitor of the aminoacyl-t-RNA synthetase) is applied during the temperature shift experiments, the inhibitor does not reappear indicating that new protein synthesis is required for the recovery of its activity.  相似文献

Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disease. Neuronal vacuolization and glial activation are pathologic hallmarks in the superoxide dismutase 1 (SOD1) mouse model of ALS. Previously, we found the neuropeptide calcitonin gene-related peptide (CGRP) associated with vacuolization and astrogliosis in the spinal cord of these mice. We now show that CGRP abundance positively correlated with the severity of astrogliosis, but not vacuolization, in several motor and non-motor areas throughout the brain. SOD1 mice harboring a genetic depletion of the βCGRP isoform showed reduced CGRP immunoreactivity associated with vacuolization, while motor functions, body weight, survival, and astrogliosis were not altered. When CGRP signaling was completely disrupted through genetic depletion of the CGRP receptor component, receptor activity-modifying protein 1 (RAMP1), hind limb muscle denervation, and loss of muscle performance were accelerated, while body weight and survival were not affected. Dampened neuroinflammation, i.e., reduced levels of astrogliosis in the brain stem already in the pre-symptomatic disease stage, and reduced microgliosis and lymphocyte infiltrations during the late disease phase were additional neuropathology features in these mice. On the molecular level, mRNA expression levels of brain-derived neurotrophic factor (BDNF) and those of the anti-inflammatory cytokine interleukin 6 (IL-6) were elevated, while those of several pro-inflammatory cytokines found reduced in the brain stem of RAMP1-deficient SOD1 mice at disease end stage. Our results thus identify an important, possibly dual role of CGRP in ALS pathogenesis.  相似文献

Résumé Dans un milieu minimal la glycine et la sérine, ajoutées en concentrations physiologiques, augmentent le taux de la sérine-hydroxyméthyltransférase durant la croissance logarithmique duNeurospora crassa. La glycine influence également la synthèse de cet enzyme chez le mutantSer-1 deNeurospora. Ainsi la glycine exogène exerce une régulation de la sérine-hydroxyméthyltransférase et cet enzyme joue un rôle possible dans la synthèse de la sérine dans cet organisme en phase logarithmique de croissance.

The antiviral activity of Shigyaku-to (TJS-109), a traditional Chinese herbal medicine, was investigated in mice infected with herpes simplex virus type 1 (HSV-1). TJS-109 is a combination of the medicinal plant extracts fromZingiberis siccatum rhizoma,Aconiti tuber andGlycyrrhizae radix in a specific proportion. Mice infected with a 10 LD50 dose of HSV-1 were treated with TJS-109 orally at doses of 1.25 to 20 mg/kg 2 days before, and 1 and 4 days after the infection. The treated groups had 80% (1.25 mg/kg), 40% (5 mg/kg) and 23% (20 mg/kg) mortality rates 25 days after the infection as compared with a 100% mortality rate in control mice treated with saline. When HSV-1 infected mice (recipients) received CD8+T cell fractions derived from spleens of mice treated with TJS-109 (donors), 70% of recipients survived, as compared with 0% survivors in the groups of mice treated with saline, B cell fractions, CD4+ T cell fractions or macrophage-enriched fractions prepared from the same donors. TJS-109 did not show any virucidal activities against HSV-1 or any virostatic activities on the growth of HSV-1 in Vero cells. These results suggest that TJS-109 protected mice exposed to lethal amounts of HSV-1 through the activation of CD8+ T cells.  相似文献

Summary After i.p. inoculation with the Guajira strain of Venezuelan equine encephalomyelitis virus a significant decrease in the density of (3H) spiroperidol binding sites in the striatum, midbrain and frontal cortex was observed. No changes in the affinity of the receptors could be demonstrated. This finding is compatible with neuronal degeneration caused by the viral infection.Acknowledgments. Summary The orally-effective antiallergic compound [N-(2-oxo-3,5,7-cycloheptatrien-1-yl)] aminooxoacetic acid ethyl ester (AY-25,674) exhibited a potency equivalent to or 3 times less than theophylline in inhibiting guinea-pig lung and beef heart PDE, respectively. AY-25,674 did not affect the basal activity of guinea-pig lung adenyl cyclase. Although part of the antiallergic activity of AY-25,674 may be due to the ability to elevate cyclic AMP levels by PDE inhibition, other modes of action appear to be of greater relevance.The authors acknowledge the excellent technical assistance of Ms M.T.
